    Notes: Abstract. The presence and localization of voltage-gated Ca2+ channels of L-type were investigated in intestinal cells of the Atlantic cod. Enterocytes were loaded with the fluorescent Ca2+ probe, fure-2/AM and changes in intracellular Ca2+ concentrations ([Ca2+] i ) were measured, in cell suspensions, in the presence of high potassium levels (100 mm), BAY K-8644 (5 μm), nifedipine (5 μm) or ω-conotoxin (1 μm). L-type Ca2+ channels were visualized on intestinal sections using the fluorescent dihydropyridine (-)-STBodipy. Depolarization of the plasma membrane produced a rapid (within 5 sec) and transient (at basal levels after 21 sec) increase in [Ca2+] i . BAY K-8644 increased the [Ca2+] i by 7.2%. Cells in a Ca2+-free buffer increased [Ca2+] i after addition of 10 mm Ca2+, and this increase was abolished by nifedipine in both depolarizing and normal medium but not by ω-conotoxin. Single cell experiments using video microscopy revealed that enterocytes remained polarized several hours after preparation and that the Ca2+ entry and extrusion occurred at specific and different regions of the enterocyte outer membrane. Fluorescent staining of L-type Ca2+ channels in the intestinal mucosa showed the most intense staining at the brushborder membrane. These results demonstrate the presence of voltage gated L-type Ca2+ channels in enterocytes from the Atlantic cod. The channels are mainly located at the apical side of the cells, and there is a polarized uptake of Ca2+ into the enterocytes. This suggests that the L-type Ca2+ channels are involved in the transcellular Ca2+ entry into the enterocytes.

    Notes: Changes were measured in some of the major physiological variables associated with seawater adaptability, growth and energetics in wild Atlantic salmon Salmo salar smolts and post-smolts migrating from the river and through the estuary, fjord and coastal areas in the River Orkla and the Trondheimsfjord, Norway during late May to early June. Gill Na+,K+-ATPase activity increased to levels of 12–16 µmol ADP mg protein−1 h−1 in post-smolts caught in higher salinity zones, probably representing long-term levels of Atlantic salmon post-smolts in oceanic conditions. Muscle moisture was regulated within narrow limits (77·7–78·7%) in fish from all zones during both years, suggesting that post-smolts adapt to marine conditions without any long-term disturbance of hydro-mineral balance. Lipid and glycogen content showed a general trend towards depletion from the river, through the fjord and into the ocean. There was, however, no significant change in protein content. The present results confirm that smolts are naturally ‘energy deficient’ during downstream migration, and suggest that post-smolts also mobilize energy reserves during their early marine phase, while protein is allocated for somatic growth. Plasma growth hormone (GH) levels increased transiently during passage through the estuary and fjord, with lower levels observed in post-smolts caught off-shore, i.e. in fish which were feeding on marine prey and had adapted to the marine environment. These physiological changes may confer substantial selective advantages during the critical early marine phase of anadromous salmonids, and hence are adaptive for long-term survival in sea water.

    Notes: As Atlantic salmon return from the ocean to undertake the anadromous spawning migration up the river of origin, profound changes in calcium metabolism and osmoregulation take place. Using tartrate resistant acid phosphatase as a marker, scale osteoclast activity was found to increase throughout sexual maturation and spawning migration. Thus, the participation of osteoclasts in the elevated scale resorption observed during this phase is established. As calcium was simultaneously accumulated in the female gonads, it is proposed that the scales are resorbed in order to provide calcium for the growing ovaries. Plasma oestradiol-17β levels were elevated in females during sexual maturation, and had decreased at the time of spawning. Plasma testosterone levels were similar in males and females during the first part of the upriver migration, but had increased in males and decreased in females at spawning. In addition to the role of these sex steroids in the gonadal growth, their possible involvement in the increased scale resorption during this phase is discussed. Plasma growth hormone and thyroxine levels were elevated in both sexes at spawning, with the triiodothyronine/thyroxine (T3/T4) ratio declining sharply, indicating possible roles for these hormones in the maturational process. The relatively low gill Na+, K+-ATPase activity of salmon caught in the estuary implies that the fish had already adapted to a hypoosmotic environment. During the upriver migration, the gill Na+, K+-ATPase activity decreased further, indicating that the hypoosmoregulatory ability was suppressed further during sexual maturation and spawning migration.

