    Beschreibung: To determine whether the rat hindlimb elevation model can be used to study the effects of spaceflight and loss of gravitational loading on bone in the adult animal, and to examine the effects of age on bone responsiveness to mechanical loading, we studied 6-mo-old rats subjected to hindlimb elevation for up to 5 wk. Loss of weight bearing in the adult induced a mild hypercalcemia, diminished serum 1,25-dihydroxyvitamin D, decreased vertebral bone mass, and blunted the otherwise normal increase in femoral mass associated with bone maturation. Unloading decreased osteoblast numbers and reduced periosteal and cancellous bone formation but had no effect on bone resorption. Mineralizing surface, mineral apposition rate, and bone formation rate decreased during unloading. Our results demonstrate the utility of the adult rat hindlimb elevation model as a means of simulating the loss of gravitational loading on the skeleton, and they show that the effects of nonweight bearing are prolonged and have a greater relative effect on bone formation in the adult than in the young growing animal.

    Beschreibung: Human DNA polymerase and DNA ligase utilization for the repair of a major class of ionizing radiation-induced DNA lesion [DNA single-strand breaks containing 3'-phosphoglycolate (3'-PG)] was examined using a novel, chemically defined vector substrate containing a single, site-specific 3'-PG single-strand break lesion. In addition, the major human AP endonuclease, HAP1 (also known as APE1, APEX, Ref-1), was tested to determine if it was involved in initiating repair of 3'-PG-containing single-strand break lesions. DNA polymerase beta was found to be the primary polymerase responsible for nucleotide incorporation at the lesion site following excision of the 3'-PG blocking group. However, DNA polymerase delta/straightepsilon was also capable of nucleotide incorporation at the lesion site following 3'-PG excision. In addition, repair reactions catalyzed by DNA polymerase beta were found to be most effective in the presence of DNA ligase III, while those catalyzed by DNA polymerase delta/straightepsilon appeared to be more effective in the presence of DNA ligase I. Also, it was demonstrated that the repair initiating 3'-PG excision reaction was not dependent upon HAP1 activity, as judged by inhibition of HAP1 with neutralizing HAP1-specific polyclonal antibody.

    Beschreibung: Our objective is to propose a landing site that the Mars Surveyor 2001 Lander and Curie Rover could go to on Mars that should meet the safety requirements of the spacecraft landing system and optimize surface operations (chiefly driven by power and communications requirements). This site lies between 1.5-3.5 deg S latitude, 195-198 deg W longitude, along a sharp albedo contact between the low-viscosity flow units of southern Elysium Planitia and the eroded highlands margin east of Aeolis Mensae. A relatively-bright "peninsula-like" protrusion of the eroded highlands into the south Elysium plains in this area reminds us of the head of an Ibis, and so we nickname this site "Ibishead Peninsula". This site is designed to be situated as close to a diversity of geologic units within view of the lander instruments. Based on our experience with the visibility of horizon details from the Mars Pathfinder and Viking landing sites, we stipulate that for horizon features to be resolved suitably for detailed study from the lander, they must be no more than several kilometers distant. This is so that diversity can be placed in a geologic context in a region that we feel has some exciting science potential. This objective is different from the Mars Pathfinder requirement to land at a site with a maximum chance for containing a diversity of rocks within a few tens of meters of the lander, which resulted in the selection of a "grab bag" site.

    Beschreibung: One of the original objectives of the Mars Orbiter Camera (MOC), as proposed in 1985, was to acquire observations to be used in assessing future spacecraft landing sites. Images obtained by the Mars Global Surveyor MOC since March 1999 provide the highest resolution views (1.5-4.5 m/pixel) of the planet ever seen. We have been examining these new data to develop a general view of what Mars is like at meter-scale within the latitudes and elevations that are accessible to the Mars Surveyor 2001 lander. Our goal is to provide guidance to the 2001 landing site selection process, rather than to use MOC images to recommend a specific landing site.

