Description: Mechanisms underlying experience-dependent refinement of cortical connections, especially GABAergic inhibitory circuits, are unknown. By using a line of mutant mice that lack activity-dependent BDNF expression (bdnf-KIV), we show that experience regulation of cortical GABAergic network is mediated by activity-driven BDNF expression. Levels of endogenous BDNF protein in the barrel cortex are strongly regulated by sensory inputs from whiskers. There is a severe alteration of excitation and inhibition balance in the barrel cortex of bdnf-KIV mice as a result of reduced inhibitory but not excitatory conductance. Within the inhibitory circuits, the mutant barrel cortex exhibits significantly reduced levels of GABA release only from the parvalbumin-expressing fast-spiking (FS) interneurons, but not other interneuron subtypes. Postnatal deprivation of sensory inputs markedly decreased perisomatic inhibition selectively from FS cells in wild-type but not bdnf-KIV mice. These results suggest that postnatal experience, through activity-driven BDNF expression, controls cortical development by regulating FS cell-mediated perisomatic inhibition in vivo.

Description: We report, in this paper, several findings about the swimming and attachment mechanisms of Giardia lamblia trophozoites. These data were collected using a combination of a high-contrast CytoViva imaging system and a particle image velocimetry camera, which can capture images at speeds greater than 800 frames/s. Using this system, we discovered that, during rapid swimming of Giardia trophozoites, undulations of the caudal region contributed to forward propulsion combined with the beating of the flagella pairs. It was also discovered, in contrast to previous studies with 10 times slower image sampling technique, that the anterior and posterolateral flagella beat with a clearly defined power stroke and not symmetrical undulations. During the transition from free swimming to attachment, trophozoites modified their swimming behavior from a rapid rotating motion to a more stable planar swimming. While using this planar swimming motion, the trophozoites used the flagella for propulsion and directional control. In addition to examination of the posterolateral and anterior flagella, a model to describe the motion of the ventral flagella was derived, indicating that the ventral flagella beat in an expanding sine wave. In addition, the structure of the ventrocaudal groove creates boundary conditions that determine the form of beating of the ventral flagella. The results from this study indicate that Giardia is able to simultaneously generate both ciliary beating and typical eukaryotic flagellar beating using different pairs of flagella.

Description: On the morning of 14 April 2010, the M s  7.1 Yushu earthquake struck the eastern Tibet Plateau and an M s  6.3 aftershock followed west-southwest of the mainshock epicenter one and a half hours later. The Yushu earthquake occurred on the northwestern continuation of the Ganzi–Xianshuihe fault zone and reactivated two segments of the fault. Ruptures associated with Yushu earthquake span a zone 70 km long, extending from south of Jielong in the west to south of Gyêgu (Yushu) in the east. Detailed mapping of the surface rupture zone shows that it consists of two strike-slip segments, the western and eastern segments, separated by the Longbao Lake step-over (pull apart) basin. Our analysis suggests that the 18-km-long western segment of the surface rupture south of the Longbao Lake basin may have been associated with the M s  6.3 aftershock. The eastern rupture segment, which is inferred to be associated with the mainshock, is 32 km long. The main surface rupture can also be divided into two sections, with a 2-km-long gap between them. The surface rupture exhibits left-lateral slip and is localized in a zone about 50 m wide along the present-day trace of the Yushu fault. A single break, without splays or branching traces, characterizes most of the surface rupture zone. The observed magnitudes of surface displacement are typically 0.5–1 m with a maximum slip of 2 m. Both field observations and seismic inversion results suggest, rupture of M s  7.1 Yushu earthquake nucleated near the Longbao Lake step-over basin in the west, propagated unilaterally eastward, and terminated within the east segment margin of the Yushu fault.

