Notes: Summary A comparison of the sequence of DNA replication in an isodicentric (idic) X chromosome was made between peripheral blood lymphocytes and skin fibroblasts from a 33-year-old female with primary amenorrhea, somatic stigmata of Turner syndrome, and normal stature and intelligence. The patient had a karyotype 45,X/46,X,idic(X)(q27.1) in lymphocytes and 46,X,idic(X)(q27.1) in skin fibroblasts. Both centromeric regions of the idic X showed C-staining but only one primary constriction. BrdU-33258 Hoechst-Giemsa techniques were used to analyze regional DNA replication patterns. The idic X chromosome was always late replicating in lymphocytes and skin fibroblasts, except that about 1–2% of cells completed replication simultaneously in both normal and idic X chromosomes. Fifty-six percent of the asymmetric patterns in lymphocytes showed an equal proportion of early and late functional and non-functional centromere halves. In skin fibroblasts, 60.8% of cells were asymmetric: the functional half tended to replicate later than the non-functional half. Some differences were observed between these two cell types. As examples, band q23 was late replicating in lymphocytes, but early replicating in fibroblasts; q25 was intermediate to late replicating in lymphocytes, but one of the last bands to complete replication in fibroblasts. Thus, different cell typed influenced the replication kinetics in the idic(X). Furthermore, several variants of the replication sequence were found in both cell types. The findings support the hypothesis that the control of DNA replication in the inactive X chromosome is multifocal, and suggest that the active idic X chromosome replication may reflect a relative lack of self-control or heterogeneity of cell population.

Notes: Summary Variation of DA/DAPI intensity in the Yq12 band was observed in five amniotic cell specimens and one blood specimen from the father of one fetus. Three distinct classes of Yq heterochromatin were identified by distamycin A (DA) treatment of the cell cultures and various staining techniques. The heterochromatin in the Yq11.23 sub-band does not under-condense when exposed to DA, and shows pale fluorescence with quinacrine staining, positive C-banding, and bright fluorescence with DA/DAPI technique. This class of heterochromatin was consistently observed in all specimens studied. The other two classes of heterochromatin are in the Yq12 band. Both show undercondensation when exposed to DA, quinacrine-bright fluorescence, and positive C-banding; howover, one class of heterochromatin shows DA/DAPI-bright fluorescence and the other shows pale fluorescence. The size and banding intensity of the two classes of heterochromatin in Yq12 are variable. These results provide cytological evidence of heterogeneity within the Y heterochromatin region containing AT-rich DNA.

Notes: Abstract An analysis is given of phase-reversal electrode structures used for phase matching in travelling-wave integrated-optic phase modulators. Both modulus and argument of the phase shift are obtained; both are required for applications involving broadband excitation. Periodic and non-periodic lossy structures are treated and the advantage of non-periodic structures for wide-band operation is demonstrated. Optimum structure arrangements and upper bounds to phase shift and figure of merit are derived.

Notes: Abstract Spectral changes in inhomogeneous media depending on the spatial coherence of the source can be described by a quasi-optical approach. Assuming a Gaussian-Schell model source the spectral changes are described by using a Gaussian beam and ABCD matrices for the propagation in the medium.

Notes: Abstract In the past decade spectral changes depending on the spatial coherence of the source have been described by a cross-spectral density method. It is shown that for Gaussian-Schell model sources a quasi-optical approach is valid which uses Gaussian beams and ABCD matrices to describe the optical system.

Notes: Summary The changes in tension produced by small step or sinusoidal changes of length have been measured for chemically skinned flight muscle fibres of the giant tropical water bugLethocerus at MgATP concentrations in the range 0.1–10 mM. In the presence of calcium ion concentrations of about 20 µm, the rates of the rapid mechanical processes observed were found to increase with increasing MgATP, exhibiting saturation with an apparent half-saturation constant lying between 0.1 and 1mM MgATP, depending upon the conditions used. Under the same conditions, an increase in MgATP concentration was found to lead to a slight decrease in the isometric tension generated by the preparations. The results are discussed with reference to some current crossbridge models of muscle contraction.

Notes: Summary The heat and force produced in tetanic contraction of single fibres from anterior tibialis muscle of the frogRana temporaria have been observed at measured temperatures close to 1 and 10° C. Heat was measured using a Hill-Downing type thermopile. In control experiments with a resistor of known heat capacity comparable to a single muscle fibre, it was found that Peltier and Joule heating produced identical thermopile outputs. The Peltier method was used to introduce a known amount of heat into the system in each experiment with a muscle fibre. From the response to this heating the heat capacity of each preparation was obtained and used to calculate the absolute amount of heat production from the thermopile output. The heat produced during tetanic contraction (H) could be described by Aubert's equation [H=H a (1−e t/τ)+h b t]. In some fibres there was no labile heat (Ha), whereas in others it was clearly present. The stable heat rate (h b ) was strongly temperature dependent (Q 10 = 4.06). At 0° C the stable heat rate (normalized by dry weight) in the single fibres was significantly greater than that in whole anterior tibialis muscle.
