    Notes: Abstract The crossover breakpoints for Charcot-Marie-Tooth disease type 1A (CMT1A) and hereditary neuropathy with liability to pressure palsies (HNPP) are located in the CMT1A-REP repeat flanking a 1.5-Mb region of chromosome 17p11.2–12. The precise locations of the breakpoints are heterogeneous, and we analyzed the relative frequency distribution of breakpoints in 33 unrelated Japanese CMT1A and 3 unrelated HNPP families. The CMT1A-REP repeat region was divided into five regions, A, B, C, D and E, based on restriction site differences between the proximal and distal CMT1A-REP repeats. The frequency distribution of breakpoints within the CMT1A-REP repeat in the Japanese patients was 3% in region A, 78% in B/C and 19% in D, which is similar to that in Caucasian patients. This result also indicates that an 8-kb region defined by region B/C is a recombinational hotspot within the CMT1A-REP repeat in Japanese patients.

    Notes: The effects of the ratio of Rubisco activase to Rubisco (activase/Rubisco ratio) on light dependent activation of CO2 assimilation were investigated during leaf aging of rice. Changes of photosynthetic CO2 gas exchange rates in relation to step increases of light intensity from two photon flux densities of 60 µmol m−2 s−1 (low initial PFD) and 500 µmol m−2 s−1 (high initial PFD) to saturated PFD of 1 800 µmol m−2 s−1 were measured. These photosynthetic activation processes were considered to be limited by the Rubisco activation rate when analyzed by the relaxation method. The relaxation time of low initial PFD gradually declined from 3 to 33 days after leaf emergence and showed high and negative correlation to the activase/Rubisco ratio. The initial rate of Rubisco activation under low initial PFD linearly correlated to the amounts of Rubisco activase, whereas these were almost constant from 3 to 23 days after leaf emergence. But these correlations could not be recognized in the case of high initial PFD. Moreover, the relaxation times were more sensitive to intercellular CO2 concentration (Ci) under high initial PFD than under low initial PFD, especially, at Ci below 300 µl l−1. These results suggest the involvement of the activase/Rubisco ratio in the photosynthetic activation under relatively low initial PFD, and the limitation of photosynthetic activation under relatively high initial PFD by Rubisco carbamylation during leaf aging of rice.

    Notes: Internodal elongation in floating rice (Oryza sativa L. cv. Habiganj Aman II) is known to be enhanced by treatment with ethylene or gibberellic acid (GA3) at high relative humidity (RH). However, ethylene-induced internodal elongation is inhibited at low RH. while GA3-induced internodal elongation is hardly affected by humidity. We examined the possible involvement of osmoregulation in the stimulation by GA3 of the elongation of internodes at low RH. Submergence and treatment with ethylene or GA33 at 100% RH increased the osmotic potential in internodes of excised stem segments, while GA3 at 20% RH maintained the osmotic potential at a low level. In internodes of stem segments that had been treated with GA3 at 20% RH, the activity of invertase and the level of soluble sugars were almost 2- and 1.5-fold higher, respectively, than those in internodes that had been treated with GA3 at 100% RH. These results indicate that one of the possible mechanisms by which GA3 promotes elongation of internodes at low RH involves the osmoregulation that is achieved by promotion of the synthesis of invertase.

    Notes: The cDNA clone encoding a putative germin-like protein (GLP) was isolated from the halophyte Atriplex lentiformis (Torr.) Wats. The deduced amino acid sequence of the protein showed high homology to those of various plant germins or germin-like proteins. The protein was designated as AlGLP (A. lentiformis germin-like protein). It was found that AlGLP has superoxide dismutase (SOD; EC 1.15.1.1) activity by in-gel SOD assays after immunoprecipitation, and that it is a glycosylated oligomer in native form. The AlGLP was strongly expressed in calli and weakly in roots, but not in stems or leaves. Interestingly, the transcript level in roots was decreased by salt or abscisic acid (ABA) treatment. We present evidence that the accumulation level of AlGLP mRNA in leaves is increased by methyl jasmonate treatment or wounding, and that the induction is suppressed by ABA treatment. These results suggest that AlGLP expression is increased to fulfil H2O2 supply to modify the structure of cell wall in response to wounding.

