    Description: In the course of this century, rising anthropogenic CO2 emissions will likely cause a decrease in ocean pH, know as ocean acidification, together with an increase of water temperature. Only in the last years, studies have focused on synergetic effects of both stressors on marine invertebrates, particularly on early life stages considered more vulnerable. Disparate responses of their singular and combined effects were reported, highlighting the importance of extending the studies to different species and populations of marine invertebrates. Here, we observed the response of important parameters such as growth, mortality and oxygen consumption of juvenile pen shell Pinna nobilis at supplied pCO2 gas levels of 400 ppm (ambient) and 1000 ppm and at three temperatures (20, 23 and 26 °C) during 36 days. To our knowledge, this is the first study on ocean acidification and temperature effects on juveniles of this species. We show that the two stressors play roles at distinct levels, with pCO2 influencing growth and partially mortality, and temperature increasing mortality rates and oxygen consumption strongly. Therefore, juveniles of P. nobilis are more likely affected by increasing temperature than the pCO2 levels expected by the end of the twenty-first century.

    Notes: Summary The effect of ceruletide® (CRL), a synthetic decapeptide analogue of cholecystokinin, on rest pain and arterial blood flow was evaluated in 8 patients with advanced, occlusive atherosclerosis of the lower extremities. CRL 1, 2, or 4 ng kg−1 or placebo were infused intravenously in random order, and in a double-blind fashion. Pain relief, assessed by a scoring system, was significantly better (p〈0.01) following the 2 and 4 ng kg−1 doses of CRL (2.71 and 2.66, respectively) than following placebo (0.75). Arterial blood flow was not affected by either CRL in any dose or by placebo. Pretreatment with naloxone, a pure opioid antagonist, abolished the analgesic effect of CRL. Following the 2 ng dose of CRL, β-endorphin levels were significantly elevated from a basal value of 125±15 pg/ml to 191±35 pg/ml 5 h after CRL administration (p〈0.05). Circulating levels of ACTH, prolactin and GH were not affected by CRL. It is concluded that CRL was effective in relieving ischaemic rest pain, and that the mechanism was related to the release of endogenous opioids.

    Description: The Mar Piccolo basin is an internal sea basin located along the Ionian coast (Southern Italy), and it is surrounded primarily by fractured carbonate karstic environment. Because of the karstic features, the main continental water inflow is from groundwater discharge. The Mar Piccolo basin represents a peculiar and sensitive environment and a social emergency because of sea water and sediments pollution. This pollution appears to be caused by the overlapping effects of dangerous anthropogenic activities, including heavy industries and commercial and navy dockyards. The paper aims to define the contribution of subaerial and submarine coastal springs to the hydrological dynamic equilibrium of this internal sea basin. A general approach was defined, including a hydrogeological basin border assessment to detect inflowing springs, detailed geological and hydrogeological conceptualisation, in situ submarine and subaerial spring measurements, and flow numerical modelling. Multiple sources of data were obtained to define a relevant geodatabase, and it contained information on approximately 2,000 wells, located in the study area (1,600 km2). The conceptualisation of the hydrogeological basin, which is 978 km2 wide, was supported by a 3D geological model that interpolated 716 stratigraphic logs. The variability in hydraulic conductivity was determined using hundreds of pumping tests. Five surveys were performed to acquire hydro-geochemical data and spring flow-yield measurements; the isotope groundwater age was assessed and used for model validation. The mean annual volume exchanged by the hydrogeological basin was assessed equal to 106.93 106 m3. The numerical modelling permitted an assessment of the mean monthly yield of each spring outflow (surveyed or not), travel time, and main path flow.

    Description: A good characterization of the geomagnetic field strength at centennial to millennial time scales in the Southern Hemisphere is particularly crucial to disentangle the long-term evolution of the South Atlantic Anomaly (SAA), an intriguing geomagnetic feature currently observed at the Earth’s surface. Here we present 59 new archeointensities obtained from four well-dated groups of potteries with ages ranging between 1300 and 1500 CE and collected in Northwest Argentina. The new data were obtained in accordance with the Thellier paleointensity method including partial thermoremanent magnetization (pTRM) checks and TRM anisotropy and cooling rate corrections. We have also performed a comparative study of the efficiency of magnetic susceptibility, ARM and TRM anisotropy tensors to correct the TRM anisotropy effect upon intensity estimates. Our results suggest that the magnetic susceptibility tensor systematically underestimates the TRM anisotropy effect by 10–30%. Our new data, together with selected archeointensities already published, confirm that the decrease of the geomagnetic field intensity in South America started around 1600–1650 CE, due to the arrival of the SAA.

    Description: A wide variety of processes controls the time of occurrence, duration, extent, and severity of river floods. Classifying flood events by their causative processes may assist in enhancing the accuracy of local and regional flood frequency estimates and support the detection and interpretation of any changes in flood occurrence and magnitudes. This paper provides a critical review of existing causative classifications of instrumental and preinstrumental series of flood events, discusses their validity and applications, and identifies opportunities for moving toward more comprehensive approaches. So far no unified definition of causative mechanisms of flood events exists. Existing frameworks for classification of instrumental and preinstrumental series of flood events adopt different perspectives: hydroclimatic (large‐scale circulation patterns and atmospheric state at the time of the event), hydrological (catchment scale precipitation patterns and antecedent catchment state), and hydrograph‐based (indirectly considering generating mechanisms through their effects on hydrograph characteristics). All of these approaches intend to capture the flood generating mechanisms and are useful for characterizing the flood processes at various spatial and temporal scales. However, uncertainty analyses with respect to indicators, classification methods, and data to assess the robustness of the classification are rarely performed which limits the transferability across different geographic regions. It is argued that more rigorous testing is needed. There are opportunities for extending classification methods to include indicators of space–time dynamics of rainfall, antecedent wetness, and routing effects, which will make the classification schemes even more useful for understanding and estimating floods.

    Description: Pain is a main symptom of inflammatory diseases and often persists beyond clinical remission. Although we have a good understanding of the mechanisms of sensitization at the periphery during inflammation, little is known about the mediators that drive central sensitization. Recent reports have identified hematopoietic colony-stimulating factors as important regulators of tumor- and nerve injury-associated pain. Using a mouse model of colitis, we identify the proinflammatory cytokine granulocyte-colony–stimulating factor (G-CSF or Csf-3) as a key mediator of visceral sensitization. We report that G-CSF is specifically up-regulated in the thoracolumbar spinal cord of colitis-affected mice. Our results show that resident spinal microglia express the G-CSF receptor and that G-CSF signaling mediates microglial activation following colitis. Furthermore, healthy mice subjected to intrathecal injection of G-CSF exhibit pronounced visceral hypersensitivity, an effect that is abolished by microglial depletion. Mechanistically, we demonstrate that G-CSF injection increases Cathepsin S activity in spinal cord tissues. When cocultured with microglia BV-2 cells exposed to G-CSF, dorsal root ganglion (DRG) nociceptors become hyperexcitable. Blocking CX3CR1 or nitric oxide production during G-CSF treatment reduces excitability and G-CSF–induced visceral pain in vivo. Finally, administration of G-CSF–neutralizing antibody can prevent the establishment of persistent visceral pain postcolitis. Overall, our work uncovers a DRG neuron–microglia interaction that responds to G-CSF by engaging Cathepsin S-CX3CR1-inducible NOS signaling. This interaction represents a central step in visceral sensitization following colonic inflammation, thereby identifying spinal G-CSF as a target for treating chronic abdominal pain.