    Notes: Abstract Summer flounder (Paralichthys dentatus) that survived flounder infectious necrotizing enteritis (FINE) provided an unusual model for testing the role of the intestine in maintaining salt and water balance. The survivors lost 67% of the posterior-most intestine. The remaining stump of the anterior intestine healed and became a blind pouch. We sampled the fish three months after the epizootic ended, demonstrating that these fish survive with only a portion of their intestine. We hypothesized that salt and water balance would be disturbed in the survivors of FINE. However, plasma osmolality and concentrations of Na+, Cl−, and K+ were the same in intact flounder and those with only an intestinal stump. No compensatory changes in gill Na+, K+-ATPase activity were detected. The fish with intestinal stumps continued to feed, but their body weight was only 41% that of intact fish. Summer flounder can maintain salt and water homeostasis, but fail to thrive, using only one-third of their intestine.

    Description: 40 Ar/ 39 Ar ages from detrital muscovites have been analyzed from six modern rivers in central and western Nepal; the size of the drainage basins associated with these samples ranges from a few square kilometers to 〉40,000 km 2 . These data, when combined with previously published ages of detrital muscovites from other modern rivers in the region, suggest that a good correspondence between normalized age and normalized topography (the comparison of t * and z *) is rare, due to either nonuniform rates of passage through the ~400 °C isotherm or subsequent faulting in the drainage area. The closure temperature of Ar in muscovite is perhaps too high to make meaningful comparisons to modern topography in tectonic analysis of active orogens. The distribution of 40 Ar/ 39 Ar ages from detrital muscovites from the Karnali basin in western Nepal is much older than that for the Narayani basin in central Nepal. The Karnali muscovites, when combined with previously published muscovites from the Siwalik Group in western Nepal and zircon fission track ages from modern and ancient samples from the region, suggest a thermal history for western Nepal consistent with vigorous tectonics (and attendant erosion) before the middle Miocene but a significant diminution in the rate of erosion since ca. 10 Ma. 40 Ar/ 39 Ar ages of detrital muscovites from the Narayani basin in central Nepal suggest a markedly different history with an acceleration of the rate of erosion since ca. 10 Ma and reactivation of major faults; this is consistent with the abundant bedrock data from the Narayani basin. The strong difference in the erosional history of the adjacent Karnali and Narayani basins, as evidenced by the 40 Ar/ 39 Ar ages from detrital muscovites, is not likely to have been due to variations in climate, but rather due to strain partitioning within the Himalaya during and after the Miocene.

    Description: The increase in detrital geochronological data presents challenges to existing approaches to data visualization and comparison, and highlights the need for quantitative techniques able to evaluate and compare multiple large data sets. We test five metrics commonly used as quantitative descriptors of sample similarity in detrital geochronology: the Kolmogorov-Smirnov (K-S) and Kuiper tests, as well as Cross-correlation, Likeness, and Similarity coefficients of probability density plots (PDPs), kernel density estimates (KDEs), and locally adaptive, variable-bandwidth KDEs (LA-KDEs). We assess these metrics by applying them to 20 large synthetic data sets and one large empirical data set, and evaluate their utility in terms of sample similarity based on the following three criteria. (1) Similarity of samples from the same population should systematically increase with increasing sample size. (2) Metrics should maximize sensitivity by using the full range of possible coefficients. (3) Metrics should minimize artifacts resulting from sample-specific complexity. K-S and Kuiper test p-values passed only one criterion, indicating that they are poorly suited as quantitative descriptors of sample similarity. Likeness and Similarity coefficients of PDPs, as well as K-S and Kuiper test D and V values, performed better by passing two of the criteria. Cross-correlation of PDPs passed all three criteria. All coefficients calculated from KDEs and LA-KDEs failed at least two of the criteria. As hypothesis tests of derivation from a common source, individual K-S and Kuiper p-values too frequently reject the null hypothesis that samples come from a common source when they are identical. However, mean p-values calculated by repeated subsampling and comparison (minimum of 4 trials) consistently yield a binary discrimination of identical versus different source populations. Cross-correlation and Likeness of PDPs and Cross-correlation of KDEs yield the widest divergence in coefficients and thus a consistent discrimination between identical and different source populations, with Cross-correlation of PDPs requiring the smallest sample size. In light of this, we recommend acquisition of large detrital geochronology data sets for quantitative comparison. We also recommend repeated subsampling of detrital geochronology data sets and calculation of the mean and standard deviation of the comparison metric in order to capture the variability inherent in sampling a multimodal population. These statistical tools are implemented using DZstats, a MATLAB-based code that can be accessed via an executable file graphical user interface. It implements all of the statistical tests discussed in this paper, and exports the results both as spreadsheets and as graphic files.
