    Description: Eyeblink classical conditioning (EBC) is a model paradigm for associative (also termed Pavlovian) learning, one of the simplest and best understood forms of learning and memory. Because EBC paradigms are readily adapted across species, the neural substrates of EBC have been well characterized, and include but are not limited to the cerebellum and anterior interpositus nucleus, the hippocampus, and prefrontal cortices. The ability to collect EBC data across many different species (i.e. including but not limited to humans) also has the distinct advantage of facilitating translational research, and therefore may be of particular benefit to elucidate mechanistic changes associated with a wide variety of psychiatric disorders. In fact, EBC paradigms have been employed to assess individuals with a wide range of neurological deficits (including Korsakoff’s amnesia, Alzheimer's disease as well as normal aging, dyslexia, inflammatory tremor, dystonia, and multiple sclerosis) and psychiatric disorders (including major depressive disorder, anxiety disorders, schizophrenia, autism, and alcohol use/addiction disorders). Individuals with these disorders exhibit differential impairments across different EBC task types (e.g., delay vs. trace EBC), with some showing impairment in one but not the other task and some showing impairments in both; across learning stage (e.g., acquisition, discrimination, or extinction), and across response variables (e.g., magnitude and timing of the conditioned eyeblink motor response, modality of the conditioned stimulus). Evaluating specific individual differences in the context of variable brain pathology should aid characterization and refinement of our understanding of complex neuropsychiatric disorders. The field of psychiatry has seen a transition from more traditional use of symptom clusters to define psychiatric disorders with subsequent examination of associated behaviors and traits, to the use of physiological and behavioral indicators to characterize individuals with respect to various psychological domains [in line with the National Institute of Mental Health Research Domain Criteria (RDoC) initiative]. This approach employs a neuroscience-based framework to assess the pathophysiology of chronic mental illnesses. Behavioral and cognitive processes are critical domains of interest in evaluating potential maladaptive patterns that may be indicative of specific psychopathologies. Furthermore, the rapid development of technological advances that allow for more detailed examination (e.g., EEG, MEG, MRI, fMRI, infrared imaging) and manipulation (e.g. transcranial magnetic and direct current stimulation) of brain functions should enhance our ability to better characterize EBC performance and its utility in characterizing aspects of particular neuropathologies. Substantial research evidence exists for the value of EBC paradigms to inform our understanding of the pathophysiologies underlying a wide variety of neurological and psychiatric disorders. Despite these findings, this readily implemented classic cognitive-behavioral paradigm is relatively underutilized in clinical settings. This e-book highlights recent convergence of clinical and research efforts in this area and aims to promote a resurgent interest in eyeblink classical conditioning, and to emphasize the potential for future translational and diagnostic applications of EBC in combination with other techniques to strengthen our understanding of alterations in brain function manifested in behaviors characteristic of specific psychopathologies.

    Description: We present the results of ammonia observations towards 66 massive star forming regions identified by the Red Midcourse Space Experiment Source survey. We have used the Green Bank Telescope and the K-Band Focal Plane Array to map the ammonia (NH 3 ) (1,1) and (2,2) inversion emission at a resolution of 30 arcsec in 8 arcmin regions towards the positions of embedded massive star formation. We have identified a total of 115 distinct clumps, approximately two-thirds of which are associated with an embedded massive young stellar object or compact H ii region, while the others are classified as quiescent. There is a strong spatial correlation between the peak NH 3 emission and the presence of embedded objects. We derive the spatial distribution of the kinetic gas temperatures, line widths, and NH 3 column densities from these maps, and by combining these data with dust emission maps we estimate clump masses, H 2 column densities and ammonia abundances. The clumps have typical masses of ~1000 M and radii ~0.5 pc, line widths of ~2 km s –1 and kinetic temperatures of ~16–20 K. We find no significant difference between the sizes and masses of the star-forming and quiescent subsamples; however, the distribution maps reveal the presence of temperature and line width gradients peaking towards the centre for the star-forming clumps while the quiescent clumps show relatively uniform temperatures and line widths throughout. Virial analysis suggests that the vast majority of clumps are gravitationally bound and are likely to be in a state of global free fall in the absence of strong magnetic fields. The similarities between the properties of the two subsamples suggest that the quiescent clumps are also likely to form massive stars in the future, and therefore provide an excellent opportunity to study the initial conditions of massive pre-stellar and protostellar clumps.

    Description: We present the chemical abundance measurements of the first large, medium-resolution, uniformly selected damped Lyman α system (DLA) survey. The sample contains 99 DLAs towards 89 quasars selected from the SDSS DR5 DLA sample in a uniform way. We analyse the metallicities and kinematic diagnostics, including the velocity width of 90 per cent of the optical depth, v 90 , and the equivalent widths of the Si ii 1526 ( W 1526 ), C iv 1548 and Mg ii 2796 transitions. To avoid strong line-saturation effects on the metallicities measured in medium-resolution spectra (FWHM ~ 71 km s –1 ), we derived metallicities from metal transitions which absorbed at most 35 per cent of the quasar continuum flux. We find the evolution in cosmic mean metallicity of the sample, 〈 Z 〉 = (–0.04 ± 0.13) z  – (1.06 ± 0.36), consistent with no evolution over the redshift range z ~ [2.2, 4.4], but note that the majority of our sample falls at z ~ [2.2, 3.5]. The apparent lack of metallicity evolution with redshift is also seen in a lack of evolution in the median v 90 and W 1526 values. While this result may seem to conflict with other large surveys that have detected significant metallicity evolution, such as Rafelski et al. who found 〈 Z 〉 = (–0.22 ± 0.03) z  – (0.65 ± 0.09) over z ~ [0, 5], several tests show that these surveys are not inconsistent with our new result. However, over the smaller redshift range covered by our uniformly selected sample, the true evolution of the cosmic mean metallicity in DLAs may be somewhat flatter than the Rafelski et al. estimate.

    Description: The defining event in apoptosis is mitochondrial outer membrane permeabilization (MOMP), allowing apoptogen release. In contrast, the triggering event in primary necrosis is early opening of the inner membrane mitochondrial permeability transition pore (mPTP), precipitating mitochondrial dysfunction and cessation of ATP synthesis. Bcl-2 proteins Bax and Bak are the principal activators of MOMP and apoptosis. Unexpectedly, we find that deletion of Bax and Bak dramatically reduces necrotic injury during myocardial infarction in vivo. Triple knockout mice lacking Bax/Bak and cyclophilin D, a key regulator of necrosis, fail to show further reduction in infarct size over those deficient in Bax/Bak. Absence of Bax/Bak renders cells resistant to mPTP opening and necrosis, effects confirmed in isolated mitochondria. Reconstitution of these cells or mitochondria with wild-type Bax, or an oligomerization-deficient mutant that cannot support MOMP and apoptosis, restores mPTP opening and necrosis, implicating distinct mechanisms for Bax-regulated necrosis and apoptosis. Both forms of Bax restore mitochondrial fusion in Bax/Bak-null cells, which otherwise exhibit fragmented mitochondria. Cells lacking mitofusin 2 (Mfn2), which exhibit similar fusion defects, are protected to the same extent as Bax/Bak-null cells. Conversely, restoration of fused mitochondria through inhibition of fission potentiates mPTP opening in the absence of Bax/Bak or Mfn2, indicating that the fused state itself is critical. These data demonstrate that Bax-driven fusion lowers the threshold for mPTP opening and necrosis. Thus, Bax and Bak play wider roles in cell death than previously appreciated and may be optimal therapeutic targets for diseases that involve both forms of cell death.
