    Notes: Using heavily methionine-substituted T4 lysozyme as an example, it is shown how the addition or deletion of a small number of methionines can simplify the location of selenium sites for use in MAD phasing. By comparing the X-ray data for a large number of singly substituted lysozymes, it is shown that the optimal amino acid to be substituted by methionine is leucine, followed, in order of preference, by phenylalanine, isoleucine and valine. The identification of leucine as the first choice agrees with the ranking suggested by the Dayhoff mutation probability, i.e. by the frequency of amino-acid substitutions in the sequences of related proteins. The ranking of the second and subsequent choices, however, differ significantly.

    Notes: Abstract  Water proton T 1 –1 measurements at magnetic fields between 0.01 and 50 MHz [nuclear magnetic relaxation dispersion (NMRD) measurements] have been performed on solutions of phthalate dioxygenase (PDO) reconstituted at the catalytic iron site with copper(II) or manganese(II). The data show evidence of a weakly coordinated water molecule in CuPDO; in the presence of the substrate, phthalate, this water appears to become even less tightly bound, and an additional tightly coordinated water can be detected. In PDO reconstituted with manganese, one tightly coordinated water is detected in the presence and in the absence of phthalate. An attempt is made to reconcile these data with low-temperature near-IR magnetic circular dichroism and X-ray absorption data, which show that PDO reconstituted with iron or cobalt is six-coordinate in the absence of substrate and five-coordinate in the presence of substrate.

    Notes: The melting, crystallization and dynamic mechanical behaviour of blends of bacterially produced poly[D(-)-3-hydroxybutyrate] (PHB) and poly[D(-)-3-hydroxyvalerate] (PHV) have been investigated. Results showed that melt-pressed PHB-PHV blends contained phase-separated domains in the melt which subsequently crystallized as PHB and PHV type spherulites respectively. The two melting regions detected by DTA related to separate melting of PHB and PHV crystallites, which were almost unaffected by the blend composition. The mechanical behaviour of a random copolymer of PHB/HV was compared with that of a blend of almost the same composition, and found to be markedly different.

    Notes: New dumb-bell-type fullerene adducts 2, 4, and 8a-b could be synthesized via bifunctional nitrile oxides. The [60]fullerene derivative 16 of this type was synthesized by twofold esterification with an isoxazolo[60]fullerenecarboxylic acid derivative. In the latter case the reaction was used to link the archimedic polyhedron fullerene with the platonic element cubane. When a bulky spacer between the fullerene units was used, atropisomeres 8a-b occurred. The fullerene derivative 11 with an anthracene moiety exhibited axial chirality.
