    Notes: Summary The effect of intramuscular atropine (0.6 mg) on the absorption of orally administered lidocaine (400 mg) was investigated in four healthy volunteers and in 12 laparoscopy patients. The time taken to achieve peak plasma lidocaine concentration was significantly delayed when lidocaine and atropine were given at the same time. The mean individual peak plasma lidocaine concentrations were reduced by atropine but the differences were not statistically significant. Anaesthesia and laparoscopy caused a further inhibition of lidocaine absorption. Atropine probably delayed the absorption of lidocaine by inhibition of gastric emptying.

    Notes: Summary The systemic availability of a new antidepressant, zimelidine, and of its pharmacologically active metabolite, norzimelidine, was studied in six healthy male volunteers. Three single doses of zimelidine (25 mg and 100 mg orally and 25 mg i.v.) and two single doses of norzimelidine (25 mg orally and i. v.) were given to each volunteer allowing at least seven days between administrations. Plasma concentrations of zimelidine and norzimelidine were determined in serial blood samples by HPLC. Following oral zimelidine peak plasma concentrations of the metabolite were attained about 3 h after dosing. Oral administration of norzimelidine itself resulted in a plasma concentration profile for this compound that was similar to that observed after oral zimelidine. Utilising the plasma concentration data following intravenous infusion of each compound, the elimination half-lives for zimelidine and norzimelidine were calculated to be 5.1 h (range 4.3–6.0) and 15.5 h (range 10.6–22.9) respectively. The total body clearances of the 2 compounds were similar at 0.52 l · min−1 (range 0.26–0.70) for zimelidine and 0.56 l · min−1 (range 0.28–0.83) for norzimelidine. The substantially longer elimination half-life of norzimelidine was apparently the result of a larger volume of distribution (9.4 l · kg−1; range 7.8–11.4) for this metabolite, as compared to zimelidine (3.21 · kg−1; range 1.6–4.9). The calculated bioavailability of zimelidine was 26% (range 9.1–39) after the 25 mg oral dose, and 29% (range 14–46) after the 100 mg dose. The bioavailability of norzimelidine was 66% (range 36–91). However, oral administration of zimelidine resulted in as much or more norzimelidine reaching the systemic circulation, as the oral administration of norzimelidine itself. This is important as a large part of the activity of the drug may be due to the metabolite.

    Notes: The reproductive cycle of Coregonus lavaretus (L.) in Loch Lomond, Scotland, was investigated by monthly sampling. Spawing takes place during the first 3 weeks of January, on offshore gravel banks. Males congregate on the spawning-grounds throughout the spawning period, while ripe females cruise in unisexual shoals in deeper water. Individual females migrate to the spawning-grounds as ovulation takes place, spawn, and return to deep water. After spawning, the gonadosomatic ratio and somatic condition factor of both sexes decrease until June or July. Gonad recrudescence occurs in males between July and October, and in females between July and December. The somatic condition factor of both sexes rises from its minimum in June or July to its maximum in September. A competitive protein binding assay was used to determine cortisol levels in 0–1 ml plasma samples of individual fish. The lowest cortisol levels occurred in fish caught by seine-netting and killed immediately by anaesthesia in MS 222 (Sandoz). Seine-netting and killing by concussion induced higher cortisol levels, in the range of 3–5 ng/100 ml. Cortisol levels in fish caught by gill-netting for 18 h were 3–6 times higher than in seine-netted fish, and the method of killing was immaterial in this case. The length of time spent in the gill-net had no significant effect on cortisol level, but maintenance of the fish in aquaria for 24–80 h elicited high cortisol levels of over 50 μg/100 ml. Post-mortem delay before blood-sampling resulted in lowered cortisol levels. Seasonal variations in cortisol level were determined in gill-netted fish throughout the year, and in seine-netted fish when available. Cortisol levels were high in both sexes in September; and very high in ovulating females caught on the spawning-grounds, though not in ovulating females caught off the spawning-grounds.

    Notes: This is a study of the reproductive biology and morphology of the pituitary gland of the osteoglossid fish Scleropages formosus (Müller & Schlegel). The fish spawn in the Tasek Bera waterways of Pahang, Malaysia during the rainy season beginning in August. Young fish are incubated in the mouth of the male parent until free-swimming. Males mature in their 2+ year, when 30–45 cm in length. In mature specimens the testis is small, and little sperm is produced. Some females mature in their 2+ year, although at that time, most have only reached the stage of secondary oocyte production. The chorion and follicular granulosa of secondary oocytes are deeply invaginated into the cytoplasm. All fish spawn in their 3+ year, when 45–60 cm in length. The single ripe ovary contains 20–30 ova, each 1.9 cm in diameter. No fish of 4+ years were caught, which may be the result of mortality or emigration, or due to extraneous causes such as selectivity in the fishing techniques used.The pituitary gland accords with the generalized teleost pattern and embryonic development. There is no saccus vasculosus. Adrenocorticotropic cells predominate in the proadenohypophysis, and somatotropic and luteotropic cells occur here. The mesoadenohypophysis contains gonadotropic and somatotropic cells; the former grow large in ripe specimens, so that the mesoadenohypophysis becomes much enlarged. The metaadenohypophysis contains relatively few cells, much neurohypophyseal ramification, and is deeply penetrated by the third ventricle of the brain.

    Notes: A study of the epideictic behaviour patterns of male Xiphophorus helleri Heckel was made, and the social rank of individual fish in 12 groups of three was assessed over a ten-day period. Straight-line hierarchies were established in most groups. An anatomical study was made to locate the adrenocortical tissue, and the adrenocortical activity of each fish was assessed by measuring mean nuclear diameter of 25 adrenocortical cells. Although results were somewhat variable, the highest-ranking fish in 7 out of 12 groups showed the least adrenocortical activity. The remaining two fish showed no significant difference from each other in adrenocortical activity, though their rank was in most cases clearly distinguishable on behavioural grounds.

    Notes: The pattern of diel vertical migration of Chaoborus flavicans larvae in a shallow Scottish loch varies according to the instar and, in third and fourth instars, according to the season.The planktonic phase of the migration is not exclusively concerned with predation, the larvae feeding at least equally actively in the benthos.     Notes: Thirty-six isolates of the fasciation-inducing bacterium Rhodococcus fascians were evaluated for the presence and location of the fas-1 gene, which codes for an isopentenyl transferase, the committed step in cytokinin biosynthesis. The presence of fas-1 was determined by PCR using a set of primers to the most conserved regions of the gene and by Southern hybridization to genomic digests using the PCR fragment as a probe. Both methods revealed the presence of the gene in 18 virulent isolates and the absence of the gene in 18 avirulent isolates. Thus, there is a strong relationship between the presence of the gene and virulence of the organism. The location of fas-1 was determined by probing blots of linear and circular DNA. For most of the virulent isolates, the gene was localized to a 200 ± 10 kb linear plasmid. Three virulent isolates lacked a plasmid of this size, but contained fas-1 either on a linear plasmid of 130 kb or on a large circular plasmid.
