Notizen: Summary Stimulation of secretion by pilocarpine results in a 70% loss of zymogen granules from pancreatic acinar cell during the first hr after injection of the drug. In previous work (Geuze and Poort, 1973), we found that the amount of membrane stored in the surface of the microvilli and of the numerous infoldings present in highly stimulated cells, increases during the first 2 hr and then decreases again during the 3rd hr after stimulation, concurrently with maximal endocytosis of sorbitol-[su14C]. Further observations on the fine structure of stimulated cells at various time intervals after injection of pilocarpine showed that during the first hr numerous smooth vesicles and multivesicular bodies (mvb's) appear in the apical cytoplasm, while the number of coated vesicles and their relative total volume increase significantly 3 hr after stimulation. By infusion of ferritin in the pancreatic duct system in vivo and application of cytochemical techniques (osmium impregnation, electron microscope autoradiography and acid phosphatase cytochemistry) it could be established that after stimulated exocytotic secretion, redundant apical cell membrane is withdrawn by at least two routes: 1) During the initial rapid increase of the amount of apical cell membrane, withdrawal is accomplished by interiorization of luminal invaginations into smooth endocytotic vesicles, which in turn give rise to mvb's by infolding and subsequent fission of their limiting membrane. 2) Once the bulk of stored secretion granules has been discharged, endocytotic coated vesicles become gradually more prominent as carriers for redundant cell membrane. The contents of endocytotic structures ultimately become incorporated in residual bodies, suggesting lysosomal degradation of cell membrane prior to eventual reutilization. Coated vesicles also originate by pinching off from mature Golgi cisternae and condensing vacuoles. A possible function of the coated membranes in the concentration of exportable protein within forming secretory granules is discussed.

Notizen: Abstract Filar micrometer measurements of the central body during its conversion to the vegetative cell indicated an initial spherical body of about 1.7 microns in diameter. The central body first increases in diameter then elongates such that its length is 2–2.5 times the diameter. Central bodies supplemented with casein hydrolysate in addition to sucrose formed vegetative cells in only 8 hr instead of the usual 12. Central bodies were quite susceptible to lysis by EDTA-lysozyme or citrate-lysozyme, but relatively insensitive to citrate-EDTA or single employment of EDTA or citrate. Comparative electron microscopy of cysts and central bodies following sonication revealed little alteration in cysts but reduction of central bodies to cellular debris after only 2 min. Central bodies were resistant to bacteriophage adsorption at 0 hr and required about 4 hr incubation before adsorption could occur.

Notizen: An overall mechanistic scheme for the suspension polymerization of vinyl chloride is presented. The process can be resolved into five discrete stages, each of which presents a unique environment for the interaction of the systems parameters. It is shown that the surface area of the polymer formed during the reaction is not a major factor in autoacceleration and that the increase of kinetic chain length with conversion is due to a radical dilution effect. The latter is a direct result of the difference in rates between polymerization and radical formation, the former being greater. The increase of the initial polymerization rate and the reduction of autoacceleration brought about by chain transfer agents can be explained by the lower diffusion rate and greater bulkiness of the chain transfer agent radical relative to that of the monomer radical. The chaintransfer agent CBr4 is preferentially absorbed by PVC from solution in vinyl chloride. With lauryl peroxide as initiator it is shown that the “hot spot” is the result of a build-up of initiator in the monomer caused by its exclusion from the polymer phase. Vinyl chloride was found to dissolve 0.03% PVC at ambient temperature and to have no effect on the decomposition rate of lauryl peroxide.

Beschreibung: We present timing solutions and analyses of 11 pulsars discovered by the Five-hundred-meter Aperture Spherical radio Telescope (FAST). These pulsars were discovered using an ultrawide bandwidth receiver in drift-scan observations made during the commissioning phase of FAST, and were then confirmed and timed using the 64-m Parkes Radio Telescope. Each pulsar has been observed over a span of at least one year. Highlighted discoveries include PSR J0344−0901, which displays mode-changing behaviour and may belong to the class of so-called swooshing pulsars (alongside PSRs B0919+06 and B1859+07); PSR J0803−0942, whose emission is almost completely linearly polarized; and PSRs J1900−0134 and J1945+1211, whose well-defined polarization angle curves place stringent constraints on their emission geometry. We further discuss the detectability of these pulsars by earlier surveys, and highlight lessons learned from our work in carrying out confirmation and monitoring observations of pulsars discovered by a highly sensitive telescope, many of which may be applicable to next-generation pulsar surveys. This paper marks one of the first major releases of FAST-discovered pulsars, and paves the way for future discoveries anticipated from the Commensal Radio Astronomy FAST Survey.

