    Notes: Abstract Phylogenetic relationships among the seven species of deep-sea giant clams Calyptogena (Bivalvia: Vesicomyidae) collected around Japan were examined using parts of nucleotide sequences of mitochondrial genes for cytochrome oxidase I (COI) and cytochrome oxidase III (COIII) and the encoded amino acid sequences. The seven species were C. soyoae (Sagami Bay), C. fausta (Suruga Bay), C. kaikoi (Nankai Trough), C. nautilei (Nankai Trough), C. phaseoliformis (Japan Trench), C. solidissima (Minami-Ensei Knoll, Okinawa Trough) and Calyptogena sp. (Iheya Ridge, Okinawa Trough). A clear phylogenetic split was observed between one group of three species (C. kaikoi, C. phaseoliformis and C. fausta) and the remaining species. This clustering corresponds to the two previously described subgenera within the genus Calyptogena (Calyptogean and Ectenagena) with the exception of the placement of C. nautilei, which had been placed in the subgenus Ectenagena. Genetic distances between two haplotypes of C. soyoae were 0.043 for the COI region and 0.055 for the COIII region, and three amino acid substitutions were detected with the COIII region. Calyptogena sp. from the Iheya Ridge could be distinguished from one of the two haplotypes (type A) of C. soyoae by only a single nucleotide substitution, a result that suggests that Calyptogena sp. of the Iheya Ridge diverged from C. soyoae after the two haplotypes had diverged, and it is now isolated from C. soyoae in Sagami Bay.

    Notes: An alkaline serine protease, M-protease, from Bacillus sp. KSM-K16 has been crystallized. Two morphologically different crystal forms were obtained. Crystal data of form 1: space group P212121, a = 47.3, b = 62.5, c = 75.6 Å, V = 2.23 × 105 Å3, Z = 4 and Vm = 2.09 Å3 Da−1. Crystal data of form 2: space group P212121, a = 75.82 (2), b = 57.79 (2), c = 54.19 (1) Å, V = 2.29 (2) × 105 Å3, Z = 4 and Vm = 2.15 Å3 Da−1. The crystal structure of M-protease in form 2 has been solved by molecular replacement using the atomic model of subtilisin Carlsberg (SBC) which is 60% homologous with M-protease, and refined to the crystallographic R-factor of 0.189 for 7004 reflections with Fo/σ(F) 〉 3 between 7 and 2.4 Å resolution. The final model of M-protease contains 1882 protein atoms, two calcium ions and 44 water molecules. The three-dimensional structure of M-protease is essentially similar to other subtilisins of known structure. The 269 Cα positions of M-protease have an r.m.s. difference of 1.06 Å with the corresponding positions of SBC. The crystal data of form 2 are close to those of SBC, though the structure determination of form 2 made it clear that it is not isomorphous to the crystal structure of SBC. The deletions of amino acids occur at the residues 36′ and 160′–163′ compared with SBC (numerals with primes show the numbering for SBC). The deletion of the four residues (160′–163′) may significantly affect the lack of isomorphism between M-protease and SBC.

    Notes: Two sympatric morphs (type A with a vertebral number of 25 and type B with a vertebral number of 24) of striped jack Pseudocaranx dentex (Bloch & Schneider) were analysed genetically. A part of the 16S–rRNA region of mtDNA was amplified with polymerase chain reaction for 24 specimens, and a restriction enzyme fragment polymorphism showed significant differences between the two types. While all specimens sampled in Ogasawara were identified as type B, about 90% of striped jack in Oita were type A and 10% were type B. Although the spawning areas of these two types are still unknown, significant genetic differences between the two sympatric morphs show that recruitment and migration patterns might differ from each other. The current system suggests the possibility that the juveniles of type B in Oita may migrate from the Ogasawara Islands.

    Notes: Adhesion between thin Te-based alloy films and fluorocarbon polymer sublayers, prepared by sputtering or plasma polymerization, was investigated by observing the 1 μm-sized ablative hole opening process with a focused laser beam. Interpretations of the mechanisms for the change in energy required for the hole opening and pit geometry were based on the framework of studies of the ablative hole opening process for optical recording. Observations suggest that the molten material flow during the hole opening includes a ductile fracture and a viscous flow of the molten sublayer material as well as of active layer material. Adhesion acts as an energy barrier against the above mentioned flow of molten material during the hole opening process. Since the fluorocarbon films used in the present work had highly cross-linked structures, the adhesion was mainly dominated by the dynamic force of adhesion. Therefore, the hole opening process was mainly affected by the dynamic force of adhesion rather than the static force, which is dominated by the surface energy of the sublayer. There was a good correlation between the dynamic force of adhesion estimated by the peel-off strength and the concentrations of the -CF- and -C-CF- structures estimated from C1s spectra obtained by x-ray photoelectron spectroscopy. © 1995 American Institute of Physics.

    Notes: The dependence on the source location of photothermal bistabilities of a thin CdS:Cu film is demonstrated by the investigation of bistability in luminescence in reflection geometry. Particularly, it is shown that the infrared portion in the reflected beam clearly exhibits contrasting bistable loops in contrast to the reflection itself. The unexpected effect is explained by the fact that bistability in luminescence is a bulk feature which is not affected by the surface region. © 1995 American Institute of Physics.

    Notes: Abstract— Usually, scatter in fracture toughness values is studied by assuming the employed steel plate to be macroscopically homogeneous in toughness, but it is probable that inhomogeneity contributes to scatter. This paper proposes a method which can discriminate scatter due to inhomogeneity from scatter measured in tests by using small size test pieces, cut out of previously fractured test specimens used for fracture toughness tests. By this method, the observed scatter of Kc(J) in a previous paper by some of the present authors was shown to be affected considerably by inhomogeneity, although the observed scatter was fairly well described by a Weibull distribution analysis.

    Notes: Abstract Alkaline protease (EC 3.4.21.14) activity, suitable for use in detergents, was detected in the alkaline culture medium of Bacillus sp. KSM-K16, which was originally isolated from soil. The enzyme, designated M protease, was purified to homogeneity from the culture broth by column chromatographies. The N-terminal amino acid sequence was Ala-Gln-Ser-Val-Pro-Trp-Gly-Ile-Ser-Arg-Val-Gln-Ala-Pro-Ala-Ala-His-Asn-Arg-Gly-Leu-Thr-Gly. The molecular mass of the protease was 28 kDa, and its isoelectric point was close to pH 10.6. Maximum activity toward casein was observed at 55°C and at pH 12.3 in 50 mM phosphate/NaOH buffer. The activity was inhibited by phenylmethylsulfonyl flouride and chymostatin. The enzyme was very stable in long-term incubation with liquid detergents at 40°C. The enzyme cleaved the oxidized insulin B chain initially at Leu15-Tyr16 and efficiently at ten more sites. Among various oligopeptidyl p-nitro-anilides (pNA) tested, N-succinyl-Ala-Ala-Pro-Phe-pNA was efficiently hydrolyzed by M protease. M protease was precipitated in (NH4)2SO4-saturated acetate buffer (pH 5.0) as plank-like cyrstals.

    Notes: Abstract The longitudinal momentum distribution of7Be was measured after the break-up reaction of8B in C, Al, and Pb targets at 1471 A·MeV. We observed a narrow distribution with a FWHM of (81±6) MeV/c in all targets. The experimental results indicate an extended spatial distribution of the loosely bound proton in8B, and agree with QRPA calculations.
