    Notizen: A variety of diploid human fibroblast lines have been successfully grown to high densities (〉106 cell/ml) on recently developed microcarriers. Interferon induction using poly I·poly C and a superinduction procedure resulted in yields greater than 10,000 units/ml with one cell line. A direct comparison of microcarrier cultures to roller bottle cultures showed equivalent interferon yields on a per cell basis and some apparent differences relating to optimum inducer concentrations and kinetics of interferon accumulation.

    Notizen: A study of the application of the Finite Element Method to compressible potential flows, typified by the airfoil problem, is undertaken. Some novel approaches, believed to simplify solution techniques, are presented.The solutions use two pseudo-variational integrals, appropriate to subsonic flows, and possessing a physical iterative basis. With constant-derivatives triangular elements formulated for cylindrical co-ordinates, accurate solutions are easily obtained for the flow over a circular cylinder. For arbitrary airfoils a simple mapping is used to transform them into near circles. An appropriate mesh is then constructed in the mapped plane. The paper then presents two solution approaches by which this non-linear problem is solved in both the near circle plane and the airfoil plane.

    Notizen: The electronic properties of a variety of methylated isoalloxazine derivatives have been investigated with absorption and fluorescece techniques (Table 1). The spectral effects of methyl substitution at different positions in the isoalloxazine ring are interpreted in terms of different bond orders between the C-atoms of the methyl groups and of the aromatic ring. Methyl groups at C(6) or C(9) induce quenching of the fluorescence and shortening of the lifetime whereas a methyl group at C(7) has an opposite effect. The quenching effect might be due to strain in the molecular framework caused by the methyl groups.The lowest energy levels of the isoalloxazine cations are shifted to higher energy as compared to the neutral isoalloxazines. Apart from these shifts the trends in spectral behaviour upon methylation are similar to those observed for the neutral isoalloxazine (cf. Table 2). The fluorescence of the isoalloxazine cations is strongly quenched in auqeous solution and their lifetime much shortened. These observations indicated that there is substantial interaction between the charged fluorophore and the water dipoles and that this interaction produces efficient radiationless deactivation of the first excited singlet state.

    Notizen: This paper described a technique for obtaining three-dimensional mine design information using a two-dimensional finite element program where the mining geometry consists of an extensive array of underground rooms and pillars. The technique is based upon a simple augmentation of forces in a two-dimensional analysis to produce the same average pillar stress that would occur in a full three dimensional analysis. Detailed comparisons between a three-dimensional analysis, a two-dimensional analysis (plane stress and plane strain) and an augmented two-dimensional analysis (also plane stress and plane strain) of stress about a typical coal mine pillar are presented. A local factor of safety is defined and then mapped over the pillar midplane, the immediate roof and immediate floor using the results from the full three-dimensional analysis. Comparisons of roof and pillar safety factor distributions obtained by the three-dimensional, two-dimensional and augmented two-dimensional analyses show that the minimum safety factors in the pillar (at the pillar sides) are predicted quite closely by the augmented two-dimensional techniqe (plane stress). The same is true of the immediate roof, although the three-dimensional safety factor tends to be higher in the roof (over the room) than that calculated by the augmented twodimensional technique. The augmented loading procedure appears to hold considerable promise as a very efficient and cost reducing techniqe for mine pillar design.

    Notizen: Thermal analysis and kinetic studies have shown that oxidative reactions are responsible for acceleration in the rates of weight loss and depolymerization of cellulose on pyrolysis in air at temperatures below 300°C. The oxidative reactions include production of hydroperoxide, carbonyl, and carboxyl groups, which have been investigated at lower temperatures along with the rates of depolymerization and production of carbon monoxide and carbon dioxide. The experimental results are consistent with an autoxidation mechanism involving initiation, propagation, and decomposition reactions. At temperatures above 300°, the rate of pyrolysis is essentially the same in both air and nitrogen, indicating that thermal degradation is independent of the oxidative reactions.

    Notizen: It has been shown that the pyrolysis of cellulose at low pressure (1.5 Torr) can be described by a three reaction model. In this model, it is assumed that an “initiation reaction” leads to formation of an “active cellulose” which subsequently decomposes by two competitive first-order reactions, one yielding volatiles and the other char and a gaseous fraction. Over the temperature range of 259-341°C, the rate constants of these reactions, ki (for cellulose → “active cellulose”), kv (for “active cellulose” → “volatiles”), and kc (for “active cellulose” → char + the gaseous fraction) are given by ki = 1.7 × 1021e- (58,000/RT) min -1, kv = 1.9 × 1016e- (47,300/RT) min-1, and kc = 7.9 × 1011e- (36,600/RT) min-1, respectively.