Beschreibung: Context. The gravitational strong equivalence principle (SEP) is a cornerstone of the general theory of relativity (GR). Hence, testing the validity of SEP is of great importance when confronting GR, or its alternatives, with experimental data. Pulsars that are orbited by white dwarf companions provide an excellent laboratory, where the extreme difference in binding energy between neutron stars and white dwarfs allows for precision tests of the SEP via the technique of radio pulsar timing. Aims. To date, the best limit on the validity of SEP under strong-field conditions was obtained with a unique pulsar in a triple stellar system, PSR J0337+1715. We report here on an improvement of this test using an independent data set acquired over a period of 6 years with the Nançay radio telescope. The improvements arise from a uniformly sampled data set, a theoretical analysis, and a treatment that fixes some short-comings in the previously published results, leading to better precision and reliability of the test. Methods. In contrast to the previously published test, we use a different long-term timing data set, developed a new timing model and an independent numerical integration of the motion of the system, and determined the masses and orbital parameters with a different methodology that treats the parameter Δ, describing a possible strong-field SEP violation, identically to all other parameters. Results. We obtain a violation parameter Δ = ( + 0.5 ± 1.8) × 10−6 at 95% confidence level, which is compatible with and improves upon the previous study by 30%. This result is statistics-limited and avoids limitation by systematics as previously encountered. We find evidence for red noise in the pulsar spin frequency, which is responsible for up to 10% of the reported uncertainty. We use the improved limit on SEP violation to place constraints on a class of well-studied scalar-tensor theories, in particular we find ωBD 〉  140 000 for the Brans-Dicke parameter. The conservative limits presented here fully take into account current uncertainties in the equation for state of neutron-star matter.

Beschreibung: Context. Realistic synthetic observations of theoretical source models are essential for our understanding of real observational data. In using synthetic data, one can verify the extent to which source parameters can be recovered and evaluate how various data corruption effects can be calibrated. These studies are the most important when proposing observations of new sources, in the characterization of the capabilities of new or upgraded instruments, and when verifying model-based theoretical predictions in a direct comparison with observational data. Aims. We present the SYnthetic Measurement creator for long Baseline Arrays (SYMBA), a novel synthetic data generation pipeline for Very Long Baseline Interferometry (VLBI) observations. SYMBA takes into account several realistic atmospheric, instrumental, and calibration effects. Methods. We used SYMBA to create synthetic observations for the Event Horizon Telescope (EHT), a millimetre VLBI array, which has recently captured the first image of a black hole shadow. After testing SYMBA with simple source and corruption models, we study the importance of including all corruption and calibration effects, compared to the addition of thermal noise only. Using synthetic data based on two example general relativistic magnetohydrodynamics (GRMHD) model images of M 87, we performed case studies to assess the image quality that can be obtained with the current and future EHT array for different weather conditions. Results. Our synthetic observations show that the effects of atmospheric and instrumental corruptions on the measured visibilities are significant. Despite these effects, we demonstrate how the overall structure of our GRMHD source models can be recovered robustly with the EHT2017 array after performing calibration steps, which include fringe fitting, a priori amplitude and network calibration, and self-calibration. With the planned addition of new stations to the EHT array in the coming years, images could be reconstructed with higher angular resolution and dynamic range. In our case study, these improvements allowed for a distinction between a thermal and a non-thermal GRMHD model based on salient features in reconstructed images.

Beschreibung: We present the results from the low-frequency (40–78 MHz) extension of the first pulsar census of non-recycled pulsars carried out with the LOw-Frequency ARray (LOFAR). We used the low-band antennas of the LOFAR core stations to observe 87 pulsars out of 158 that had been previously detected using high-band antennas. We present flux densities and flux-calibrated profiles for the 43 pulsars we detected. Of this sample, 17 have not, to our knowledge, previously been detected at such low frequencies. Here we recalculate the spectral indices using the new low-frequency flux density measurements from the LOFAR census and discuss the prospects of studying pulsars at very low frequencies using current and upcoming facilities, such as the New Extension in Nançay Upgrading LOFAR (NenuFAR).
