    Description: This eBook provides a compendium of the current state-of-the-art in research tools for, and understanding of, the critical research areas in epithelial ovarian cancer (EOC) with a strong emphasis on (HG-SOC). Research areas covered include therapy response and development, microenvironmental influences and the etiology and progression of EOC. Ten articles detail established and novel in vivo and in vitro model systems. These include primary and immortalized cell culture in 2D and 3D as well as genetically engineered, transgenic, spontaneous, syngeneic, classical xenograft and patient derived xenograft mouse models. The generation of genetically engineered mouse models of HG-SOC has been a major dilemma as models with the oncogenic aberrations common in the human malignancy do not accurately recapitulate HG-SOC. Conversely, commonly used HG-SOC cell lines have been found to not harbor the expected genetic changes. These issues as well as the rapid acceptance of patient derived xenograft models are reviewed. Five articles discuss different aspects of the tumor microenvironment including its role in therapy resistance, disease progression and metastasis. Mutation of BRCA1/2 continues to be the best defined risk factor for HG-SOC. Three articles discuss BRCA-loss in the context of disease development, targeted therapies and changes in preventative measures proposed for mutation carriers in light of the recent advances in knowledge regarding the origins of this malignancy. An image of HG-SOC with patchy BRCA1 expression is featured on the cover (image by VM Howell). A major clinical issue for patients with HG-SOC is the development of therapy resistance. Five articles focus on therapy resistance and different ways to overcome resistance. Overall, this eBook is an outstanding resource to aid researchers design their programs of research and determine the most appropriate and up-to-date EOC model systems to address their research questions.

    Notes: A feeding experiment was conducted in floating cages (1.5 ± 1.0 ± 2.0 m) to determine the requirement of dietary phosphorus by juvenile orange-spotted grouper Epinephelus coicodes. Five isonitrogenous (approximately 54% CP) fishmeal-casein-based diets that contained 0.68, 0.77, 0.86, 1.02 and 1.21% total phosphorus from ingredients and supplemental monocalcium phosphate (MCP: 0, 0.75,1.51,2.26,3.02%) were fed to the fish previously fed a basal diet (0.68% total phosphorus) for 2 wk. Fish were fed in triplicate cages with one of the diets twice daily to satiation. After 8 wk of feeding, survival ranged from 70.0% to 81.7% with no significant difference among dietary treatments. The relative weight gain, specific growth rate, muscle lipid, ash, calcium, iron, zinc and phosphorus, bone ash, phosphorus, iron, and zinc were significantly affected by dietary phosphorus level. There were no significant effects of increasing total dietary phosphorus on hepatosomatic index, viscerasomatic index, condition factor, muscle moisture and protein, muscle magnesium, and manganese; and bone calcium, magnesium and manganese. Based on relative weight gain, specific growth rate and bone phosphorus, a dietary level of 0.86% total phosphorus is recommended for juvenile grouper growing in floating cages.

    Notes: A growth trial was conducted to feed juvenile tilapia (initial weight, 9.1±0.1 g), Oreochromis niloticus×O. aureus, isonitrogenous diets for 8 weeks. Six diets were formulated containing 29% crude protein from casein and gelatin, 10% crude fat from soybean oil and refined soybean lecithin and varying levels of corn starch ranging from 6% to 46% at increments of 8%, with corresponding energy to protein (E/P) ratios of 35.6, 37.9, 40.2, 42.5, 44.8 and 47.1 kJ g−1. Weight gain (WG), specific growth rate, feed efficiency ratio and protein efficiency ratio were significantly higher in fish fed diets with starch ≥22% (or E/P ratio ≥40.2 kJ g−1) than in fish fed diets with 6% or 14% starch (or E/P ratio of 35.6 or 37.9 kJ g−1). No further improvement was measured when dietary starch content increased beyond 22%. Body protein retention showed the same general pattern as WG, and was highest in fish fed the 22% starch diet. Body composition was significantly affected by dietary starch level. Fish fed diets with starch ≥30% had significantly higher lipid content than fish fed diets with 6% or 14% starch. Ash content was negatively correlated with starch inclusion level, but moisture and protein contents did not show discernible trends among treatments. Results indicate that hybrid tilapia can utilize 46% dietary starch without growth retardation, while 22% starch in feed for juvenile tilapia containing 29% protein and 10% lipid, or an E/P ratio of 37.9 kJ g−1 is optimal.

