    Description: In this work, we present results from the largest study of measured, whole-building energy performance for commercial LEED-certified buildings, using 2016 energy use data that were obtained for 4417 commercial office buildings (114 million m2) from municipal energy benchmarking disclosures for 10 major U.S. cities. The properties included 551 buildings (31 million m2) that we identified as LEED-certified. Annual energy use and greenhouse gas (GHG) emission were compared between LEED and non-LEED offices on a city-by-city basis and in aggregate. In aggregate, LEED offices demonstrated 11% site energy savings but only 7% savings in source energy and GHG emission. LEED offices saved 26% in non-electric energy but demonstrated no significant savings in electric energy. LEED savings in GHG and source energy increased to 10% when compared with newer, non-LEED offices. We also compared the measured energy savings for individual buildings with their projected savings, as determined by LEED points awarded for energy optimization. This analysis uncovered minimal correlation, i.e., an R2 〈 1% for New Construction (NC) and Core and Shell (CS), and 8% for Existing Euildings (EB). The total measured site energy savings for LEED-NC and LEED-CS was 11% lower than projected while the total measured source energy savings for LEED-EB was 81% lower than projected. Only LEED offices certified at the gold level demonstrated statistically significant savings in source energy and greenhouse gas emissions as compared with non-LEED offices.

    Description: The Scientific Committee on Oceanographic Research (SCOR) Working Group 144 Microbial Community Responses to Ocean Deoxygenation workshop held in Vancouver, B.C on July 2014 had the primary objective of initiating a process to standardize operating procedures for compatible process rate and multi-omic (DNA, RNA, protein, and metabolite) data collection in marine oxygen minimum zones and other oxygen depleted waters. Workshop attendees participated in practical sampling and experimental activities in Saanich Inlet, British Columbia, a seasonally anoxic fjord. Experiments were designed to compare and cross-calibrate in situ versus bottle sampling methods to determine effects on microbial community structure and potential activity when using different filter combinations, filtration methods, and sample volumes. Resulting biomass was preserved for small subunit ribosomal RNA (SSU or 16S rRNA) and SSU rRNA gene (rDNA) amplicon sequencing followed by downstream statistical and visual analyses. Results from these analyses showed that significant community shifts occurred between in situ versus on ship processed samples. For example, Bacteroidetes, Alphaproteobacteria, and Opisthokonta associated with on-ship filtration onto 0.4 μm filters increased fivefold compared to on-ship in-line 0.22 μm filters or 0.4 μm filters processed and preserved in situ. In contrast, Planctomycetes associated with 0.4 μm in situ filters increased fivefold compared to on-ship filtration onto 0.4 μm filters and on-ship in-line 0.22 μm filters. In addition, candidate divisions and Chloroflexi were primarily recovered when filtered onto 0.4 μm filters in situ. Results based on rRNA:rDNA ratios for microbial indicator groups revealed previously unrecognized roles of candidate divisions, Desulfarculales, and Desulfuromandales in sulfur cycling, carbon fixation and fermentation within anoxic basin waters. Taken together, filter size and in situ versus on-ship filtration had the largest impact on recovery of microbial groups with the potential to influence downstream metabolic reconstruction and process rate measurements. These observations highlight the need for establishing standardized and reproducible techniques that facilitate cross-scale comparisons and more accurately assess in situ activities of microbial communities.

    Notes: Abstract. Receptors were identified pharmacologically in functional studies where K+ secretion was monitored as transepithelial current (I sc ). Further, receptors were identified as transcripts by cloning and sequencing of reverse-transcriptase polymerase chain reaction (RT-PCR) products. I sc under control conditions was 796 ± 15 μA/cm2 (n= 329) in gerbilline VDC and 900 ± 75 μA/cm2 (n= 6) in murine VDC. Forskolin (10−5 m) but not 1,9-dideoxy-forskolin increased I sc by a factor of 1.42 ± 0.05 (n= 7). 10−9 m Arg8-vasopressin and 10−9 m desmopressin had no significant effect in gerbilline and murine VDC. Isoproterenol, norepinephrine, epinephrine and prenalterol stimulated I sc maximally by a factor of 1.38 ± 0.04 (n= 7), 1.59 ± 0.06 (n= 6), 1.64 ± 0.03 (n= 8) and 1.37 ± 0.03 (n= 6), respectively. The EC 50 values were (1.4 ± 0.7) × 10−8 m (n= 36), (2.5 ± 1.0) × 10−8 m (n= 31), (1.7 ± 0.7) × 10−7 m (n= 36) and (5 ± 4) × 10−7 m (n= 32), respectively. Propanolol inhibited isoproterenol-induced stimulation of I sc . Atenolol, ICI118551 and CGP20712A inhibited isoproterenol-induced stimulation of I sc with a pKDB of 5.0 × 10−8 m (pK DB = 7.30 ± 0.07, n= 38), 4.4 × 10−8 m (pK DB = 7.36 ± 0.14, n= 37) and 6.8 × 10−12 m (pK DB = 11.17 ± 0.12, n= 37), respectively. RT-PCR of total RNA isolated from microdissected vestibular labyrinth tissue using specific primers revealed products of the predicted sizes for β1- and β2-adrenergic receptors but not for β3-adrenergic receptors. Sequence analysis of the amplified cDNA fragments from gerbilline tissues revealed a 96.4%, 91.5% and 89.6% identity compared to rat β1-, β2- and β3-adrenergic receptors, respectively. These results demonstrate that K+ secretion in VDC is under the control of β1- but not β2- or β3-adrenergic receptors or vasopressin-receptors.
