    Description: Aptamers have emerged as promising molecular probes for in vivo cancer imaging, but the reported “always-on” aptamer probes remain problematic because of high background and limited contrast. To address this problem, we designed an activatable aptamer probe (AAP) targeting membrane proteins of living cancer cells and achieved contrast-enhanced cancer visualization inside mice. The AAP displayed a quenched fluorescence in its free state and underwent a conformational alteration upon binding to target cancer cells with an activated fluorescence. As proof of concept, in vitro analysis and in vivo imaging of CCRF-CEM cancer cells were performed by using the specific aptamer, sgc8, as a demonstration. It was confirmed that the AAP could be specifically activated by target cancer cells with a dramatic fluorescence enhancement and exhibit improved sensitivity for CCRF-CEM cell analysis with the cell number of 118 detected in 200 μl binding buffer. In vivo studies demonstrated that activated fluorescence signals were obviously achieved in the CCRF-CEM tumor sites in mice. Compared to always-on aptamer probes, the AAP could substantially minimize the background signal originating from nontarget tissues, thus resulting in significantly enhanced image contrast and shortened diagnosis time to 15 min. Furthermore, because of the specific affinity of sgc8 to target cancer cells, the AAP also showed desirable specificity in differentiating CCRF-CEM tumors from Ramos tumors and nontumor areas. The design concept can be widely adapted to other cancer cell-specific aptamer probes for in vivo molecular imaging of cancer.

    Description: Using linear regression (parametric), Mann-Kendall (non-parametric) and attribution analysis methods, this study systematically analyzed the changing properties of reference evapotranspiration (ETr) calculated using the Penman–Monteith method over the Poyang Lake catchment during 1960–2008, and investigated the contribution of major climatic variables to ETr changes and their temporal evolution. Generally, a significant decreasing trend of annual ETr is found in the catchment. The decrease of annual ETr in the Poyang Lake basin is mostly affected by the decline of summer ETr. Over the study period, climatic variables, i.e. sunshine duration (SD), relative humidity (RH), wind speed (WS) and vapour pressure (VP) all showed decreasing trends, while mean daily temperature (DT) increased significantly. Multivariate regression analysis indicated that SD is the most sensitive climatic variable to the variability of ETr on annual basis, followed by RH, WS and DT, while the effect of VP is obscure. Although recent warming trend and decrease of relative humidity over the catchment could have increased ETr, the combined effect of shortened sunshine duration and reduced wind speed negated the effect and caused significant decrease of ETr. Our investigation reveals that the relative contributions of climatic variables to ETr are temporally unstable and vary considerably with large fluctuation. In consideration of the changes of climatic variables over time, further analysis indicated that changes of mean annual ETr in 1970–2008 were primarily affected by SD followed by WS, RH and DT with reference to 1960s. However, WS became the predominant factor during the period 2000–2008 compared to reference period 1960s, and followed by SD. This article is protected by copyright. All rights reserved.

    Description: In order to study the flow characteristic in turbulence generator of medium consistency pump, a particle image velocimetry (PIV) test rig was established. 2D-plane flow field was acquired fast and effective by adjusting the angle and position of mirror. For investigating the effect of speed on flow field, velocity and turbulent kinetic energy were measured at speed 80r/min, 130r/min and 200r/min. Dimensionless method was adopted to analyze flow field by quantitative approach. The results showed that on vertical flow plane axial velocities decreased with radius increasing in the region of turbulence generator blade, and axial velocity direction was changed and increased with radius increasing outside the region of turbulence generator blade.     Description: 〈p〉Steering the evolution of single spin systems is crucial for quantum computing and quantum sensing. The dynamics of quantum systems has been theoretically investigated with parity-time–symmetric Hamiltonians exhibiting exotic properties. Although parity-time symmetry has been explored in classical systems, its observation in a single quantum system remains elusive. We developed a method to dilate a general parity-time–symmetric Hamiltonian into a Hermitian one. The quantum state evolutions ranging from regions of unbroken to broken 〈f〉〈/f〉 symmetry have been observed with a single nitrogen-vacancy center in diamond. Owing to the universality of the dilation method, our result provides a route for further exploiting and understanding the exotic properties of parity-time symmetric Hamiltonian in quantum systems.〈/p〉

    Description: Whole-genome search of genes is an essential approach to dissecting complex traits, but a marginal one-single-nucleotide polymorphism (SNP)/one-phenotype regression analysis widely used in current genome-wide association studies fails to estimate the net and cumulative effects of SNPs and reveal the developmental pattern of interplay between genes and traits. Here we describe a computational framework, which we refer to as two-side high-dimensional genome-wide association studies (2HiGWAS), to associate an ultrahigh dimension of SNPs with a high dimension of developmental trajectories measured across time and space. The model is implemented with a dual dimension-reduction procedure for both predictors and responses to select a sparse but full set of significant loci from an extremely large pool of SNPs and estimate their net time-varying effects on trait development. The model can not only help geneticists to precisely identify an entire set of genes underlying complex traits but also allow them to elucidate a global picture of how genes control developmental and dynamic processes of trait formation. We investigated the statistical properties of the model via extensive simulation studies. With the increasing availability of GWAS in various organisms, 2HiGWAS will have important implications for genetic studies of developmental compelx traits.

    Description: 〈p〉A parasite-infected host may promote performance of associated insect vectors; but possible parasite effects on nonvector insects have been largely unexplored. Here, we show that 〈i〉Begomovirus〈/i〉, the largest genus of plant viruses and transmitted exclusively by whitefly, reprogram plant immunity to promote the fitness of the vector and suppress performance of nonvector insects (i.e., cotton bollworm and aphid). Infected plants accumulated begomoviral βC1 proteins in the phloem where they were bound to the plant transcription factor WRKY20. This viral hijacking of WRKY20 spatiotemporally redeployed plant chemical immunity within the leaf and had the asymmetrical benefiting effects on the begomoviruses and its whitefly vectors while negatively affecting two nonvector competitors. This type of interaction between a parasite and two types of herbivores, i.e., vectors and nonvectors, occurs widely in various natural and agricultural ecosystems; thus, our results have broad implications for the ecological significance of parasite-vector-host tripartite interactions.〈/p〉

    Description: 〈p〉The Rag/Gtr GTPases serve as a central module in the nutrient-sensing signaling network upstream of TORC1. In yeast, the anchoring of Gtr1-Gtr2 to membranes depends on the Ego1-Ego2-Ego3 ternary complex (EGO-TC), resulting in an EGO-TC-Gtr1-Gtr2 complex (EGOC). EGO-TC and human Ragulator share no obvious sequence similarities and also differ in their composition with respect to the number of known subunits, which raises the question of how the EGO-TC fulfills its function in recruiting Gtr1-Gtr2. Here, we report the structure of EGOC, in which Ego1 wraps around Ego2, Ego3, and Gtr1-Gtr2. In addition, Ego3 interacts with Gtr1-Gtr2 to stabilize the complex. The functional roles of key residues involved in the assembly are validated by in vivo assays. Our structural and functional data combined demonstrate that EGOC and Ragulator-Rag complex are structurally conserved and that EGO-TC is essential and sufficient to recruit Gtr1-Gtr2 to membranes to ensure appropriate TORC1 signaling.〈/p〉