    Beschreibung: The p53 tumor suppressor gene has been shown to be involved in a variety of repair processes, and recent findings have suggested that p53 may be involved in DNA double strand break repair in irradiated cells. The role of p53 in DNA double strand break repair, however, has not been fully investigated. In this study, we have constructed a novel Epstein-Barr virus (EBV)-based shuttle vector, designated as pZEBNA, to explore the influence of p53 on DNA strand break repair in human lymphoblasts, since EBV-based vectors do not inactivate the p53 pathway. We have compared plasmid survival of irradiated, restriction enzyme linearized, and calf intestinal alkaline phosphatase (CIP)-treated pZEBNA with a Simian virus 40 (SV40)-based shuttle vector, pZ189, in TK6 (wild-type p53) and WTK1 (mutant p53) lymphoblasts and determined that p53 does not modulate DNA double strand break repair in these cell lines.     Beschreibung: Gamma-ray bursts (GRBs) are thought to arise when an extremely relativistic outflow of particles from a massive explosion (the nature at which is still unclear) interacts with material surrounding the site of the explosion. Observations of the evolving changes in emission at many wavelengths allow us to investigate the origin of the photons, and so potentially determine the nature of the explosion. Here we report the results of gamma-ray, optical, infrared, submillimeter, millimeter and radio observations of the burst ORB990123 and its afterglow. Our interpretation of the data indicates that the initial and afterglow emissions are associated with three distinct regions in the fireball. The peak flux of the afterglow, one day after the burst, has a lower frequency than observed for other bursts; this explains the short-lived radio emission. We suggest that the differences between bursts reflect variations in the magnetic-field strength in the afterglow-emitting regions.

    Beschreibung: We report the results from an ASCA observation of the high-luminosity, radioloud quasar PKS 2149-306 (redshift 2.345), covering the approximately 1.7 - 30 keV band in the quasar-frame. We find the source to have a luminosity of approximately 6 x 10(exp 47) ergs/s in the 2 - 10 keV band (quasar frame). We detect an emission line centered at approximately 17 keV in the quasar frame. Line emission at this energy has not been observed in any other active galaxy or quasar to date. We present evidence rejecting the possibility that this line is the result of instrumental artifacts, or a serendipitous source. The most likely explanation is blueshifted Fe-K emission (the equivalent width, is EW approximately 300 +/- 200 eV, quasar frame). Bulk velocities of the order of 0.75c are implied by the data. We show that Fe-K line photons originating in an accretion disk and Compton-scattering off a leptonic jet aligned along the disk axis can account for the emission line. Curiously. if the emission-line feature recently discovered in another quasar (PKS 0637-752, z = 0.654) at 1.6 keV in the quasar frame, is due to blueshifted OVII emission, the Doppler blueshifting factor in both quasars is similar (approximately 2.7 - 2.8).

    Beschreibung: The discovery of afterglows associated with gamma-ray bursts at X-ray, optical and radio wavelengths and the measurement of the redshifts of some of these events has established that gamma-ray bursts lie at extreme distances, making them the most powerful photon-emitters known in the Universe. Here we report the discovery of transient optical emission in the error box of the gamma-ray burst GRB980425, the light curve of which was very different from that of previous optical afterglows associated with gamma-ray bursts. The optical transient is located in a spiral arm of the galaxy ESO 184-GS2, which has a redshift velocity of only 2,550 km/ s. Its optical spectrum and location indicate that it is a very luminous supernova, which has been identified as SN1998bw. If this supernova and GRB980425 are indeed associated, the energy radiated in gamma-rays is at least four orders of magnitude less than in other gamma-ray bursts, although its appearance was otherwise unremarkable: this indicates that very different mechanisms can give rise to gamma-ray bursts. But independent of this association, the supernova is itself unusual, exhibiting an unusual light curve at radio wavelengths that requires that the gas emitting the radio photons be expanding relativistically.
