    Description: In the low-redshift Universe, the most powerful radio sources are often associated with gas-rich galaxy mergers or interactions. We here present evidence for an advanced, gas-rich (‘wet’) merger associated with a powerful radio galaxy at a redshift of z  ~ 2. This radio galaxy, MRC 0152-209, is the most infrared-luminous high-redshift radio galaxy known in the Southern hemisphere. Using the Australia Telescope Compact Array, we obtained high-resolution CO(1–0) data of cold molecular gas, which we complement with Hubble Space Telescope ( HST )/ Wide Field Planetary Camera 2 ( WFPC 2) imaging and William Herschel Telescope long-slit spectroscopy. We find that, while roughly M H2  ~ 2 10 10  M of molecular gas coincides with the central host galaxy, another M H2  ~ 3 10 10  M is spread across a total extent of ~60 kpc. Most of this widespread CO(1–0) appears to follow prominent tidal features visible in the rest-frame near-UV HST / WFPC 2 imaging. Lyα emission shows an excess over He II, but a deficiency over L IR , which is likely the result of photoionization by enhanced but very obscured star formation that was triggered by the merger. In terms of feedback, the radio source is aligned with widespread CO(1–0) emission, which suggests that there is a physical link between the propagating radio jets and the presence of cold molecular gas on scales of the galaxy's halo. Its optical appearance, combined with the transformational stage at which we witness the evolution of MRC 0152-209, leads us to adopt the name ‘Dragonfly Galaxy’.

    Description: As a discipline, marine historical ecology (MHE) has contributed significantly to our understanding of the past state of the marine environment when levels of human impact were often very different from those today. What is less widely known is that insights from MHE have made headway into being applied within the context of present-day and long-term management and policy. This study draws attention to the applied value of MHE. We demonstrate that a broad knowledge base exists with potential for management application and advice, including the development of baselines and reference levels. Using a number of case studies from around the world, we showcase the value of historical ecology in understanding change and emphasize how it either has already informed management or has the potential to do so soon. We discuss these case studies in a context of the science–policy interface around six themes that are frequently targeted by current marine and maritime policies: climate change, biodiversity conservation, ecosystem structure, habitat integrity, food security, and human governance. We encourage science–policy bodies to actively engage with contributions from MHE, as well-informed policy decisions need to be framed within the context of historical reference points and past resource or ecosystem changes.

    Description: The approach to fisheries termed "balanced harvesting" (BH) calls for fishing across the widest possible range of species, stocks, and sizes in an ecosystem, in proportion to their natural productivity, so that the relative size and species composition is maintained. Such fishing is proposed to result in higher catches with less negative impact on exploited populations and ecosystems. This study examines the models and the empirical evidence put forward in support of BH. It finds that the models used unrealistic settings with regard to life history (peak of cohort biomass at small sizes), response to fishing (strong compensation of fishing mortality by reduced natural mortality), and economics (uniform high cost of fishing and same ex-vessel price for all species and sizes), and that empirical evidence of BH is scarce and questionable. It concludes that evolutionary theory, population dynamics theory, ecosystem models with realistic assumptions and settings, and widespread empirical evidence do not support the BH proposal. Rather, this body of evidence suggests that BH will not help but will hinder the policy changes needed for the rebuilding of ecosystems, healthy fish populations, and sustainable fisheries.

    Description: In a recent publication (Froese et al. , ICES Journal of Marine Science ; 73: 1640–1650), we presented a critique of the balanced harvesting (BH) approach to fishing. A short section dealt with the size-spectrum models used to justify BH, wherein we pointed out the lack of realism of these models, which mostly represented ecosystems as consisting of a single cannibalistic species. Andersen et al. ( ICES Journal of Marine Science ; 73: 1651–1655) commented on our paper and suggested that we criticized size-spectrum models in general and that we supposedly made several erroneous statements. We stress that we only referred to the size-spectrum models that we cited, and we respond to each supposedly erroneous statement. We still believe that the size-spectrum models used to justify BH were highly unrealistic and not suitable for evaluating real-world fishing strategies. We agree with Andersen et al. that BH is unlikely to be a useful guiding principle for ecosystem-based fisheries management, for many reasons. The use of unrealistic models is one of them.

    Description: Cyanobacteria are the primary taxa responsible for freshwater harmful algal blooms (HABs), with several genera capable of producing potent intracellular toxins and off-flavor compounds. There is considerable growing interest in methods to rapidly quantify cyanobacteria in water samples. Past studies have demonstrated poor correlations between phycocyanin in vivo fluorescence and cyanobacterial cell densities. We conducted a series of laboratory experiments aimed at refining a protocol that uses benchtop fluorometry to measure the cyanobacterial pigment, phycocyanin, to accurately estimate cyanobacterial biovolume. In our study, we found strong correlations between phycocyanin concentration and cyanobacterial biovolume (but not for cell densities) both within and across ponds, which varied widely in productivity and algal diversity. Thus, benchtop fluorometry of phycocyanin is a viable method for water resource managers to quickly estimate cyanobacterial biovolume.

