    Notes: The temporal cholesterolemic patterns to skim milk powder (SMP) and reprocessed SMP (RSMP) diets were compared to diets containing casein or laboratory rat chow over a 24-day period. SMP was hypocholestetemic relative to casein in rats fed a 1% dietary cholesterol. Reprocessing of SMP resulted in an apparent loss of the relative hypocholesterolemic reponse of native SMP. Amino acid analysis of SMP and RSMP, showed only marginally lower lysine levels than casein; however, the Iysine/arginine ratio was higher in SMP than either in casein or RSMP. Available lysine content in SMP was higher than in RSMP, suggesting occurrence of nonenyzmatic browning reactions. Although a similar cholesterolemic response was observed in casein and RSMP, the available lysine content of these two protein sources were markedly different, suggesting that reduced available lysine alone was not totally responsible for the lower cholesterolemic response of SMP, relative to casein.

    Notes: Abstract A comparative study of the protein and lipopolysaccharide (LPS) profiles of 8 strains of Haemophilus ducreyi revealed that protein patterns remained unaffected by changes in medium composition, atmospheric conditions and temperature. In contrast, the LPS patterns exhibited marked variations under the different cultural conditions.

    Notes: Abstract Lipopolysaccharides (LPS) extracted from 33 clinical isolates of Bacteroides ureolyticus were examined by polyacrylamide gel electrophoresis (PAGE) and silver staining. Variable results were obtained with proteinase K-digested whole-cell lysates, but the LPS was shown conclusively to be of the smooth type, exhibiting O side chains, when phenol-water extracts were used. Heterogeneity among the smooth LPS profiles of the strains of B. ureolyticus suggested that such profiles might be useful for typing unknown clinical strains.

    Notes: The relatively scarcity of flat or moderately sloping land in Central Appalachia make reclaimed surface mined lands attractive for agricultural uses. A reclaimed surface coal mine in southern West Virginia was placed under grazing management during the 1984 and 1985 growing seasons. Discharge was collected from summer-grazed watersheds of about 2.8 ha and 8.9 ha and analyzed, by the membrane-filtration method, for fecal coliforms (FC). Prior to grazing, in 1984, FC counts were 〈 20/100 ml. During the grazing season, FC ranged from 〈0/100 ml to〉 1000/100 ml in 1984 and from 0/100 ml to 〉 2500/100 ml in 1985. FC counts remained high during warm periods for several months after grazing ceased. It was concluded that the bacteriological quality of receiving streams was impacted by grazing the reclaimed area and recommended standards for point sources were often exceeded; however, the FC counts did not appear to be any greater than what would have been expected from grazed, undisturbed areas. Reclaimed surface mine areas in Appalachia have the potential to be a valuable “flat land” resource and grazing appears to be an alternative post mine land use that affects bacteriological water quality in a similar manner as “natural” pastures. However, good management practices may be necessary to avoid bacterial contamination of adjacent bodies of water.

    Notes: A bovine calf liver DNA library in λ2001 bacteriophage has been screened with a human Igγ4 heavy-chain constant-region gene probe. Four hybridizing clones have been identified, and the DNA sequences in two of these, which have high homology with CHγ genes, are reported here. Within the bovine sequences, four separate exons can be identified, corresponding to the three CH domains and the hinge of gamma heavy-chain genes. Both of these genes contain atypical sequences around one or more of their exon/intron boundaries with consequent loss of splice sites, indicating that these are probably γ pseudogenes. One sequence codes for a C-terminal peptide which matches the 18-mer C-terminal heavy-chain peptide of bovine serum IgG2, the other encodes a C-terminal peptide unknown in the bovine. These results suggest that evolutionary duplication of CHγ genes has occurred in the bovine.

