    Notes: An electronic device is described which allows for counting of the filling/emptying cycles which occur during high-pressure soxhlet extraction. Previously, the number of cycles which occurred during an extraction run were counted manually from a strip chart recording of the thermal excursions associated with the cycling events. This device automates the detection, accumulation, and display of these events and was found to exhibit a counting accuracy of at least 94%. It extends the application of high-pressure soxhlet extraction to areas which routinely require a high degree of precision.

    Notes: Abstract Proliferating cultures ofGarrya elliptica were established from shoot tip and nodal explants grown on woody plant medium containing benzyladenine. Optimum numbers of healthy shoots were induced on media containing 1.25 μM benzyladenine; higher concentrations caused more hyperhydricity and less extension. Following growth for 28 days on quarter-strength woody plant medium containing 0.2% activated charcoal, 90% of shoots rooted on the same medium lacking charcoal, but with 2.5 μM indolebutyric acid. Survival during weaning was poor, and was strongly correlated with the time of year at which plants were removed from culture.

    Notes: Epidermal growth factor and cartilage-derived basic fibroblast growth factor (EGF and CD-bFGF) are mitogens shown to increase the rate of wound repair in animal models. In addition to being a mitogen for granulation tissue, CD-bFGF stimulates the recruitment of cells to the wound site. CD-bFGF and a closely-related chondrosarcoma-derived fibroblast growth factor stimulated chemotaxis of granulation tissue cells in vitro, each factor having a maximum activity at a concentration of 55 pM. Epidermal growth factor was also a potent chemoattractant for rat granulation tissue fibroblasts; however, maximum activity was obtained at 1.7 nM. Cells from all stages of wound repair were chemotactically responsive to these factors, but there was some attenuation of the response to bFGF in cells derived from fully-organized day 28 granulation tissue. Collagenase-catalyzed restructuring of collagen, an additional significant feature of wound repair, is probably critical to cell movement in an extracellular matrix. Cells derived from organizing (6-day old) sponge granulation tissue secreted latent collagenase constituitively in vitro. In the presence of serum, the production of collagenase was stimulated three-four fold by 1.8 nM bFGF derived either from cartilage or chondrosarcoma. When serum was present, as at a wound site, collagenase production was not enhanced by the addition of EGF. Cells from fully organized, day 21 sponge granulation tissue did not secrete latent collagenase constitutively and could not be stimulated to do so by the addition of EGF, bFGF, or phorbol ester. Human skin fibroblast collagenase production was also stimulated by bFGF and was refractory to EGF. While both classes of growth factor have the ability to promote wound healing, the varying responses they elicit in cell populations from the wound site emphasize the different pathways of cellular activation.

    Notes: Sutured, stapled, and methyl- or thickened n-pentyl-2-cyanoacrylate-adhered end-to-end arterial anastomoses were compared in two series of 10 dogs, employing the three 4-mm diameter terminal branches of the aorta for these comparisons. Patency rate, speed of construction, and gross and microscopic appearances were employed as criteria for evaluation. In a series of adult white rabbits the same criteria were applied to compare sutured and thickened n-pentyl-2-cyanoacrylateadhered end-to-end anastomoses of the 2-mm diameter abdominal aorta. Anastomoses adhered with methyl-2-cyanoacrylate were significantly more often thrombosed than any other variety; a marked inflammatory response followed application of this material. Thickened n-pentyl-2-cyanoacrylate adhesive was well tolerated, and the results obtained with this material were equivalent to those obtained by suturing or stapling in the dog, or by suturing in the rabbit.

    Notes: A series of tests permitting evaluations of cyanoacrylates as tissues adhesives is presented. Closed coaption of skin is an effective, rapid screening test capable of identifying weak and strong adhesives under use conditions, and is more sensitive for this purpose than bond strength determinations employing aortic lap joints. Tensile strength testing employing skin incisions closed with cyanoacrylates permits evaluation of the continuing effects of cyanoacrylate upon healing skin. In this test system, a number of alkyl cyanoacrylates provides results equivalent to those obtained with sutures. The incorporation of cyanoacrylates into subcutaneous implants permits evaluation of the effects of these materials upon fibroplasia and collagen elaboration, the most critical aspects of wound repair. Implant testing also permits estimation of rates of resportion of cyanoacrylates from subcutaneous deposits.

    Description: Fusarium circinatum, the causal agent of pine pitch canker (PPC), is currently one of the most important threats of Pinus spp. globally. This pathogen is known in many pine-growing regions, including natural and planted forests, and can affect all life stages of trees, from emerging seedlings to mature trees. Despite the importance of PPC, the global distribution of F. circinatum is poorly documented, and this problem is also true of the hosts within countries that are affected. The aim of this study was to review the global distribution of F. circinatum, with a particular focus on Europe. We considered (1) the current and historical pathogen records, both positive and negative, based on confirmed reports from Europe and globally; (2) the genetic diversity and population structure of the pathogen; (3) the current distribution of PPC in Europe, comparing published models of predicted disease distribution; and (4) host susceptibility by reviewing literature and generating a comprehensive list of known hosts for the fungus. These data were collated from 41 countries and used to compile a specially constructed geo-database. A review of 6297 observation records showed that F. circinatum and the symptoms it causes on conifers occurred in 14 countries, including four in Europe, and is absent in 28 countries. Field observations and experimental data from 138 host species revealed 106 susceptible host species including 85 Pinus species, 6 non-pine tree species and 15 grass and herb species. Our data confirm that susceptibility to F. circinatum varies between different host species, tree ages and environmental characteristics. Knowledge on the geographic distribution, host range and the relative susceptibility of different hosts is essential for disease management, mitigation and containment strategies. The findings reported in this review will support countries that are currently free of F. circinatum in implementing effective procedures and restrictions and prevent further spread of the pathogen.

    Description: Background: Sentinel Lymph Node (SLN) sampling may significantly reduce surgical morbidity by avoiding needless radical lymphadenectomy. In gynaecological cancers, the current practice in the UK is testing the accuracy of SLN detection using radioactive isotopes within the context of clinical trials. However, radioactive tracers pose significant logistic problems. We, therefore, conducted a pilot, observational study to assess the feasibility of a novel optical imaging device for SLN detection in gynaecological cancers using near infrared (NIR) fluorescence. Methods: A novel, custom-made, optical imaging system was developed to enable detection of multiple fluorescence dyes and allow simultaneous bright-field imaging during open surgery and laparoscopic procedures. We then evaluated the performance of the system in a prospective study of 49 women with early stage vulval, cervical and endometrial cancer who were scheduled to undergo complete lymphadenectomy. Clinically approved fluorescent contrast agents indocyanine green (ICG) and methylene blue (MB) were used. The main outcomes of the study included SLN mapping detection rates, false negative rates using the NIR fluorescence technique and safety of the procedures. We also examined the association between injection sites and differential lymphatic drainage in women with endometrial cancer by fluorescence imaging of ICG and MB. Results: A total of 64 SLNs were detected during both open surgery and laparoscopy. Following dose optimisation and the learning phase, SLN detection rate approached 100 % for all cancer types with no false negatives detected. Fluorescence from ICG and MB detected para-aortic SLNs in women with endometrial cancer following uterine injection. Percutaneous SLN detection was also achieved in most women with vulval cancer. No adverse reactions associated with the use of either dyes were observed. Conclusions: This study demonstrated the successful clinical application of a novel NIR fluorescence imaging system for SLN detection across different gynaecological cancers. We showcased the first in human imaging, during the same procedure, of two fluorescence dyes in women with endometrial cancer.
