    Description: Although it is generally accepted that cellular differentiation requires changes to transcriptional networks, dynamic regulation of promoters and enhancers at specific sets of genes has not been previously studied en masse. Exploiting the fact that active promoters and enhancers are transcribed, we simultaneously measured their activity in 19 human and 14 mouse time courses covering a wide range of cell types and biological stimuli. Enhancer RNAs, then messenger RNAs encoding transcription factors, dominated the earliest responses. Binding sites for key lineage transcription factors were simultaneously overrepresented in enhancers and promoters active in each cellular system.     Description: The results of a pilot study into the application of an unsupervised clustering approach to the analysis of catchment-based National Geochemical Survey of Australia (NGSA) geochemical data combined with geophysical and geological data across northern Australia are documented. NGSA Mobile Metal Ion® (MMI) element concentrations and first and second order statistical summaries across catchments of geophysical data and geological data are integrated and analysed using Self-Organizing Maps (SOM). Input features that contribute significantly to the separation of catchment clusters are objectively identified and assessed. A case study of the application of SOM for assessing the spatial relationships between Au mines and mineral occurrences in catchment clusters is presented. Catchments with high mean Au code-vector concentrations are found downstream of areas known to host Au mineralization. This knowledge is used to identify upstream catchments exhibiting geophysical and geological features that indicate likely Au mineralization. The approach documented here suggests that catchment-based geochemical data and summaries of geophysical and geological data can be combined to highlight areas that potentially host previously unrecognised Au mineralization.

    Description: Haematopoietic stem cells (HSCs), which sustain production of all blood cell lineages, rely on glycolysis for ATP production, yet little attention has been paid to the role of mitochondria. Here we show in mice that the short isoform of a critical regulator of HSCs, Prdm16 (refs 4, 5), induces mitofusin 2 (Mfn2), a protein involved in mitochondrial fusion and in tethering of mitochondria to the endoplasmic reticulum. Overexpression and deletion studies, including single-cell transplantation assays, revealed that Mfn2 is specifically required for the maintenance of HSCs with extensive lymphoid potential, but not, or less so, for the maintenance of myeloid-dominant HSCs. Mfn2 increased buffering of intracellular Ca(2+), an effect mediated through its endoplasmic reticulum-mitochondria tethering activity, thereby negatively regulating nuclear translocation and transcriptional activity of nuclear factor of activated T cells (Nfat). Nfat inhibition rescued the effects of Mfn2 deletion in HSCs, demonstrating that negative regulation of Nfat is the prime downstream mechanism of Mfn2 in the maintenance of HSCs with extensive lymphoid potential. Mitochondria therefore have an important role in HSCs.     Description: An understanding of collective effects is of fundamental importance for the design and optimisation of the performance of modern accelerators. In particular, the design of an accelerator with strict requirements on the beam quality, such as a free electron laser (FEL), is highly dependent on a correspondence between simulation, theory and experiments in order to correctly account for the effect of coherent synchrotron radiation (CSR), and other collective effects. A traditional approach in accelerator simulation codes is to utilise an analytic one-dimensional approximation to the CSR force. We present an extension of the 1D CSR theory in order to correctly account for the CSR force at the entrance and exit of a bending magnet. A limited range of applicability to this solution—in particular, in bunches with a large transverse spot size or offset from the nominal axis—is recognised.     Description: ResumoA exploração desordenada ao longo dos anos das argilas bentoníticas do município de Boa Vista, PB, resultou no esgotamento das variedades nobres. Assim, o que existe atualmente são bentonitas com elevado percentual de minerais acessórios, elementos contaminantes a exemplo do quartzo, os quais influem negativamente nas propriedades reológicas das dispersões. O presente trabalho propõe concentrar dispersões dessas argilas pouco nobres pela operação de hidrociclonagem e posterior secagem por spray dryer, visando à obtenção de amostras com valores de reologia de acordo com norma EP-1EP-00011-A da Petrobrás. Assim, preparou-se dispersões de três composições com as argilas Bofe, Chocolate, Verde-Lodo, Chocobofe e Sortida previamente selecionadas por planejamento experimental com concentrações de 4% em massa de argila para serem concentradas por hidrociclonagem. Os concentrados foram secos por spray dryer em duas condições de secagem. Os resultados evidenciaram a eficiência das operações de hidrociclonagem e secagem por spray dryer das misturas com notável melhoria nas propriedades reológicas.

    Description: As argilas estão entre as mais importantes matérias-primas devido as suas múltiplas funções industriais. No caso especial da indústria de cerâmica vermelha, as argilas têm ampla utilidade, sendo o principal produto para a fabricação de telhas e blocos cerâmicos, que podem ser incorporados com resíduos industriais. Portanto, o objetivo deste trabalho foi estudar a influencia dos diferentes tipos de argilas na qualidade dos blocos cerâmicos de vedação, incorporados com lodo têxtil, por meio do processo de solidificação/estabilização. Avaliou-se a incorporação do lodo têxtil em 5 tipos de argilas, coletadas na região norte do Paraná, e realizou-se uma análise qualitativa dos blocos cerâmicos, com composição de 15% de resíduo têxtil, em função de suas características, estabelecidas nas normas da Associação Brasileira de Normas Técnicas. O resíduo utilizado foi proveniente de 12 indústrias têxteis de Maringá, PR. Inicialmente foi feita uma caracterização do lodo e das argilas. Os blocos cerâmicos foram fabricados em escala reduzida e submetidos a testes de resistência a compressão, lixiviação, solubilização e absorção de água, de acordo com as respectivas normas. Verificou-se nas condições estudadas que os blocos cerâmicos não apresentaram lixiviação nem solubilização. Portanto, o processo de solidificação/estabilização foi capaz de imobilizar de forma eficaz, os metais presentes no resíduo têxtil, mostrando-se como um promissor processo de minimização do impacto ambiental.
