    Notes: The rheological properties and phase orientation of liquid crystalline polymer (LCP)melts flowing in a nanochannel with different surface roughness are investigated by moleculardynamics (MD) simulations. Simulation results show the surface roughness has great impact on therheological properties and phase orientation of LCP melts in the nanochannel (cross section is12nm). As the amplitude of serrations increases, the shear viscosity increases nonlinearly and thevalue of orientational order parameter decreases. When the serration amplitude is larger than 1.1nm,a phase transition (from nematic to isotropic phase) of LCP melt happens, which makes flowing innanochannels more difficult. On the other hand, the influence of serration period on the shearviscosity and orientational order parameter are found not so obvious. Findings in this study will behelpful for injection molding plastic products with nanofeatures

    Notes: The use of the Phillips–Tikhonov regularization is proposed for numerically stabilizing the ill-conditioned plasma image reconstructions. An objective function to be minimized leads to a linear estimator of the image intensity distribution and, with the aid of the singular value decomposition, makes it possible to use the generalized cross validation for optimizing a regularization parameter. An excellent behavior of the estimator with computational facility is obtained on the Hα emission computerized tomography of a toroidal plasma.

    Notes: . The peritrichs have been recognized as a higher taxon of ciliates since 1968. However, the phylogenetic relationships among them are still unsettled, and their placement within the class Oligohymenophorea has only been supported by the analysis of the small subunit rRNA gene sequence of Opisthonecta henneguyi. DNA was isolated directly from field-sampled species for PCR, and was used to resolve relationships within the genus Epistylis and to confirm the stability of the placement of peritrichs. Small subunit rRNA gene sequences of Epistylis plicatilis, Epistylis urceolata, Epistylis chrysemydis, Epistylis hentscheli, Epistylis wenrichi, and Vorticella campanula were sequenced and analyzed using both distance-matrix and maximum-parsimony methods. In phylogenetic trees, the monophyly of both the genus Epistylis and the subclass Peritrichia was strongly supported, while V. campanula clustered with Vorticella microstoma. The topology in which E. plicatilis and E. hentscheli formed a strongly supported sister clade to E. urceolata, E. chrysemydis, and E. wenrichi was consistent with variations in the thickness of the peristomial lip. We concluded that the peristomial area, especially the peristomial lip, might be the important phylogenetic character within the genus Epistylis.

    Notes: . The phylogenetic relationships among peritrichs remain unresolved. In this study, the complete small subunit rRNA (SSrRNA) gene sequences of seven species (Epistylis galea, Campanella umbellaria, Carchesium polypinum, Zoothamnium arbuscula, Vaginicola crystallina, Ophrydium versatile, and Opercularia microdiscum) were determined. Trees were constructed using distancematrix, maximum-likelihood and maximum-parsimony methods, all of which strongly supported the monophyly of the subclass Peritrichia. Within the peritrichs, 1) E. galea grouped with Opercularia microdiscum and Campanella umbellaria but not the other Epistylis species, which indicates that the genus Epistylis might not be monophyletic; 2) the topological position of Carchesium and Campanella suggested that Carchesium should be placed in the family Zoothamniidae, or be elevated to a higher taxonomic rank, and that Campanella should be independent of the family Epistylididae, and probably be given a new rank; and 3) Opisthonecta grouped strongly with Astylozoon, which suggested that Opisthonecta species were not the ancestors of the stalked peritrichs.

    Notes: Abstract. Microsatellite DNA has been developed into one of the most popular genetic markers. We have identified and cloned microsatellite loci in the genome of a free-living protozoan Euglena gracilis FACHB-848, using the random amplified microsatellites method (RAMS). The digoxigenin-labelled oligonucleotides (CT)10 and (GT)10 served as probes to detect complementary sequences in the randomly amplified polymorphic DNA (RAPD) fingerprints produced by means of Southern blotting. Subsequently, positive RAPD fragments were cloned. From a total of 31 RAPD primer profiles, eight microsatellite loci of E. gracilis were detected and characterized. Further, six sites (i.e. EGMS1, EGMS3, EGMS4, EGMS5, EGMS6, and EGMS7) showed polymorphisms. We found a GT or CT microsatellite every 10.5 kb in the genome of E. gracilis, and similar to animal genomes, the (GT)n motif was much more abundant than the (CT)n motif. These polymorphic microsatellite DNA will serve as advantageous molecular markers for studying the genetic diversity and molecular ecology of Euglena.

