    Notes: We introduce a reaction–diffusion system capable of modeling ligand migration inside of proteins as well as conformational fluctuations of proteins, and present a detailed analytical and numerical analysis of this system in general dimension. The main observable, the probability of finding the system in the starting state, exhibits dimension-dependent as well as dimension-independent properties, allowing for sharp experimental tests of the effective dimension of the process in question. We discuss the application of this theory to ligand migration in myoglobin and to the description of gating fluctuations of ion channel proteins. © 1996 American Institute of Physics.

    Notes: Delivery of phosphorodiamidate morpholino oligomers (PMO) into fish cells in vitro and tissues in vivo was examined. Uptake was evaluated by fluorescence microscopy and flow cytometry after treating cultured cells or live rainbow trout with 3′ fluorescein-tagged PMO. Arginine-rich peptide conjugated to the 5′ end of the PMO markedly enhanced cellular uptake in culture by 8- to 20-fold compared with non-peptide-conjugated PMO as determined by flow cytometry. Enhanced uptake of PMO conjugated to peptide was also observed in tissues of fish treated by immersion. The efficacy of PMO as inhibitors of infectious haematopoietic necrosis virus (IHNV) replication was determined in vitro. Peptide-conjugated PMOs targeting sequences within the IHNV genomic RNA (negative polarity) or antigenomic RNA (positive polarity) significantly inhibited replication in a dose-dependent and sequence-specific manner. A PMO complementary to sequence near the 5′ end of IHNV genomic RNA was the most effective, diminishing titre by 97%, as measured by plaque assay and Western blot. These data demonstrate that replication of a negative-stranded non-segmented RNA virus can be inhibited by antisense compounds that target positive polarity viral RNA, or by a compound that targets negative polarity viral RNA.

    Notes: Reactive Q switching of a CO2 laser by means of a piezoelectrically actuated end reflector is reported. The high-frequency response of the piezoelectric transducer was maintained by optically polishing the transducer's surface directly instead of employing a separate mirror. Submicrosecond pulses at rates in excess of 100 kHz were obtained. This technique is especially appropriate for generating high pulse rates in low-power lasers, as no lossy intracavity elements are required.

    Notes: A new wide-annulus Couette-type viscometer/rheometer has been designed, fabricated, and tested with the objective of measuring the shear flow properties of silicate melts and magmatic suspensions containing vapor bubbles and/or solid particles at 10−4 GPa and temperatures from 600 to 1500 °C. The goal of this development has been to advance the precision of high-temperature multiphase silicate rheometry in the viscosity regime of 104–1010 Pa s, and to attempt experiments on multiphase magmatic mixtures that have heretofore been difficult to perform in the concentric cylinder geometry. With this instrument, fluids with viscosities in the design viscosity range may be tested at shear rates from 10−5 to 1 s−1, employing a series of interchangeable torque transducers capable of measurements from 0.003 to 3 N m, preserving precision of the stress measurement across the entire dynamic span. A microprocessor-controlled motor drive enables precisely specified rotational motions (position and/or speed of the sample container) for accurate constant-shear-rate or finite-strain experiments. Use of high-purity cast ceramic components rather than platinum alloy for the rotating sample container (cup) and fixed cylindrical bob permit sectioning of the sample following an experiment to inspect and analyze textural characteristics of the magmatic suspensions. Computerized data acquisition is accomplished via a standard microcomputer interface and commercially available software system that allows data to be recorded, visualized, and statistically modeled as an experiment progresses. The instrument has been tested using NIST (NBS) reference material 717 (a borosilicate glass) at viscosities from 104 to 109 Pa s. Value of the standard viscosity has been recovered within the published precision (0.03 log units) at temperatures between 650 and 850 °C. © 1998 American Institute of Physics.
