    Description: The persistent open-vent activity of basaltic volcanoes is periodically interrupted by spectacular but hazardous paroxysmal explosions. The rapid transition from quiescence to explosive eruption poses a significant challenge for volcanic hazard assessment and mitigation, and improving our understanding of the processes that trigger these paroxysmal events is critical. Although magmatic gas is unquestionably the driver, direct measurements of a paroxysm's gas flux budget have remained challenging, to date. A particularly violent paroxysmal sequence took place on Etna on December 2015, intermittently involving all summit craters, especially the Voragine (VOR) that had previously displayed no activity for several years. Here, we characterize the volcano's SO2 degassing budget prior to, during and after this paroxysmal sequence, using ground-based (UV-Camera) and satellite (OMI) observations, complemented with ground- and space-borne thermal measurements. We make use of the high spatial resolution of UV-cameras to resolve SO2 emissions from the erupting VOR crater for the first time, and to characterize temporal switches in degassing activity from VOR to the nearby New Southeast Crater (NSEC). Our data show that onset of paroxysmal activity on December 3–5 was marked by visible escalation in VOR SO2 fluxes (4,700–8,900 tons/day), in satellite-derived thermal emissions (2,000 MW vs. ~2–11 MW in July-November 2015), and in OMI-derived daily SO2 masses (5.4 ± 0.7 to 10.0 ± 1.3 kilotonnes, kt; 0.5 kt was the average in the pre-eruptive period). Switch in volcanic activity from VOR to NSEC on December 6 was detected by increasing SO2 fluxes at the NSEC crater, and by decaying SO2 emissions at VOR, until activity termination on December 19. Taken together, our observations infer the total degassed SO2 mass for the entire VOR paroxysmal sequence at 21,000 ± 2,730 t, corresponding to complete degassing of ~1.9 ± 0.3 Mm3 of magma, or significantly less than the measured erupted magma volumes (5.1–12 Mm3). From this mismatch we propose that only a small fraction of the erupted magma was actually emplaced in the shallow plumbing system during (or shortly prior) the paroxysmal sequence. Rather, the majority of the erupted magma was likely stored conduit magma, having gone through extensive degassing for days to weeks prior to the paroxysm.

    Description: The aim of this study was to assess potential candidate gene regions and corresponding universal primer pairs as secondary DNA barcodes for the fungal kingdom, additional to ITS rDNA as primary barcode. Amplification efficiencies of 14 (partially) universal primer pairs targeting eight genetic markers were tested across 〉 1 500 species (1 931 strains or specimens) and the outcomes of almost twenty thousand (19 577) polymerase chain reactions were evaluated. We tested several well-known primer pairs that amplify: i) sections of the nuclear ribosomal RNA gene large subunit (D1–D2 domains of 26/28S); ii) the complete internal transcribed spacer region (ITS1/2); iii) partial β-tubulin II (TUB2); iv) γ-actin (ACT); v) translation elongation factor 1-α (TEF1α); and vi) the second largest subunit of RNA-polymerase II (partial RPB2, section 5–6). Their PCR efficiencies were compared with novel candidate primers corresponding to: i) the fungal-specific translation elongation factor 3 (TEF3); ii) a small ribosomal protein necessary for t-RNA docking; iii) the 60S L10 (L1) RP; iv) DNA topoisomerase I (TOPI); v) phosphoglycerate kinase (PGK); vi) hypothetical protein LNS2; and vii) alternative sections of TEF1α. Results showed that several gene sections are accessible to universal primers (or primers universal for phyla) yielding a single PCR-product. Barcode gap and multi-dimensional scaling analyses revealed that some of the tested candidate markers have universal properties providing adequate infra- and inter-specific variation that make them attractive barcodes for species identification. Among these gene sections, a novel high fidelity primer pair for TEF1α, already widely used as a phylogenetic marker in mycology, has potential as a supplementary DNA barcode with superior resolution to ITS. Both TOPI and PGK show promise for the Ascomycota, while TOPI and LNS2 are attractive for the Pucciniomycotina, for which universal primers for ribosomal subunits often fail.

    Description: Offering a scientific perspective, this paper provides a rationale for the use of dispersants in oil spill remediation by discussing their formulations and modes of action and connecting their physics and chemistry to a their environmental fates and impacts. With the first use of dispersants at the source of the oil release during the Deepwater Horizon incident, there is a new great need for understanding the efficiency and the environmental impacts of their use. The paper concludes with some cautionary recommendations on dispersant research.