    Notes: The ELY-I locus controls the expression of a polymorphic cell surface antigen of equine lymphocytes which was detected using antibodies generated by alloimmunization with peripheral blood lymphocytes. The ELY-1 antigens were not detected on erythrocytes or platelets by absorption experiments. The two alleles, which have been designated ELY-1.1 and ELY-1.2, are expressed codominantly and appear to constitute a closed system at the population level. In family studies, the ELY-1 antigens segregated as products of an autosomal locus not linked to the major histocompatibility complex (MHC) of the horse. In the complement mediated lymphocyte microcytotoxicity test, antisera to the ELY-1 antigens selectively killed peripheral blood lymphocytes which did not express surface immunoglobulin. The ELY-1 antigens may be useful markers for equine T cells when assayed in this fashion. Three alloantisera were used in immune precipitation of iodinated and solubilized cell surface proteins from peripheral blood lymphocytes. Electrophoresis of the precipitates in sodium dodecyl sulphate (SDS)-polyacrylamide gels demonstrated strong bands in the Mr 180–190K range that were shared in the three different preparations. These results suggest that the ELY-1 allospecificities are expressed on an equine equivalent of the murine T200 molecule.

    Notes: Regional procedures to estimate flood magnitudes for ungaged watersheds typically ignore available site-specific historic flood information such as high water marks and the corresponding flow estimates, otherwise referred to as limited site-specific historic (LSSH) flood data. A procedure to construct flood frequency curves on the basis of LSSH flood observations is presented. Simple inverse variance weighting is employed to systematically combine flood estimates obtained from the LSSH data base with those from a regional procedure to obtain improved estimtes of flood peaks on the ungaged watershed. For the region studied, the variance weighted estimates of flow had a lower logarithmic standard error than either the regional or the LSSH flow estimates, when compared to the estimates determined by three standard distributions for gaged watersheds investigated in the development of the methodology. Use of the simple inverse variance weighting procedure is recommended when “reliable” estimates of LSSH floods for the ungaged site are available.

    Notes: The storage stability of eviscerated Indian mackerel (Rastrelliger kanagurta) irradiated at a dose of 1.5 kGy and held under melting ice was examined. Packaging in polyethylene pouches prior to irradiation gave a shelf-life of 25 days, while, nonpackaged, irradiated fish remained in acceptable condition up to 20 days. The nonpackaged irradiated fish has less irradiation odor and better appearance as compared with packaged irradiated fish during the course of ice storage. Uni-radiated and nonpackaged mackerel kept in ice were acceptable only for 14 days. The results suggest that pre-irradiation packaging was not essential for commercially adequate extension in shelf-life of the iced fish.

A study of the diet of E. octoculata (L) and H. stagnalis (L) using a serological technique was carried out in a lot ic habitat subject to organic pollution and seasonal sewage fungus growth.2. The results indicate that E. octoculata consumed predominantly Chironomidae and Oligochaeta, while H. stagttalis had a less pronounced prey preference, consuming rather similar proportions of chironomids, oligochaetes, Asellus and gastropod snails.3. At enriched sites, E. ocioculata consumed relatively more chironomids and fewer oligochaetes, while H. stagnalis took more gastropods and fewer chironomids.4. Seasonal differences were noted in the diet of both leeches with chironomids predominating chiefly from April to August, and molluscs, oligochaetes and Asellus at other times of the year. Intra-specific differences (on a weight-age basis) were only seen for E. octoculata, small forms consuming chiefly oligochaetes and larger ones concentrating on chironomids.

Major landscape features and hydrological parameters indicative of black fly species assemblages were examined at 101 stream sites in Alberta, northern British Columbia, the Yukon and Alaska during the summer. Forty-one black fly taxa were recorded at seventy- nine sites using qualitative sampling procedures. River sites lacking black flies had significantly higher conductivity, greater depth, shallower slope and were farther from the Pacific Ocean than sites with black flies.2. Classification of sites by taxon occurrence using hierarchical cluster analysis suggested five groupings: A. Simulium tuberosum(Lundstrëm) complex + several taxa. B, S. venustum Saylverecundum Stone and Jamnback complexes +S, tuberosum complex; C. 5. arcticum Malloch complex +S. corbis Twinn complex; D, Gymnopais Stone/Prosimulium Roubaud; and E. P. onychodactytum Dyar and Shannon complex + several taxa.3. Multiple discriminant analysis (MDA) was used to predict group membership of the seventy-nine sites using nineteen environmental variables; 71% of the sites were classified correctly. MDA identified latitude and distance from stream source as important factors separating group D from other groups. Stream width and drainage basins entering the Arctic Oeean and Hudson Bay delineated group B. There was no clear separation among groups A. C or E. The presence of sibling species probably accounts for the overlap of black fly assemblages.4. Our findings are briefly discussed in the context of stream classification systems, notably the river continuum concept.