Description: Central Asia is currently a semiarid-arid region, dominated by the Westerlies. It is important to understand mechanisms of climate and precipitation changes here, as water availability in the region is crucial today and in the future. High-resolution, absolutely-dated oxygen isotope (δ18O) records of stalagmites from Kesang Cave characterize a dynamic precipitation history over most of the past 500,000 years. This record demonstrates, for the first time, that climate change in the region exhibits a processional rhythm with abrupt inceptions of low δ18O speleothem growth at times of high Northern Hemisphere summer insolation followed by gradual δ18O increases that track decreases of insolation. These observations and interpretations contrast with the interpretation of nearby, but higher elevation ice core records. The absolutely-dated cave δ18O shifts can be used to correlate the regional climate variability by providing chronological marks. Combined with other paleoclimate records, the Kesang observations suggest that possible incursions of Asian summer monsoon rainfall or related moisture into the Kesang site and/or adjacent areas during the high insolation times may play an important role in changing orbital-scale hydrology of the region. Based on our record, arid climate will prevail in this region for the next several millennia, providing that anthropogenic effects do not supersede natural processes.

Description: A continuous countercurrent supercritical CO 2 extraction-distillation dual-column process was developed to extract and concentrate natural vitamin E (VE) from soybean oil deodorizer distillate (SODD). The experimental results demonstrated that process parameters such as extraction pressure, temperature, and solvent-to-feed ratio significantly impacted on the extraction efficiency of natural VE. A new five-parameter mass transfer model for the continuous countercurrent supercritical CO 2 extraction-distillation dual-column process was presented based on the Penetration and Double-Film theories. The calculated values of the mathematical model agreed well with the experimental data, with absolute average relative deviation values of less than 25 %. Description: Persistent organic pollutants (POPs) have been associated with a wide range of adverse health effects. Our case–control study was performed to explore the association between placental levels of selected POPs and risks for neural tube defects (NTDs) in a Chinese population with a high prevalence of NTDs. Cases included 80 fetuses or newborns with NTDs, whereas the controls were 50 healthy, nonmalformed newborn infants. Placental concentrations of polycyclic aromatic hydrocarbons (PAHs), organochlorine pesticides, polychlorinated biphenyls, and polybrominated diphenyl ethers were analyzed by gas chromatography–mass spectrometry. The medians of PAHs, o,p′-isomers of dichlorodiphenyltrichloroethane (DDT) and metabolites, α- and γ-hexachlorocyclohexane (HCH), and α-endosulfan were significantly higher in case placentas than in controls. PAH concentrations above the median were associated with a 4.52-fold [95% confidence interval (CI), 2.10–9.74) increased risk for any NTDs, and 5.84- (95% CI, 2.28–14.96) and 3.71-fold (95% CI, 1.57–8.79) increased risks for anencephaly and spina bifida, respectively. A dose–response relationship was observed between PAH levels and the risk of NTDs, with odds ratios for the second, third, and fourth quartiles, compared with the first, of 1.77- (95% CI, 0.66–4.76), 3.83- (95% CI, 1.37–10.75), and 11.67-fold (95% CI, 3.28–41.49), respectively. A dose–response relationship was observed for anencephaly and spina bifida subtypes. Similar results were observed for o,p′-DDT and metabolites, α-HCH, γ-HCH, and α-endosulfan, whereas no dose–response relationship was observed for the last two pollutants. Elevated placental concentrations of PAHs, o,p′-DDT and metabolites, and α-HCH were associated with increased risks of NTDs in this population.

Description: Soot oxidation with NO (in the absence of gas phase O 2 ) on potassium-supported Mg-Al hydrotalcite mixed oxides (K/MgAlO) was studied using a temperature-programmed reaction and in situ FTIR techniques. Nitrite and the ketene group were identified as the reaction intermediates and thus a nitrite-ketene mechanism was proposed in which surface active oxygen on K sites of K/MgAlO is transferred to soot by NO through nitrites. In the absence of gas phase O 2 , soot oxidation with NO at lower temperatures (below 450 °C) is limited by the amount of active oxygen on the K sites. This kind of active oxygen is not reusable but can be replenished in the presence of gas phase O 2 . Soot oxidation with NO on K/MgAIO was studied by temperature-programmed reaction and in situ FTIR. The nitrite and the ketene group were identified as the reaction intermediates and thus a nitrite–ketene mechanism was proposed, in which surface active oxygen on K sites is transferred to soot by NO through nitrites, forming the ketene group.
