    Notes: Abstract In response to a mandate in Section 118(c)(3) of the Water Quality Act of 1987, a program called Assessment and Remediation of Contaminated Sediments (ARCS) was established. Four technical work groups were formed. This paper details the research strategy of the Toxicity-Chemistry Work Group. The Work Group's general objectives are to develop survey methods and to map the degree of contamination and toxicity in bottom sediments at three study areas, which will serve as guidance for future surveys at other locations. A related objective is to use the data base that will be generated to calculate sediment quality concentrations by several methods. The information needed to achieve these goals will be collected in a series of field surveys at three areas: Saginaw Bay (MI), Grand Calumet River (IN), and Buffalo River (NY). Assessments of the extent of contamination and potential adverse effects of contaminants in sediment at each of these locations will be conducted by collecting samples for physical characterization, toxicity testing, mutagenicity testing, chemical analyses, and fish bioaccumulation assays. Fish populations will be assessed for tumors and external abnormalities, and benthic community structure will be analyzed. A mapping approach will use low-cost indicator parameters at a large number of stations, and will extrapolate by correlation from traditional chemical and biological studies at a smaller number of locations. Sediment toxicity testing includes elutriate, pore water and whole sediment bioassays in a three-tiered framework. In addition to the regular series of toxicity tests at primary mater stations, some stations are selected for a more extensive suite of tests.

    Description: Familial medullary thyroid cancer (MTC) and its precursor, C cell hyperplasia (CCH), is associated with germline RET mutations causing multiple endocrine neoplasia type 2. However, some rare families with apparent MTC/CCH predisposition do not have a detectable RET mutation. To identify novel MTC/CCH predisposition genes we undertook exome resequencing studies in a family with apparent predisposition to MTC/CCH and no identifiable RET mutation. We identified a novel ESR2 frameshift mutation, c.948delT, which segregated with histological diagnosis following thyroid surgery in family members and demonstrated loss of ESR2 -encoded ERβ expression in the MTC tumour. ERα and ERβ form heterodimers binding DNA at specific oestrogen-responsive elements (EREs) to regulate gene transcription. ERβ represses ERα-mediated activation of the ERE and the RET promoter contains three EREs . In vitro , we showed that ESR2 c.948delT results in unopposed ERα mediated increased cellular proliferation, activation of the ERE and increased RET expression. In vivo , immunostaining of CCH and MTC using an anti-RET antibody demonstrated increased RET expression. Together these findings identify germline ESR2 mutation as a novel cause of familial MTC/CCH and provide important insights into a novel mechanism causing increased RET expression in tumourigenesis.

    Description: Surface truth and LANDSAT measurements collected July 31, 1975, for Saginaw Bay were used to demonstrate a technique for producing a color coded water quality map. On this map, color was used as a code to quantify five discrete ranges in the following water quality parameters: (1) temperature, (2) Secchi depth, (3) chloride, (4) conductivity, (5) total Kjeldahl nitrogen, (6) total phosphorous, (7)chlorophyll a, (8) total solids and (9) suspended solids. The LANDSAT and water quality relationship was established through the use of a set of linear regression equations where the water quality parameters are the dependent variables and LANDSAT measurements are the independent variables. Although the procedure is scene and surface truth dependent, it provides both a basis for extrapolating water quality parameters from point samples to unsampled areas and a synoptic view of water mass boundaries over the 3000 sq. km bay area made from one day's ship data that is superior, in many ways, to the traditional machine contoured maps made from three day's ship data.

    Description: A technique is being developed for use of ERTS in estimating and monitoring trophic levels of inland lakes. Preliminary findings are that Michigan lakes and ponds of one acre or more are resolvable in bands 5, 6 and 7 of NASA MSS imagery under fair conditions (haze and 70% cloud cover). In processed imagery (CCT) smaller features, including water color patterns, are evident within some lakes of 40 acres or more. Image distortion of lake size, shape, orientation, etc. is minimal; discrimination of lakes and ponds from various wetlands is good. Subsequent ERTS and aircraft imagery will be correlated with detailed ground truth of water color and quality in eutrophic test lakes.

    Description: The author has identified the following significant results. ERTS-1 computer compatible tapes were used as a basis to generate land use maps in lake watersheds in southeastern Michigan. These maps, generated on a repetitive basis, provide information essential to governmental agencies concerned with planning and control of lake eutrophication. The ERTS mapping products included geometrically current land use map overlays at 1:250,000 and 1:48,000 scale and area measurement printouts. The printouts provide, within the watershed boundaries and by land use category, a quantitative measure of the amount of land, in square kilometers and acres. This quantitative measure of land use in watersheds is essential to the development and application of deterministic models, which compute nutrient flows into lakes and establish lake eutrophication rates.

    Description: The author has identified the following significant results. LANDSAT digital data and ground truth measurements for Saginaw Bay (Lake Huron), Michigan, for 31 July 1975 were correlated by stepwise linear regression and the resulting equations used to estimate invisible water quality parameters in nonsampled areas. Chloride, conductivity, total Kjeldahl nitrogen, total phosphorus, and chlorophyll a were best correlated with the ratio of LANDSAT Band 4 to Band 5. Temperature and Secchi depth correlate best with Band 5.

    Description: The author has identified the following significant results. LANDSAT was used as a basis for inventorying land use within each of the Ohio-Kentucky-Indiana regional commissions, 225 drainage areas, and nine counties. Computer tabulations were produced to obtain the area covered by each of 16 land use categories within 225 drainage areas. The 16 categories were merged into ten categories and mapped at a scale of 1 inch = 5,000 ft, with detail to 0.44 hectares for the 2,700 sq mi region. These products were produced in less than 90 days, at a cost of $20,000.

    Description: The author has identified the following significant results. LANDSAT was used as a basis for producing geometrically-corrected, color-coded imagery of turbidity and circulation patterns in Saginaw Bay, Michigan (Lake Huron). This imagery shows nine discrete categories of turbidity, as indicated by nine Secchi depths between 0.3 and 3.3 meters. The categorized imagery provided an economical basis for extrapolating water quality parameters from point samples to unsample areas. LANDSAT furnished a synoptic view of water mass boundaries that no amount of ground sampling or monitoring could provide.

Significant findings are: (1) one acre lakes and one acre islands are detectable; (2) circular lakes of 7.5 acres and greater reach full density; (3) long channels 100 ft wide are detectable; (4) orientation of lakes is independent of scan direction; (5) lake features are observable in enlargements of CCT imagery produced in the Bendix Earth Resources Data Center; and (6) a decision surface water outline map is presented that was produced from ERTS-1 CCT. A water color literature review, baseline water quality data of the test lakes, and a discussion of geometric corrections of the CCT decision water surface outline map are also presented.