    Notes: Abstract The regulatory effects of malate on chloroplast Mg2+-ATPase were investigated and the mechanism was discussed. Malate stimulated methanol-activated membrane-bound and isolated CF1 Mg2+-ATPase activity. The γ subunit of CF1 may be involved in malate regulation of the enzyme function. Modification of γ subunit at one site of the peptide by NEM may affect malate stimulation of ATPase while at another site may have no effect. The effect of malate on the Mg2+-ATPase was also controlled by the Mg2+/ATP ratio in the reaction medium. The enhancing effect of malate on Mg2+-ATPase activity depended on the presence of high concentrations of Mg2+ in the reaction mixture. Kinetic study showed that malate raised the Vmax of catalysis without affecting the Km for Mg2+ ATP. The experiments imply that the stimulation of Mg2+-ATPase by malate is probably correlated with the Pi binding site on the enzyme. The regulation of ATPase activity by malate in chloroplasts may be relevant to its function in vivo.

    Notes: Abstract Photoinactivation of Photosystem (PS) II in vivo was investigated by cumulative exposure of pea, rice and spinach leaves to light pulses of variable duration from 2 to 100 s, separated by dark intervals of 30 min. During each light pulse, photosynthetic induction occurred to an extent depending on the time of illumination, but steady-state photosynthesis had not been achieved. During photosynthetic induction, it is clearly demonstrated that reciprocity of irradiance and duration of illumination did not hold: hence the same cumulative photon exposure (mol m−2) does not necessarily give the same extent of photoinactivation of PS II. This contrasts with the situation of steady-state photosynthesis where the photoinactivation of PS II exhibited reciprocity of irradiance and duration of illumination (Park et al. (1995) Planta 196: 401–411). We suggest that, for reciprocity to hold between irradiance and duration of illumination, there must be a balance between photochemical (qP) and non-photochemical (NPQ) quenching at all irradiances. The index of susceptibility to light stress, which represents an intrinsic ability of PS II to balance photochemical and non-photochemical quenching, is defined by the quotient (1-qP)/NPQ. Although constant in steady-state photosynthesis under a wide range of irradiance (Park et al. (1995). Plant Cell Physiol 36: 1163–1169), this index of susceptibility for spinach leaves declined extremely rapidly during photosynthetic induction at a given irradiance, and, at a given cumulative photon exposure, was dependent on irradiance. During photosynthetic induction, only limited photoprotective strategies are developed: while the transthylakoid pH gradient conferred some degree of photoprotection, neither D1 protein turnover nor the xanthophyll cycle was operative. Thus, PS II is more easily photoinactivated during photosynthetic induction, a phenomenon that may have relevance for understorey leaves experiencing infrequent, short sunflecks.

    Notes: Abstract Weak red light-induced changes in chlorophyll fluorescence parameters and in the distribution of PS I and PS II in thylakoid membranes were measured in wheat leaves to investigate effective ways to alter the excitation energy distribution between the two photosystems during state transition in vivo. Both the chlorophyll fluorescence parameter Fm/Fo and F685/F735, the ratio of fluorescence yields of the two photosystems at low temperature (77 K), decreased when wheat leaves were illuminated by weak red light of 640 nm, however, Fm/Fo decreased to its minimum in a shorter time than F685/F735. When Photosystem (PS II) thylakoid (BBY) membranes from adequately dark-adapted leaves (control) and from red light-illuminated leaves were subjected to SDS-polyacrylamide gel electrophoresis under mildly denaturing conditions, PS I was almost absent in the control, but was present in the membranes from the leaves preilluminated with the weak red light. In consonance with this result, the content of Cu, measured by means of the energy dispersive X-ray microanalysis (EDX), increased in the central region, but decreased in the margin of the grana stacks from the leaves preilluminated by the red light as compared with the control. It is therefore suggested that: (i) both spillover and absorption cross-section changes are effective ways to alter the excitation energy distribution between the two photosystems during state transitions in vivo, and the change in spillover has a quicker response to the unbalanced light absorption of the two photosystems than the change in light absorption cross-section, and (ii) the migration of PS I towards the central region of grana stack during the transition to state 2 leads to the enhancement of excitation energy spillover from PS II to PS I.

    Notes: Abstract The relation between the fast phase of ms-DLE (delayed light emission measured with a phosphoroscope) and the proton released from water oxidation in spinach chloroplasts was studied in several aspects. When photophosphorylation was allowed to be coupled to the Hill reaction the intensity of the fast phase of ms-DLE of chloroplast was lowered more at 1 °C than at 25 °C, and the photophosphorylation rate within 40 ms of flashing light was higher at 1 °C than at 25 °C. Adding the ε subunit of ATP synthase to the chloroplast preparation to block the leakage of protons through ATP synthase, the intensity of the fast phase of ms-DLE was enhanced, to a larger extent at 1 °C than at 25°C. When the ms-DLE was measured under isotonic conditions, the intensity of fast phase of ms-DLE enhanced by proton released from oxidation of water was more pronounced. The above results support the suggestion that under lower temperature and isotonic conditions, the proton released from water oxidation was liable to be localized and could enhance the intensity of the fast phase of ms-DLE more effectively.