    Notes: Abscisic acid (ABA) is a sesquiterpene compound (C15) derived from C40 carotenoids. The immediate carotenoid precursors for ABA biosynthesis, 9-cis-violaxanthin and 9′-cis-neoxanthin, are produced from β-carotene by a series of hydroxylation, epoxidation, and isomerization reactions. Carotenoid hydroxylase deficient mutants contain severely reduced levels of violaxanthin and neoxanthin ( 〈 20% of wild type level) and provide a unique system to correlate carotenoid substrate availability and ABA production in photosynthetic tissues under non-stressed conditions. Quantitative measurements indicated that ABA levels in the carotenoid hydroxylase mutants are reduced nearly 50% compared to the wild type plants under non-stressed conditions. When drought-stressed, wild type plants showed up to a 17-fold increase in ABA levels, while ABA levels in the carotenoid hydroxylase mutants were only increased 6- to 7-fold (25% of wild type drought-stressed ABA levels). Expression of AtNCED3 (Arabidopsis thaliana nine-cis-epoxycarotenoid dioxygenase 3, the rate-limiting activity for ABA biosynthesis) was induced in the carotenoid hydroxylase mutants, but to a lesser extent than the 40-fold increase in wild type plants. Therefore, the reduced ABA accumulation in response to drought-stress is at least partially due to the attenuated increase in AtNCED3 gene expression in the carotenoid hydroxylase mutants. The remaining violaxanthin and neoxanthin in the carotenoid hydroxylase mutants can not be converted into ABA, indicating that there is probably a separate pool of violaxanthin and neoxanthin that is not accessible to the cleavage enzymes, because it is sequestered in the light-harvesting complexes.

    Notes: Abstract  A semi-automated continuous flow hydride generation system with inductively coupled plasma atomic emission spectrometry (ICP-AES) was used for the determination of tin in marine materials. The effects of acids (H2SO4 and HCl) were studied. The analytical parameters were thoroughly investigated. Under optimized conditions, the detection limit is 0.4 ng/ml. Interferences from transition elements were investigated and seven masking reagents were tested. L-cysteine hydrochloride monohydrate (1%) was used to mask the interferences from foreign ions. Finally, the accuracy, checked with a marine standard reference material obtained from the National Research Council (NRC), was within the certified value.

    Notes: Abstract A hydride generation system using a small concentric hydride generator combined with inductively coupled plasma atomic emission spectrometry (ICP-AES) was established to determine tin, arsenic, bismuth and antimony in a marine sediment material with L-cysteine as a pre-reductant. Influences of concentrations of three kinds of acids (HCl, HNO3 and HClO4), L-cysteine, and sodium tetrahydroborate(III) as well as sodium hydroxide were investigated. The interferences from transition ions were found to be insignificant for determination of the four elements in presence of L-cysteine. Under optimized conditions the detection limits were 0.6 ng/mL for arsenic(III), 0.8 ng/mL for antimony(III), 1.7 ng/mL for tin(IV), and 1.2 ng/mL for bismuth(III). The method was applied to determine the four elements in standard marine sediment materials and the results were in agreement with certified values.

    Notes: TbDyFe is a rare earth-iron magnetostrictive alloy with “giant” magnetostrain, good magnetomechanical coupling factor and fast response. Giant magnetostrictive actuators (GMAs) are designed and fabricated with home-made TbDyFe rods. Their magnetostrain properties under varied operation are tested. The static output displacement up to 100μm and output force up to 1500N were obtained. The dynamic displacement increases with amplitude under fixed frequency and decreases with frequency under fixed amplitude generally. The maximum dynamic output displacement of 146µm was obtained at natural frequency around 5Hz. Active vibration control employing GMA was implemented in the flexible structure. The excellent damping effect, 20-30 dB under the frequency range from 10Hz to 100Hz was obtained. The dynamic phase delay of GMA has been analyzed. Anovel improved FSLMS algorithm is proposed to achieve a better control performance
