    Description: Phosphorylation of histone H4 serine 47 (H4S47ph) is catalyzed by Pak2, a member of the p21-activated serine/threonine protein kinase (Pak) family and regulates the deposition of histone variant H3.3. However, the phosphatase(s) involved in the regulation of H4S47ph levels was unknown. Here, we show that three phosphatases (PP1α, PP1β and Wip1) regulate H4S47ph levels and H3.3 deposition. Depletion of each of the three phosphatases results in increased H4S47ph levels. Moreover, PP1α, PP1β and Wip1 bind H3-H4 in vitro and in vivo , whereas only PP1α and PP1β, but not Wip1, interact with Pak2 in vivo . These results suggest that PP1α, PP1β and Wip1 regulate the levels of H4S47ph through directly acting on H4S47ph, with PP1α and PP1β also likely regulating the activity of Pak2. Finally, depletion of PP1α, PP1β and Wip1 leads to increased H3.3 occupancy at candidate genes tested, elevated H3.3 deposition and enhanced association of H3.3 with its chaperones HIRA and Daxx. These results reveal a novel role of three phosphatases in chromatin dynamics in mammalian cells.

    Description: Simulating seismic waves with uniform grid in heterogeneous high-velocity contrast media requires small-grid spacing determined by the global minimal velocity, which leads to huge number of grid points and small time step. To reduce the computational cost, discontinuous grids that use a finer grid at the shallow low-velocity region and a coarser grid at high-velocity regions are needed. In this paper, we present a discontinuous grid implementation for the collocated-grid finite-difference (FD) methods to increase the efficiency of seismic wave modelling. The grid spacing ratio n could be an arbitrary integer n  ≥ 2. To downsample the wavefield from the finer grid to the coarser grid, our implementation can simply take the values on the finer grid without employing a downsampling filter for grid spacing ratio n  = 2 to achieve stable results for long-time simulation. For grid spacing ratio n  ≥ 3, the Gaussian filter should be used as the downsampling filter to get a stable simulation. To interpolate the wavefield from the coarse grid to the finer grid, the trilinear interpolation is used. Combining the efficiency of discontinuous grid with the flexibility of collocated-grid FD method on curvilinear grids, our method can simulate large-scale high-frequency strong ground motion of real earthquake with consideration of surface topography.

    Description: On the morning of 14 April 2010, the M s  7.1 Yushu earthquake struck the eastern Tibet Plateau and an M s  6.3 aftershock followed west-southwest of the mainshock epicenter one and a half hours later. The Yushu earthquake occurred on the northwestern continuation of the Ganzi–Xianshuihe fault zone and reactivated two segments of the fault. Ruptures associated with Yushu earthquake span a zone 70 km long, extending from south of Jielong in the west to south of Gyêgu (Yushu) in the east. Detailed mapping of the surface rupture zone shows that it consists of two strike-slip segments, the western and eastern segments, separated by the Longbao Lake step-over (pull apart) basin. Our analysis suggests that the 18-km-long western segment of the surface rupture south of the Longbao Lake basin may have been associated with the M s  6.3 aftershock. The eastern rupture segment, which is inferred to be associated with the mainshock, is 32 km long. The main surface rupture can also be divided into two sections, with a 2-km-long gap between them. The surface rupture exhibits left-lateral slip and is localized in a zone about 50 m wide along the present-day trace of the Yushu fault. A single break, without splays or branching traces, characterizes most of the surface rupture zone. The observed magnitudes of surface displacement are typically 0.5–1 m with a maximum slip of 2 m. Both field observations and seismic inversion results suggest, rupture of M s  7.1 Yushu earthquake nucleated near the Longbao Lake step-over basin in the west, propagated unilaterally eastward, and terminated within the east segment margin of the Yushu fault.

    Description: Protein-coding genes account for only a small part of the human genome, whereas the vast majority of transcripts make up the non-coding RNAs including long non-coding RNAs (lncRNAs). Accumulating evidence indicates that lncRNAs could play a critical role in regulation of cellular processes such as cell growth and apoptosis as well as cancer progression and metastasis. LncRNA loc285194 was previously shown to be within a tumor suppressor unit in osteosarcoma and to suppress tumor cell growth. However, it is unknown regarding the regulation of loc285194. Moreover, the underlying mechanism by which loc285194 functions as a potential tumor suppressor is elusive. In this study, we show that loc285194 is a p53 transcription target; ectopic expression of loc285194 inhibits tumor cell growth both in vitro and in vivo . Through deletion analysis, we identify an active region responsible for tumor cell growth inhibition within exon 4, which harbors two miR-211 binding sites. Importantly, this loc285194-mediated growth inhibition is in part due to specific suppression of miR-211. We further demonstrate a reciprocal repression between loc285194 and miR-211; in contrast to loc285194, miR-211 promotes cell growth. Finally, we detect downregulation of loc285194 in colon cancer specimens by quantitative PCR arrays and in situ hybridization of tissue microarrays. Together, these results suggest that loc285194 is a p53-regulated tumor suppressor, which acts in part through repression of miR-211.

    Description: Paleoclimatic research can provide critical insight on causes of change in the East Asian monsoon, which influences the lives of 1.6 billion people today. In this study, we use paleoclimatic indexes from Chinese loess deposits, which have clear climatic implications and are independently dated, to reconstruct the monsoon precipitation since 21 ka. Our results show that monsoon precipitation persistently decreased from 21 ka to ca. 8 ka, and increased after ca. 8 ka, with a precipitation peak at 8–3 ka. These changes in East Asian summer monsoon precipitation are synchronous with changes in high-northern-latitude ice volume/ice cover and atmospheric CO 2 . These new data suggest that variation of the monsoon precipitation was probably driven by CO 2 -forced high-northern-latitude temperature changes, shifting the location of the intertropical convergence zone that dominates monsoon precipitation. Our TraCE-21000 modeling experiment supports this interpretation.

    Description: Central Asia is currently a semiarid-arid region, dominated by the Westerlies. It is important to understand mechanisms of climate and precipitation changes here, as water availability in the region is crucial today and in the future. High-resolution, absolutely-dated oxygen isotope (δ18O) records of stalagmites from Kesang Cave characterize a dynamic precipitation history over most of the past 500,000 years. This record demonstrates, for the first time, that climate change in the region exhibits a processional rhythm with abrupt inceptions of low δ18O speleothem growth at times of high Northern Hemisphere summer insolation followed by gradual δ18O increases that track decreases of insolation. These observations and interpretations contrast with the interpretation of nearby, but higher elevation ice core records. The absolutely-dated cave δ18O shifts can be used to correlate the regional climate variability by providing chronological marks. Combined with other paleoclimate records, the Kesang observations suggest that possible incursions of Asian summer monsoon rainfall or related moisture into the Kesang site and/or adjacent areas during the high insolation times may play an important role in changing orbital-scale hydrology of the region. Based on our record, arid climate will prevail in this region for the next several millennia, providing that anthropogenic effects do not supersede natural processes.
