    Description: Our improved capability to adapt to future changes in discharge is linked to our capability to predict the magnitude or at least the direction of these changes. For the agricultural U.S. Midwest, too much or too little water has severe socio-economic impacts. Here we focus on the Raccoon River at Van Meter, Iowa, and use a statistical approach to examine projected changes in discharge. We build on statistical models using rainfall and harvested corn and soybean acreage to explain the observed discharge variability. We then use projections of these two predictors to examine the projected discharge response. Results are based on seven global climate models part of the Coupled Model Intercomparison Project Phase 5 and two representative concentration pathways (RCPs 4.5 and 8.5). There is not a strong signal of change in the discharge projections under the RCP 4.5. However the results for the RCP 8.5 point to a stronger changing signal related to larger projected increases in rainfall, resulting in increasing trends in particular in the upper part of the discharge distribution (i.e., 60th percentile and above). Examination of two hypothetical agricultural scenarios indicates that these increasing trends could be alleviated by decreasing the extent of the agricultural production. We also discuss how the methodology presented in this study represents a viable approach to move forward with the concept of return period for engineering design and management in a non-stationary world.

    Notes: Abstract Electrochemical modelling may provide fast order-of-magnitude estimates of energy and mass transfer in crops and partly fill the gap between rigid, expensive field experiments and oversimplified mathematical modelling. The technique comprises breaking up a flowing electrolyte in a manner analogous — but not necessarily completely similar — to the way the atmospheric flow is broken up by crops and studying the flow of ions in the electrolyte. The effects of variations in free-flow velocity, plant spacing, row spacing and orientation on velocity field, transfer coefficients and eddy diffusivities have been studied in an electrochemical model under conditions relevant to well-ventilated crops with relatively simple canopy structure. A one-dimensional analytical model is developed for the transfer at solid surfaces, with direct proportionality between transfer coefficient and eddy diffusivity. The proportionality constantα has values in the model of (3.5±0.5)×10−3cm−1 at the ground and (5+-2)×10−4cm−1 (for a cylindrical probe) near canopy top. The latter is not too different from the values proposed for foliage surfaces in real canopies, viz., approximately 1×10−3 cm−1 (Philip, 1964) and 7×10−4 cm−1 (Uchijima, 1966).α may therefore become a useful parameter in scaling transfer coefficients from systems with different molecular diffusion properties. A tentative extrapolation of measured data to corresponding values in air is given. The agreement between predicted values and presently available field data is encouraging although more precise field data are required for a final judgment on the validity of the model.

    Notes: Microthane foam, a poly(ester)urethane, used as a covering of some breast implants (Meme or Replicon by Surgitek) degrades readily in phosphate buffer pH 7.4 producing multiple unknown products in addition to 2,4 and 2,6 toluenediamine (TDA). The cumulative weight loss of the foam in buffer was 1.5% at 1 week and 2% at 2 weeks. Multiple peak molecular weights from 105 to 665 000 were found in the aqueous foam extracts following 2 weeks of incubation at 37°C using size exclusion chromatography (SEC). TDA was measured in the foam buffer extracts by high pressure liquid chromatography (HPLC) and confirmed by gas chromatography/mass spectroscopy (GC/MS). The total cumulative levels of 2,4 TDA and 2,6 TDA measured were 3 ± 0.65 μg/g of foam and 1.13 ± 0.19 μg/g of foam following 36 days of incubation at 37 °C. Higher amounts, 8.96 μg/g of 2,4 TDA and 3.3 μg/g of 2,6 TDA, were obtained at 50 °C. The cumulative release of TDA in aqueous extracts was found to correlate linearly with time suggesting a slow but continuous production of TDA under neutral conditions. However, less than 0.2 μg/g of TDA was detected as residual TDA in the methylene chloride extracts following up to 60 days at 37 °C. PU foam extracted with methylene chloride under similar conditions showed higher foam weight loss, 4.4 and 5% at 1 and 2 weeks, respectively. The results presented in this study provide further evidence that Microthane undergoes hydrolysis under neutral conditions in aqueous solutions producing TDA and other unknown low molecular weight components and oligomers. © 1994 John Wiley & Sons, Inc.

    Description: Prions are unconventional infectious agents that cause transmissible spongiform encephalopathy (TSE) diseases, or prion diseases. The biochemical nature of the prion infectious agent remains unclear. Previously, using a protein misfolding cyclic amplification (PMCA) reaction, infectivity and disease-associated protease-resistant prion protein (PrPres) were both generated under cell-free conditions, which supported a nonviral hypothesis for the agent. However, these studies lacked comparative quantitation of both infectivity titers and PrPres, which is important both for biological comparison with in vivo-derived infectivity and for excluding contamination to explain the results. Here during four to eight rounds of PMCA, end-point dilution titrations detected a 〉320-fold increase in infectivity versus that in controls. These results provide strong support for the hypothesis that the agent of prion infectivity is not a virus. PMCA-generated samples caused the same clinical disease and neuropathology with the same rapid incubation period as the input brain-derived scrapie samples, providing no evidence for generation of a new strain in PMCA. However, the ratio of the infectivity titer to the amount of PrPres (specific infectivity) was much lower in PMCA versus brain-derived samples, suggesting the possibility that a substantial portion of PrPres generated in PMCA might be noninfectious.