    Notizen: Evidence was obtained supporting the theory that monomer droplets can become a locus of initiation and propagation in emulsion polymerization. This was done by reducing the size of the monomer droplets prior to initiation of polymerization using different preemulsification techniques for a typical latex recipe containing a common anionic surfactant. Monomer droplet size reduction caused an increase in the total surface area of the monomer droplets and thereby increased the competitiveness of these droplets for capturing active free radicals which are generated in the aqueous phase. As the total surface area of the monomer droplets was increased by preemulsification, a corresponding increase in the number of large particles formed by polymerization of these monomer droplets was experimentally measured. This work shows that monomer droplets are a locus of emulsion polymerization. However, the importance of monomer droplet polymerization is limited by their total surface area because of the competition for free radicals with other surfaces and/or particle nucleation processes. The results offer a basis for explaining why broad, and sometimes bimodal, particle size distributions are obtained in some commercial processes where the reactants are preemulsified prior to being charged into a reaction vessel.

    Notizen: The degradation resistance of matrix, fiber and composite systems which we have been studying as candidate orthopedic materials has been examined in two appropriate environments. Both resistance to steam sterilization in an autoclave environment and resistance to a simulated physiologic solution have been studied. In the autoclave study, samples were placed in a pressure cooker at 123°C for differing amounts of time and tested for retention of mechanical properties. Results indicate that most of the materials tested could be autoclaved several times, as along as autoclave times did not exceed 1 hr. Longer autoclave times result in an accelerated degradation and loss of strength of all materials except the polypropylene. Polysulfone degrades after even the shortest autoclave duration. Resistance to the simulated physiologic environment was tested by measuring retention of mechanical properties after immersion times in pseudo-extracellular fluid (PECF) at 37°C for as long as three years. None of the materials showed any significant changes in properties after immersion in the PECF.

    Notizen: The intracellular accumulation of free [3H] adenosine was measured by rapid kinetic techniques in P388 murine leukemia cells in which adenosine metabolism (phosphorylation and deamination) was completely prevented by depletion of cellular ATP and by treatment with deoxycoformycin. Nonlinear regression of integrated rate equations on the data demonstrate that the time courses of labeled adenosine accumulation at various extracellular adenosine concentrations in zero-trans and equilibrium exchange protocols are well described by a simple, completely symmetrical, transport model with a carrier:substrate affinity constant of about 150 μM. Adenosine transport was not affected by 1 mM deoxycoformycin indicating that this analog has a low affinity for the nucleoside transport system. The transport capacity of dog thymocytes and peripheral leukocytes was similar to that of P388 cells. Transport was not inhibited by deoxycoformycin and remained constant during the first two hours after mitogenic stimulation with concanavalin A.In untreated, metabolizing P388 cells transport was found to be the major determinant of the rate of intracellular metabolism, regardless of the extracellular adenosine concentration (up to at least 160 μM), but the long-term accumulation (longer than 30-60 seconds) of radioactivity from extracellular adenosine strictly reflected the rate of formation of nucleotides (mainly ATP). The metabolism of adenosine by whole cells was entirely consistent with the kinetic properties of the transport system and those of the metabolic enzymes. At low exogenous adenosine concentrations (1 μM and below) transport was slow enough to allow direct phosphorylation of most of the entering adenosine. The remainder was deaminated and rapidly converted to nucleotides via inosine, hypoxanthine, and IMP. At concentrations of 100 μM or higher, on the other hand, influx exceeded the maximum velocity of adenosine kinase about 100 times so that most of the entering adenosine was deaminated. But since the maximum velocity of adenosine deaminase exceeded those of nucleoside phosphorylase and hypoxanthine/guanine phosphoribosyltransferase about 5 and 100 times, respectively, hypoxanthine and inosine rapidly exited from the cells and accumulated in the medium. A 98% reduction of adenosine transport (at 100 μM), caused by the transport inhibitor Persantin, inhibited adenosine deamination by whole cells to about the same extent as transport, whereas adenosine phosphorylation was relatively little affected; thus in the presence of Persantin, transport and metabolism resembled that occurring at the low adenosine concentration. These and other results indicate that adenosine deamination is an event distinct from transport, which occurs only subsequent to adenosine's transport into the cell.
