Notizen: Five monoclonal antibodies reacting with class I MHC antigens were produced by fusing lymphocytes from WF (RT1u) rats immunized against DA (RT1a) rats with P3-X63-Ag8-653 myeloma cells. Sequential immunoprecipitation studies with the mAb and the WF anti-DA alloantiserum demonstrated the presence of four different class I molecules: all four molecules were reactive with the alloantiserum; three of them contained the determinant for mAb 155; two of the latter three molecules shared the determinants for mAb 3, 56 and 60, and one of these two molecules also contained the determinant for mAb 118. The four molecules could be isolated from the antigen preparation by sequential immunodepletion first with 118, next with 3, then with 155 and finally with the alloantiserum or by sequential absorption with affinity columns of Sepharose 4B coupled to the antibodies. The three antigens which were sequentially isolated with the mAb 118, 3, and 155, respectively, were analysed by SDS-PAGE after digestion with Staphylococcus aureus V8 protease, and they showed differences in peptide patterns. The relative amounts of the antigens expressed on red blood cells and on lymphocytes were different based on the results of sequential isolation and indirect cellular radioimmunoassay: the antigen which reacted with both mAb 3 (and 56 or 60) and 155 was the major class I antigen on red blood cells, and the antigen which reacted with mAb 118, 3 (and 56 or 60) and 155 was the major class I antigen on lymphocytes.

Notizen: The growth and reproduction complex contains recessive genes (grc) which influence body weight and gonadal development. Homozygous males are sterile, and they have an arrest of spermatogenesis at the primary spermatocyte stage. Homozygous females are fertile but have a reduced reproductive capacity. The data presented in this paper show that the latter defect is associated with a decrease in the relative number of secondary ovarian follicles and an increase in the number of atretic follicles. This finding indicates that most of the primary follicles do not mature properly. Thus, the genetic defect in gametogenesis controlled by the grc appears to occur at the same stage of development in both females and males.

Notizen: The combination of zinc (II) with phytic acid has been investigated under a variety of reaction conditions. With the P:Zn ratios used, i.e. 1:1 or 6:1, in the preparations elemental analyses of the isolated products indicated P:Zn ratios of either 1, 1.2 or 1.5 dependent upon the initial P:Zn ratios and the various bases and salts used in the preparations. Of the total number of experiments carried out elemental analyses indicated that only about 25% of the isolated complexes had stoichiometric atomic ratios.

Notizen: Two red clover (Trifolium pratense) cultivars, Red Head (tetraploid) and Kuhn (diploid), were sown at a seed rate of 13 kg ha−1 either alone or in mixture with Italian ryegrass (Lolium multiflorum) cv. RvP sown at seed rates of 10, 15, 20 or 30 kg ha−1. RvP was also sown alone at a seed rate of 30 kg ha−1 and received nil or 300 kg ha−1 fertilizer a−1 fertilizer N. All plots were established using the barley cultivar Midas sown at a seed rate of 100 kg ha−1 as a nurse crop.Neither clover cultivar nor ryegrass seed rate significantly influenced either dry matter harvested or botanical composition over the 3 harvest years. On average over all years the grass-clover mixtures produced 75% of the yield of the N-fertilized RvP, 125% of the clover monocultures and 225% of the unfertilized RvP. The red clover contribution to the total dry matter harvested of the mixtures averaged 45–60%. The dry matter concentrations of the mixtures were considerably higher than those of the pure clover stands. In the third year yields were markedly reduced in comparison with those in the first and second years. It was concluded that Italian ryegrass can be a suitable companion grass for red clover. Its superior yielding capacity over other grasses such as perennial ryegrass or timothy under a conservation management can be coupled to advantage with red clover to give a sward which Is essentially stable, at least over a 2- to 3-year cropping period, although giving slightly reduced yields in the third year. Italian ryegrass-red clover mixtures, without the use of fertilizer N, can produce high DM yields of good quality herbage.

Notizen: The length–weight relationship for Lough Neagh pollan was log W= 3·2 log L–2·24. Covariance analysis indicated that there were no differences between the sexes, with maturity, or between years. While a seasonal cycle in relative condition was in phase with gonad development during late summer and autumn, changes in somatic condition presented a more complex pattern. Somatic condition increased in spring, as did the quantity and diversity of food consumed. Despite good conditions for feeding and growth, there was an autumn fall in somatic condition, caused by the diversion of energy to gonad growth.

Notizen: The highly active, polar gibberellin-like substance found in the apical region of shoots of tall (genotype Le) peas (Pisum sativum L.) is shown by combined gas chromatography-mass spectrometry (GC/MS) to be GA1. This substance is either absent or present at only low levels in dwarf (le) plants. Multiple ion monitoring (MIM) tentatively suggests that GA8 may also be present in shoot tissue of tall peas. Gibberellin A1 is the first 3 β-hydroxylated gibberellin positively identified in peas, and its presence in shoot tissue demonstrates the organ specificity of gibberellin production since GA1 has not been detected in developing seeds. Application of GA1 can mask the Le/le gene difference. However, whilst Le plants respond equally to GA20 and GA1, le plants respond only weakly to GA20, the major biologically active gibberellin found in dwarf peas. These results suggest that the Le gene controls the production of a 3 β-hydroxylase capable of converting GA20 to GA1. Further support for this view comes from feeds of [3H] GA20 to Le and le plants. Plants with Le metabolise [3H] GA20 to three major products whilst le plants produce only one major product after the same time. The metabolite common to Le and le plants co-chromatographs with GA29. The additional two metabolites in Le peas co-chromatograph with GA1 and GA8.

Notizen: Visual damage is commonly used as the criterion for assessment of salinity resistance in rice breeding programmes. The use of other indicators, such as sodium uptake, is being evaluated: a correlation between initial sodium uptake to the third leaf and varietal survival was established and the physiological basis of this examined.Chlorophyll was used as an indicator of metabolic status and the relationship between leaf sodium and chlorophyll concentrations investigated for nine varieties differing in their resistance to salinity. By sampling a population of leaves having a wide range of salt concentrations, inverse relationships between chlorophyll and sodium concentrations were established. The salt concentration in the leaf tissue which causes equivalent toxicity (50% loss of chlorophyll) differed 3-fold amongst these varieties. Varieties showing the greatest tolerance to salt within the leaves were not necessarily those showing the greatest overall phenotypic resistance to salinity. The enzymes malate dehydrogenase and nitrate reductase were activated equally by sodium and potassium ions at 60–80 mol m−3 in both control and saline grown plants and severely inhibited at higher concentrations. If all the salt in the tissue of leaves with 50% chlorophyll remaining was in solution and uniformly distributed the concentration would be 135–500 mol m−3. This is improbable and some level of compartmentation is likely. It is concluded that while initial sodium uptake has predictive value for salinity resistance of varieties, there are other characteristics which are masked by excess salt entry and require independent assessment; no single factor confers resistance.