    Description: Coral reef communities between 26.8°N and 18.6°N latitude in the Saudi Arabian Red Sea were surveyed to provide baseline data and an assessment of fine-scale biogeography of communities in this region. Forty reefs along 1100 km of coastline were surveyed using depth-stratified visual transects of fish and benthic communities. Fish abundance and benthic cover data were analyzed using multivariate approaches to investigate whether coral reef communities differed with latitude. A total of 215 fish species and 90 benthic categories were recorded on the surveys. There were no significant differences among locations in fish abundance, species richness, or among several diversity indices. Despite known environmental gradients within the Red Sea, the communities remained surprisingly similar. The communities do, however, exhibit subtle changes across this span of reefs that likely reflect the constrained distributions of several species of reef fish and benthic fauna.

    Description: Transitions towards sustainability are urgently needed to address the interconnected challenges of economic development, ecological integrity, and social justice, from local to global scales. Around the world, collaborative science-society initiatives are forming to conduct experiments in support of sustainability transitions. Such experiments, if carefully designed, provide significant learning opportunities for making progress on transition efforts. Yet, there is no broadly applicable evaluative scheme available to capture this critical information across a large number of cases, and to guide the design of transition experiments. To address this gap, the article develops such a scheme, in a tentative form, drawing on evaluative research and sustainability transitions scholarship, alongside insights from empirical cases. We critically discuss the scheme's key features of being generic, comprehensive, operational, and formative. Furthermore, we invite scholars and practitioners to apply, reflect and further develop the proposed tentative scheme - making evaluation and experiments objects of learning.

    Description: The calving of A-68, the 5,800 km2 iceberg that separated from the Larsen C Ice Shelf in July 2017, presents a unique and time-sensitive research opportunity. This event and other ice-shelf losses (e.g., from Larsen A and B, Wilkins, Wordie) are harbingers of warming effects along the Antarctic Peninsula in particular, and ultimately around all of Antarctica. The scientific momentum and public interest created by this most recent event led to an NSF funded workshop in November 2017. A consortium of US and international researchers with a diversity of expertise in Antarctic glaciological, geological, biological, and ecological sciences shared and reviewed the latest research, identified important research priorities and knowlegde gaps, and outlined strategic plans for the research community to advance understanding of the continent-wide changes that Antarctic ice shelves and surrounding ecosystems will experience in response to warming. The workshop has leveraged an opportunity to synergize efforts in investigating Antarctic ecosystems under the direct and indirect effects of ice-shelf collapse, and climatic warming in general. Here we present a review on the known and unknown ecosystem processes that will occur in the wake of massive, abrupt ice-shelf break-off and how they will affect ice-associated, pelagic, hard-bottom and soft-sediment substrates. We also present a view on future research approaches to address gaps in our knowledge and advance our understanding of the widespread effects of ice-shelf break-off and collapse.

    Description: Whole-organism performance tasks are accomplished by the integration of morphological traits and physiological functions. Understanding how evolutionary change in morphology and physiology influences whole-organism performance will yield insight into the factors that shape its own evolution. We demonstrate that nonmigratory populations of alewife (Alosa pseudoharengus) have evolved reduced swimming performance in parallel, compared with their migratory ancestor. In contrast to theoretically and empirically based predictions, poor swimming among nonmigratory populations is unrelated to the evolution of osmoregulation and occurs despite the fact that nonmigratory alewives have a more fusiform (torpedo-like) body shape than their ancestor. Our results suggest that elimination of long-distance migration from the life cycle has shaped performance more than changes in body shape and physiological regulatory capacity.

    Description: KRAS mutations occur in ∼35% of colorectal cancers and promote tumor growth by constitutively activating the mitogen-activated protein kinase (MAPK) pathway. KRAS mutations at codons 12, 13, or 61 are thought to prevent GAP protein-stimulated GTP hydrolysis and render KRAS-mutated colorectal cancers unresponsive to epidermal growth factor receptor (EGFR) inhibitors. We report here that KRAS G13-mutated cancer cells are frequently comutated with NF1 GAP but NF1 is rarely mutated in cancers with KRAS codon 12 or 61 mutations. Neurofibromin protein (encoded by the NF1 gene) hydrolyzes GTP directly in complex with KRAS G13D, and KRAS G13D-mutated cells can respond to EGFR inhibitors in a neurofibromin-dependent manner. Structures of the wild type and G13D mutant of KRAS in complex with neurofibromin (RasGAP domain) provide the structural basis for neurofibromin-mediated GTP hydrolysis. These results reveal that KRAS G13D is responsive to neurofibromin-stimulated hydrolysis and suggest that a subset of KRAS G13-mutated colorectal cancers that are neurofibromin-competent may respond to EGFR therapies.
