    Description: The glucagon analog [l-N alpha-trinitrophenylhistidine, 12-homoarginine]-glucagon (THG) was examined for its ability to lower blood glucose concentrations in rats made diabetic with streptozotocin. In vitro, THG is a potent antagonist of glucagon activation of the hepatic adenylate cyclase assay system. Intravenous bolus injections of THG caused rapid decreases (20 to 35 percent) of short duration in blood glucose. Continuous infusion of low concentrations of the inhibitor led to larger sustained decreases in blood glucose (30 to 65 percent).     Description: A ground based search for the 856.50137/cm R(9.5) and for the 859.76765 R(12.5) transitions of stratospheric (Cl-35)O was made in the solar absorption mode using an infrared heterodyne spectrometer. Lines due to stratospheric HNO3 and tropospheric OCS were detected, at about 0.3% absorption levels. The expected lines of ClO in this same region were not detected, even though the optical depth of the ClO lines should be on the order of 0.2% using currently accepted ClO abundances. These infrared measurements suggest that stratospheric ClO is at least a factor of 7 less abundant than is indicated by indirect in situ fluorescence measurements, and the upper limit of 2.4x10 to the 13th power molecules/sq cm to the integrated column density of ClO is a factor of over 4 less than is indicted by microwave measurements. Results imply that the release of fluorocarbon precursors of ClO may be significantly less important for the destruction of stratospheric ozone than was previously thought.

    Description: A computer automated data acquisition system for atmospheric emittance, and global solar, downwelled diffuse solar, and direct solar irradiances is discussed. Hourly-integrated global solar and atmospheric emitted radiances were measured continuously from February 1981 and hourly-integrated diffuse solar and direct solar irradiances were measured continuously from October 1981. One-minute integrated data are available for each of these components from February 1982. The results of the correlation of global insolation with fractional cloud cover for the first year's data set. A February data set, composed of one-minute integrated global insolation and direct solar irradiance, cloud cover fractions, meteorological data from nearby weather stations, and GOES East satellite radiometric data, was collected to test the theoretical model of satellite radiometric data correlation and develop the cloud dependence for the local measurement site.

    Description: Contours and gridded values are given for seven years of monthly mean total ozone data derived from observations with the Backscattered Ultraviolet instrument on Nimbus-4 for the Northern and Southern Hemispheres. The instrument, algorithm, uncertainties in derived ozone and systematic changes in the bias with respect to the international groundbased ozone network of Dobson instruments, are discussed.

    Notes: A method for the quantitative determination of 1,2,3-propanetrioltrinitrate (nitroglycerin) in human plasma by gas chromatography mass spectrometry has been developed. After addition of 1,2,3-propanetriol(15N)trinitrate as internal standard, plasma (1 ml) is extracted by a mixture of pentane and methyl acetate (90:10). The extracts are purified by partition between, first, the extraction solvent and a mixture of acetonitrile and water (60:40) and, secondly, the resultant aqueous phase and benzene. The solvent is evaporated to a small volume before injection. The fragment ions at m/z 46 and 47 are monitored for the measurement of the [NO2]+ and [15NO2]+ ions using electron impact ionization. The mean recovery (%) ±SD in blind plasma samples spiked with amounts in the concentration range 0.35-3.52 nmol I-1, was 97.4 ± 9.0 (n = 58). Within a day, recovery experiments gave rise to coefficients of variation of 9.6, 6.4 and 2.7% at the levels 0.44,0.88 and 11 nmol I-1, respectively. Concentrations in plasma down to about 0.2 nmol I-1 (50 pg mI-1) could be estimated. Percent recovery of duplicate determinations in the range 0.5-1.96 nmol I-1± SD was 99.4 ± 6.0 (n = 80).

    Notes: The vicinal proton coupling constants were obtained for some 6-membered ring unsaturated heterocyclic compounds. The R values and ring dihedral angles were determined and found to be consistent with half-chair conformations. Relative to saturated heterocyclic compounds, the effect of the sulfur atom on ring puckering was attenuated. However, an increase in ring puckering resulted from the sulfone group and was attributed to torsional energy.

The observed spectra are very dependent upon the choice of instrument parameters such as the gas target density, the mode of operation of the central quadrupole, and the direct current levels of the quadrupoles (rod-offsets) which determine the collision energy and the maximum mass resolutions in each analysing quadrupole.     Notes: A detailed exploration of the application of triple quadrupoles in sequential mass spectrometry has been carried out using dimethylmorpholinophosphoramidate as a case study of an ion of relatively high mass and complex structure. Both metastable ion decomposition and dissociation by low energy collision were examined. Variation of the basic instrument parameters allowed the investigation of the cross-section for dissociation as a function of collision energy, the change in fragmentation patterns with collision energy, the kinetic energy distributions of daughter ions and the overall kinetics of dissociation in a succession of ion decomposition sequences giving daughter and granddaughter ions. The daughter ions could also be produced in the source region and examined separately so that a genealogical tree could be established.
