Notes: Abstract Neutral complex-type N-glycans of the envelope glycoprotein 120 of HIV-2, propagated in different host cells, display cell-type specific variations. In order to identify typical structural elements, glycans were analysed by gel filtration, by enzymic sequencing and, in part, by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry. The characteristic substituents of di- tri- and tetraantennary carbohydrate units thus observed include N-acetyllactosamine repeats, bisecting N-acetylglucosamine and fucose linked to the chitobiose core as well as to N-acetyllactosamine antennae. Each glycoprotein preparation displayed a characteristic set of glycoforms. Abbreviations: endo H, endo-β-N-acetylglucosaminidase H; E-PHA, Phaseolus vulgaris agglutinin E4; GlcNAcOH, N-acetyl-glucosaminitol; gp120/HUT78(MOLT4/Mφ/PBL/U937), external envelope glycoprotein 120 of HIV-2, strain D194, propagated in HUT78 (MOLT4, Mφ, PBL, U937) cells; gu, glucose units; HPAEC, high-pH anion-exchange chromatography; MALDI-TOF-MS, matrix-assisted laser desorption/ionization time-of-flight mass spectrometry; Mφ, human monocytes/macrophages; PBL, human peripheral blood lymphocytes; PNGase F, peptide-N4-(N-acetyl-β-glucosaminyl)asparagine amidase F

Notes: [Auszug] The functional properties of inactivation domains of Kv channels have been analysed in studies combining electrophysiological measurements with site-direct mutagenesis. Both electrostatic and hydrophobic interactions were found to determine the affinity of the ball domain for its binding site in ...

Notes: Different growth stages and the microstructure of amorphous CuTi films are investigated by scanning tunneling microscopy and small angle neutron scattering. During film growth at room temperature, the initially smooth films show increasing surface roughening and finally a change to a columnar growth mode with column diameters of about 20 nm. The interfacial energies associated with the column boundaries are higher than those of grain boundaries in crystalline systems. The column boundaries might be the origin of high intrinsic tensile stresses measured before in the amorphous CuTi films. © 1997 American Institute of Physics.

Notes: A new small-angle X-ray scattering instrument has been set up on the D2AM-ESRF beamline. This modular instrument, which has the capacity to handle different sample environments (variable temperature and pressure), is designed for easy wavelength changes, i.e. for anomalous-scattering studies. This paper describes its design and discusses in some detail its positioning and alignment with respect to the focusing optics. The mirrors were found to be a major source of diffuse scattering, which was eliminated by rearrangement of the slit positions and refocusing of the optics. The present configuration yields a high flux (1010photons s−1) and allows measurements down to q = 0.005 Å−1.

Notes: [Auszug] Ras-related GTP-binding proteins such as Ras, Rho, Rac/Cdc42 are signal switch molecules that cycle between the GDP-bound inactive and the GTP-bound active form. They are activated by the action of guanine-nucleotide exchange factors (GEF) and downregulated by the GTPase reaction, which is ...

Notes: Abstract In continuing our studies of temporal slicings of spacetimes, we consider the collapse from rest of an initially homogeneous dust sphere, known as Oppenheimer-Snyder collapse. It is shown that in the case of harmonic slicing the whole spacetime becomes covered by slices extending to spatial infinity in contrast to maximal slicing. The behavior of the lapse function is discussed, especially for various data on the initial slice, and it is demonstrated that not every choice of the initial lapse leads to meaningful slicings.

Notes: Abstract Traditional hay-meadows in the Alps and fens at the edge of the Alps are habitats for many rare and endangered butterfly species. Conservation efforts aim at preserving these species, but the biotopes depend on regular mowing, which in turn requires intensive financial support. The feasibility of substituting mowing of these sites by grazing is discussed and considered as a more cost effective management type which produces agriculturally valuable goods as well. In this study the butterfly fauna of mown and grazed sites were compared. Species composition, species number, and the occurrence of rare species under the two management types were in most cases rather similar for both grassland ecosystems. Nevertheless, there are hints that for single rare species this might not be true. Additionally, at one site, grazing intensity on a former hay-meadow was too high to preserve the species-rich community. Overall the results are encouraging: grazing does not have to be as detrimental as formerly thought, although details (compartments of pastures, intensity) still have to be confirmed. Experimental grazing management of aban-doned grasslands of the studied types should be started.

Notes: Abstract The observation of the production of antihydrogen atoms \overlineH 0\equiv\barpe+, the simplest atomic bound state of antimatter, is presented. A method has been used by the PS210 collaboration at LEAR which assumes that the production of \overlineH0 is predominantly mediated by the e+e--pair creation via the two-photon mechanism in the antiproton--nucleus interaction. Neutral \overlineH0 atoms are indentified by a unique sequence of characteristics. In principle \overlineH0 is well suited for investigations of fundamental CPT violation studies under different forces, however, in our investigations we concentrate on the production of this antimatter object, since so far it had not been observed. The production of eleven antihydrogen atoms is reported including possibly 2± 1 background signals, the observed yield agrees with theoretical predictions.

Notes: Abstract The neutral glycosphingolipid fraction from adults of the pig parasitic nematode, Ascaris suum, was resolved into four components on thin-layer chromatography. The high-performance liquid chromatography-isolated components were structurally analysed by: methylation analysis; exoglycosidase cleavage; gas-liquid chromatography/mass spectrometry; liquid secondary-ion mass spectrometry; and, in particular, matrix-assisted laser desorption/ionization time-of-flight mass spectrometry. Their chemical structures were determined as: Glc(β1-1)ceramide, Man(β1-4)Glc(β1-1)ceramide, GlcNAc(β1-3)Man(β1-4)Glc(β1-1)ceramide and Gal(α1-3)GalNAc(β1-4)GlcNAc(β1-3)Man(β1-4)Glc(β1-1)ceramide; and were characterized as belonging to the arthro-series of protostomial glycosphingolipids. No glycosphingolipid component corresponding to ceramide tetrasaccharide was detected during these analyses. The ceramide composition of the parent glycosphingolipids was dominated by the 2-(R)-hydroxy C24:0 fatty acid, cerebronic acid, and C17 sphingoid-bases: 15-methylhexadecasphing-4-enine and 15-methylhexadecaphinganine in approximately equal proportions. The component ceramide monohexoside was characterized by an additional 15-methylhexadecaphytosphingosine. Abbreviations: CDH, ceramide dihexoside; Cer, ceramide; CMH, ceramide monohexoside; CPH, ceramide pentahexoside; CTH, ceramide trihexoside; CTetH, ceramide tetrahexoside; Hex, hexose; HexNAc, N-acetylhexosamine; HPTLC, high-performance thin-layer chromatography; LSIMS, liquid secondary-ion mass spectrometry; MALDI-TOF-MS, matrix-assisted laser desorption/ionization time-of-flight mass spectrometry; N-, Nz- and A-glyco(sphingo)lipids, neutral, neutralzwitterionic and acidic glyco(sphingo)lipids, respectively
