    Notes: Abstract We have previously demonstrated that apical Na+ channels in A6 renal epithelial cells are associated with spectrin-based membrane cytoskeleton proteins and that the lateral mobility of these channels, as determined by fluorescence photobleach recovery (FPR) analysis, is severely restricted by this association (Smith et al., 1991. Proc. Natl. Acad. Sci. USA 88:6971–6975). Recent data indicate that the actin component of the cytoskeleton may play a role in modulating Na+ channel activity (Cantiello et al., 1991. Am. J. Physiol. 261:C882–C888); however, it is unknown if the Na+ channel's linkage to the spectrin-based membrane cytoskeleton is also involved in regulating channel activity. In this study, we have used FPR to examine if the linkage of the Na+ channels to the membrane cytoskeleton is a site for modulation of Na+ channel activity in filter grown A6 cells by vasopressin and aldosterone. We hypothesized that if the linkage of the Na+ channels to the membrane cytoskeleton is a site for regulation of Na+ channel activity by vasopressin and aldosterone, then hormone-mediated changes in either the membrane cytoskeleton or the affinity of the Na+ channel for the membrane cytoskeleton, should be reflected in changes in the lateral mobility and/or mobile fraction of Na+ channels on the cell surface. FPR revealed that although the rates of lateral mobility were not affected, there was a twofold increase in mobility fraction (f) of apical Na+ channels in aldosterone-treated (16 hr) monolayers (f = 32.31 ± 5.42%) when compared to control (unstimulated) (f = 14.2 ± 0.77%) and vasopressin-treated (20 min) (f = 12.7 ± 2.4%) monolayers. The twofold increase in mobile fraction of Na+ channels corresponds to the average increase in Na+ transport in response to aldosterone in A6 cells. The aldosterone-induced increase in Na+ transport and mobile fraction can be inhibited by the methylation inhibitor, 3-deazaadenosine, consistent with the hypothesis that a methylation event is involved in aldosterone induced upregulation of Na+ transport. We propose that the membrane cytoskeleton is involved in the aldosterone-mediated activation of epithelial Na+ channels.

    Notes: Abstract Patch clamp methods were used to characterize sodium channels on the apical membrane of Ambystoma distal nephron. The apical membranes were exposed by everting and perfusing initial collecting tubules in vitro. In cell-attached patches, we observed channels whose mean inward unitary current averaged 0.39±0.05 pA (9 patches). The conductance of these channels was 4.3±0.2 pS. The unitary current approached zero at a pipette voltage of −92 mV. When clamped at the membrane potential the channel expressed a relatively high open probability (0.46). These characteristics, together with observation that doses of 0.5 to 2 μm amiloride reversibly inhibited the channel activity, are consistent with the presence of the high amiloride affinity, high sodium selectivity channel reported for rat cortical collecting tubule and cultured epithelial cell lines. We used antisodium channel antibodies to identify biochemically the epithelial sodium channels in the distal nephron of Ambystoma. Polyclonal antisodium channel antibodies generated against purified bovine renal, high amiloride affinity epithelial sodium channel specifically recognized 110, 57, and 55 kDa polypeptides in Ambystoma and localized the channels to the apical membrane of the distal nephron. A polyclonal antibody generated against a synthetic peptide corresponding to the C-terminus of Apx, a protein associated with the high amiloride affinity epithelial sodium channel expressed in A6 cells, specifically recognized a 170 kDa polypeptide. These data corroborate that the apically restricted sodium channels in Ambystoma are similar to the high amiloride affinity, sodium selective channels expressed in both A6 cells and the mammalian kidney.

    Notes: High-resolution vacuum ultraviolet fluorescence excitation spectra of the D(O+u) ion-pair state of I2 have been obtained using a pulsed supersonic jet. Tunable vacuum ultraviolet radiation in the region 184–200 nm was produced by Raman shifting (fifth and sixth anti-Stokes) the output from a tunable excimer pumped dye laser. Vibrational constants valid up to v'=201 in the D(O+u) state have been determined.

    Notes: We have examined the surface topography and channel connectivity of a naturally crystalline porin that is known to be functional, and whose structure has not been perturbed by detergent extraction, A three-dimensional density map, calculated from two independent tilt series of negatively stained cell envelopes, reveals three separate channels per trimer on one side (the ‘smooth’ side), and a single common opening at the other (‘rough’) side. This arrangement is consistent with the molecular structures recently determined at high resolution by X-ray crystallography for three other porins after detergent solubilization, and implies that the Bordetella pertussis porin may have the same kind of folding. Surface relief maps calculated from electron micrographs of cell envelopes contrasted by unidirectional shadowing clearly show that the side with single opening (i.e. the rough side) represents the external surface.

    Notes: We study the propagation of terahertz bandwidth electrical pulses on high critical current density c-axis oriented YBa2Cu3O7−δ (YBCO) coplanar transmission lines deposited epitaxially on the low dielectric loss substrate lanthanum aluminate (LaAlO3). The losses on the YBCO transmission line are lower than on an equivalent gold line on the same substrate at temperatures below 50 K. At higher temperatures, absorption and dispersion of the ultrashort electrical pulses are observed in reasonable agreement with Mattis–Bardeen theory.

    Notes: The realization of collector-up light-emitting complementary charge injection transistors is reported. The devices have been implemented in molecular-beam-epitaxy-grown n-InGaAs/InAlAs/p-InGaAs and n-InGaAs/InP/p-InGaAs heterostructures using a self-aligned process for the collector stripe definition. Electrons, injected over the wide-gap heterostructure barrier (InAlAs or InP) by the real-space transfer (RST) process, luminesce in the low-doped p-type InGaAs active layer. An essential feature of present devices, besides their self-aligned collector-up configuration, is a relatively heavy doping of the n-type emitter channel, with the sheet dopant concentration of 4×1012 cm−2. This ensures a higher uniformity of the electric field in the channel and provides a relief from RST instabilities at a high level of collector current (linear density ∼10 A/cm). Devices with InAlAs and InP barriers show rather different optical characteristics, mainly due to the different band lineups ΔEC/ΔEV in InGaAs/InAlAs and InGaAs/InP heterostructures, leading to different ratios between the RST current and the parasitic leakage of holes from the collector into the channel. At high RST current densities, the effective carrier temperature Te in the active collector layer, determined from the high-energy tails of the luminescence spectra, is strongly enhanced compared to the lattice temperature. This decreases the device radiative efficiency and leads to a thermionic emission of carriers out of the active layer.

    Notes: Abstract. The role of the caseinase extracellular enzyme in the pathogenesis of furunculosis was investigated. Natural and mutant strains of Aeromonas salmonicida which were unable, or limited, in their ability to produce this enzyme were studied. The virulence of the mutant strains of A. salmonicida was not significantly reduced by the lack of extracellular caseinase production. Furthermore, there was no difference in the toxicity of extracellular products produced by these mutant strains and the wild-type A. salmonicida strain studied. It is reasonable to assume from these results that the caseinase enzyme has no major role in the pathogenesis of A. salmonicida infections.