    Notes: Addition of plant growth regulators (5 nM NAA and 5μM BAP) to a defined basal medium stimulated adventitious bud formation of Douglas fir (Pseudotsuga menziesii [Mirb.] Franco) cotyledon explants in culture. Cytoplasmic soluble proteins synthesized during early stages of adventitious bud formation were analyzed by electrophoresis of 3H- and 14C-leucine labeled proteins on SDS polyacrylamide gels. Increased synthesis of low molecular weight proteins (16,000 to 20,000 daltons) was detected after 2 days in culture and reached a maximal level at day 4. When cotyledon explants cultured on bud medium for 2 days were transferred to callus medium (which suppressed adventitious bud formation), suppression of the synthesis of low molecular weight proteins was also observed, suggesting that these proteins may be associated with early stages of adventitious bud formation.

    Notes: Newly synthetized cyloplasmic proteins of Douglas fir cotyledon culture capable of initiating adventitious bud formation (bud culture) were compared with those of morphologically distinct types of cultures, bud-callus culture (some adventitious buds with callus), and callus culture (no adventitious buds; only callus). SDS polyacrylamide gel electrophoretic profiles of double labeled (14C-and 3H-) proteins showed that a persistent increase of low molecular weight proteins (16,000 to 20,000 daltons) was associated with bud culture. Similar electrophoretic profiles of proteins extracted in the absence and in the presence of protease inhibitor (4-mcthylbenzene sulfonyl fluoride) or phenolic compounds absorbents (Dowex I-X8 and polyvinylpolypyrolidine) were obtained indicating that the detected low molecular weights of these proteins represented the real monomeric state of these molecules.

    Notes: During the induction of DNA synthesis in Jerusalem artichoke (Helianthus tuberosus L.) tuber by 2,4-D, the 2-14C-2, 4-D from the agar medium rapidly incorporated into the ethanol soluble and insoluble fractions. Although the 2,4-D level in the ethanol soluble fraction decreased on transplantation of the tissue from the 2-14C-2,4-D medium to medium without the auxin, its level in the buffer-soluble and -insoluble macromolecular fractions increased. The purified, buffer-insoluble macromolecules were chromatin. The 2,4-D binding to chromatin particularly increased during DNA synthesis. The histone contents of chromatin decreased as DNA synthesis progressed. The polyacrylamide gel electrophoretic patterns of the histones showed a decrease in the moderately lysine-rich histone fraction as compared to other fractions. Thus, the decrease in the histone level caused by 2,4-D and the presence of the 2,4-D moderately lysine-rich histone complex may be closely related to the induction of DNA synthesis by 2,4-D in cells.

    Notes: Abstract In excised stem segments of floating rice (Oryza sativa L.), as well as in intact plants, submergence greatly stimulates the elongation of internodes. The differences in the composition of cell wall polysaccharides along the highest internodes of submerged and air-grown stem segments were examined. The newly elongated parts of internodes that had been submerged for two days contained considerably less cellulosic and noncellulosic polysaccharides than air-grown internodes, an indication that the cell walls of the newly elongated parts of submerged internodes are extremely thin. In the young parts of both air-grown and submerged internodes, the relative amounts of noncellulosic polysaccharides were equal to those of α-cellulose, whereas the relative amounts of α-cellulose were higher than those of noncellulosic polysaccharides in the upper, old parts. In the cell-elongation zones of both air-grown and submerged internodes, glucose was predominant among the noncellulosic neutral sugars of cell wall. The relative amount of glucose in noncellulosic neutral sugars decreased toward the upper, old parts of internodes, whereas that of xylose increased.
