    Notes: Conclusions The initial discovery of p-glycoprotein in the plasma membrane of MDR cancer cell lines was followed quickly by the cloning of its gene. Sequence analysis of cloned cDNAs revealed that p-glycoprotein was a member of the ABC family of membrane transporters. Subsequent biochemical characterization demonstrated the binding of chemotherapeutic drugs and ATP to p-glycoprotein. P-glycoprotein-mediated drag transport and drug-stimulated ATPase activity were documented in plasma membrane vesicles and in proteoliposomes containing the partially purified protein. P-glycoprotein was shown to be phosphorylated and the effect of this modification on the protein's biological function was explored. P-glycoproteins were found in many normal tissues and their overexpression was documented in numerous cancers. An important role for p-glycoprotein in intrinsic and acquired drug resistance in clinical oncology was established. Despite all that has been learned about p-glycoprotein over the last few years, additional studies will be necessary to address the many questions that have been left unanswered. Determination of p-glycoprotein structure and membrane topology should help elucidate the nature of chemotherapeutic drug binding sites and the mechanism whereby drug movement is coupled to ATP hydrolysis. Complete purification and functional reconstitution of p-glycoprotein into defined lipid vesicles will permit further characterization of drug transport and ATPase activity and give us the means by which p-glycoprotein's apparent dual function as a transporter and a channel can be clarified. Structural and functional studies on p-glycoprotein will also provide information needed to develop specified inhibitors that can be used clinically to overcome MDR in cancer patients. Further study of the mechanisms whereby p-glycoprotein expression is induced and regulated during malignant transformation is indicated. The development of biliary phospholipid deficiency in mdr2 knockout mice and xenobiotic hypersensitivity in mdr3 knockout mice have given us the first clues into the normal physiologic roles for the p-glycoproteins. The search for endogenous substrates for the p-glycoproteins will continue to be an area of active investigation. Continued investigation of p-glycoprotein's functions should result in better understanding of an important class of prokaryotic and eukaryotic membrane transporters. The potential of exploiting the knowledge garnered from these studies in the treatment of neoplastic, parasitic and inherited and acquired liver disease may be greater than we can now imagine.

    Notes: Abstract  The existence of soluble organic forms of N in rain and drainage waters has been known for many years, but these have not been generally regarded as significant pools of N in agricultural soils. We review the size and function of both soluble organic N extracted from soils (SON) and dissolved organic N present in soil solution and drainage waters (DON) in arable agricultural soils. SON is of the same order of magnitude as mineral N and of equal size in many cases; 20–30 kg SON-N ha–1 is present in a wide range of arable agricultural soils from England. Its dynamics are affected by mineralisation, immobilisation, leaching and plant uptake in the same way as those of mineral N, but its pool size is more constant than that of mineral N. DON can be sampled from soil solution using suction cups and collected in drainage waters. Significant amounts of DON are leached, but this comprises only about one-tenth of the SON extracted from the same soil. Leached DON may take with it nutrients, chelated or complexed metals and pesticides. SON/DON is clearly an important pool in N transformations and plant uptake, but there are still many gaps in our understanding.

    Notes: Previous studies with purified variants of the 434 repressor having different operator-binding specificities have demonstrated the interactions of a heterodimeric repressor with a hybrid operator site. The present study investigates the interactions between the 434 repressor and its operator site. The optimum 434 operator half-site is used with a P22 operator half-site to create a hybrid 434/P22 operator. We show that this hybrid operator can be efficiently bound by a heterodimeric repressor, consisting of one wild-type 434 repressor monomer and one 434 repressor monomer with the binding specificity of the P22 repressor, to bring about repression in Escherichia coli.

    Notes: Fatty acid and amino acid profiles were determined in three tissues (eggs, muscle and midgut glands) from freshwater prawns, Macrobrachium rosenbergii (De Man), raised unfed in ponds with no organic fertilization, unfed in ponds with organic fertilization or fed a formulated, pelleted diet. Amino acid profiles of tail muscle and eggs were not treatment-dependent. Comparison of amino acid profiles of the diet with those of tissues of animals fed that diet suggest that dietary levels of arginine, histidine, methionine, and especially lysine may be suboptimal. Fatty acid profiles of the midgut gland, tail muscle, and to a lesser extent, eggs reflected the fatty acid composition of the diet in the fed treatment, the organic fertilizer material in the fertilized treatment and natural food organisms in the unfed treatment. Direct consumption of organic fertilizer particles by the prawns is supported by these data. Selective retention of essential fatty acids appears likely in all three treatments. Comparison of the fatty acid composition of prawn eggs and muscle tissue with that of the pelleted diet indicates that 18: 3(n-3), 20: 5(n-3), and especially 20: 4(n-6) are probably below optimal levels. However, under practical culture conditions, prawn growth may not be significantly reduced because of relatively low dietary requirements and contributions from natural foods in the ponds.