    Description: SKS arrivals from ocean bottom seismometer (OBS) data from an offshore southern California deployment are analysed for shear wave splitting. The project involved 34 OBSs deployed for 12 months in a region extending up to 500 km west of the coastline into the oceanic Pacific plate. The measurement process consisted of removing the effects of anisotropy using a range of values for splitting fast directions and delay times to minimize energy along the transverse seismometer axis. Computed splitting parameters are unexpectedly similar to onland parameters, exhibiting WSW–ENE fast polarization directions and delays between 0.8 and 1.8 s, even for oceanic plate sites. This is the first SKS splitting study to extend across the entire boundary between the North America and Pacific plates, into the oceanic part of the Pacific plate. The splitting results show that the fast direction of anisotropy on the Pacific plate does not align with absolute plate motion (APM), and they extend the trend of anisotropy in southern California an additional 500 km west, well onto the oceanic Pacific plate. We model the finite strain and anisotropy within the asthenosphere associated with density–buoyancy driven mantle flow and the effects of APM. In the absence of plate motion effects, such buoyancy driven mantle flow would be NE-directed beneath the Pacific plate observations. The best-fit patterns of mantle flow are inferred from the tomography-based models that show primary influences from foundering higher-density zones associated with the history of subduction beneath North America. The new offshore SKS measurements, when combined with measurements onshore within the plate boundary zone, indicate that dramatic lateral variations in density-driven upper-mantle flow are required from offshore California into the plate boundary zone in California and western Basin and Range.

    Description: Oxidative damage to mitochondria (MT) is a major mechanism for aging and neurodegeneration. We have developed a novel synthetic antioxidant, XJB-5-131, which directly targets MT, the primary site and primary target of oxidative damage. XJB-5-131 prevents the onset of motor decline in an HdhQ(150/150) mouse model for Huntington's disease (HD) if treatment starts early. Here, we report that XJB-5-131 attenuates or reverses disease progression if treatment occurs after disease onset. In animals with well-developed pathology, XJB-5-131 promotes weight gain, prevents neuronal death, reduces oxidative damage in neurons, suppresses the decline of motor performance or improves it, and reduces a graying phenotype in treated HdhQ(150/150) animals relative to matched littermate controls. XJB-5-131 holds promise as a clinical candidate for the treatment of HD.

    Description: DNA structure and topology pervasively influence aspects of DNA metabolism including replication, transcription and segregation. However, the effects of DNA topology on DNA-protein interactions have not been systematically explored due to limitations of standard affinity assays. We developed a method to measure protein binding affinity dependence on the topology (topological linking number) of supercoiled DNA. A defined range of DNA topoisomers at equilibrium with a DNA binding protein is separated into free and protein-bound DNA populations using standard nitrocellulose filter binding techniques. Electrophoretic separation and quantification of bound and free topoisomers combined with a simple normalization procedure provide the relative affinity of the protein for the DNA as a function of linking number. Employing this assay we measured topology-dependent DNA binding of a helicase, a type IB topoisomerase, a type IIA topoisomerase, a non-specific mitochondrial DNA binding protein and a type II restriction endonuclease. Most of the proteins preferentially bind negatively supercoiled DNA but the details of the topology-dependent affinity differ among proteins in ways that expose differences in their interactions with DNA. The topology-dependent binding assay provides a robust and easily implemented method to probe topological influences on DNA-protein interactions for a wide range of DNA binding proteins.

    Description: Klebsiella pneumoniae clonal group (CG) 258, comprising sequence types (STs) 258, 11, and closely related variants, is associated with dissemination of the K. pneumoniae carbapenemase (KPC). Hospital outbreaks of KPC CG258 infections have been observed globally and are very difficult to treat. As a consequence, there is renewed interest in alternative infection control measures such as vaccines and phage or depolymerase treatments targeting the K. pneumoniae polysaccharide capsule. To date, 78 immunologically distinct capsule variants have been described in K. pneumoniae. Previous investigations of ST258 and a small number of closely related strains suggested that capsular variation was limited within this clone; only two distinct ST258 capsule polysaccharide synthesis ( cps ) loci have been identified, both acquired through large-scale recombination events (〉50 kb). In contrast to previous studies, we report a comparative genomic analysis of the broader K. pneumoniae CG258 ( n = 39). We identified 11 different cps loci within CG258, indicating that capsular switching is actually common within the complex. We observed several insertion sequences (IS) within the cps loci, and show further intraclone diversification of two cps loci through IS activity. Our data also indicate that several large-scale recombination events have shaped the genomes of CG258, and that definition of the complex should be broadened to include ST395 (also reported to harbor KPC). As only the second report of extensive intraclonal cps variation among Gram-negative bacterial species, our findings alter our understanding of the evolution of these organisms and have key implications for the design of control measures targeting K. pneumoniae capsules.
