    Notes: Viral haemorrhagic septicaemia virus (VHSV) and infectious haematopoietic necrosis virus (IHNV) cause severe epizootics among most salmonid fish as well as a number of marine species. The role of the marine reservoir in the epidemiology is unclear. This epidemiological situation, i.e. the wide distribution among a lot of species, gives rise to the belief that it seems to be unsuccessful to control the disease by regulatory methods in regions in which the virus is enzootic, i.e. where feral fish carry the virus. Therefore, the development of a vaccine seems to be more promising. There is now one live vaccine against VHS available in Germany.For identification of attenuated viral strains as vaccines, molecular markers are needed. The G-genes of several VHSV strains, including two attenuated strains, were sequenced to define marker for specific recognition of the attenuated virus. The deduced amino acid sequences were compared to elucidate the molecular basis of attenuation. For rapid differentiation of the vaccine-strain from wild-type viruses, RT-PCR was used with primers specific for the attenuated virus. The PCR-product is a fragment of 251 bp which is used for identification of the vaccine after isolation of a virus from the field.The administration of an inactivated vaccine seems to be unsuitable for rainbow trout fry. Therefore, recent studies have focused on the use of recombinant viral proteins as candidates for vaccines, with particular attention given to the glycoprotein.Rainbow trout vaccinated by immersion with live Aeronionas salmonicida bacteria carrying VHS-G-gene and IHN-G-gene fragments were differentiated from survivors after infection with wild-type virus by western blot: sera from vaccinated fish react only with G-protein and not with other structural components of the virus.

    Description / Table of Contents: Zusammenfassung Herstellung und Anwendung gepackter Capillarsäulen (innerer Durchmesser: ≈0,3 mm) zur analytischen Trennung von Kationen durch Komplexelution im Mikromaßstab werden beschrieben. Als Säulenpackung dienen Kugeln eines stark sauren Kationenaustauschers mit einem mittleren Durchmesser von 25 μ. Die Detektion der getrennten Kationen erfolgt kontinuierlich im Effluent mit Hilfe einer amperometrischen Mikromeßzelle. Die Trennleistung einer 125 mm langen Capillarsäule wird am System Ni2+/Co2+/Iminodiessigsäure gezeigt. Die dosierten Probemengen liegen in der Größenordnung von 5–12 μg je Kation.
    Notes: Summary Preparation and application of packed capillary columns (interior diameter: ≈0.3 mm) for analytical separations of cations in micro-scale by means of complex elution are described. The capillary columns are filled with spherical grains in the range of 25 μ. The detection of separated cations in the effluent is performed by means of an amperometric micro-cell. The separation efficiency of a capillary column (length: 125 mm) is shown for the system Ni2+/Co2+/iminodiacetic acid. The injected quantities of samples are in the range of 5–12 μg per cation.

    Description / Table of Contents: Zusammenfassung Im Rahmen experimenteller Untersuchungen über das Wirt-Parasit-Verhältnis nach einmaliger Litomosoides carinii-Infektion der Mastomys natalensis wurde das Albumin-Globulin-Verhältnis im Serum über einen Zeitraum von 240 Tagen p.i. mit Hilfe der Cellulose-Azetat-Folien-Elektrophorese untersucht. Dabei zeigte sich zweimal eine sprunghafte, prozentuale Albuminverringerung bei gleichzeitigem Globulinanstieg, und zwar zwischen dem 21. und dem 28. Tag p.i. und am Ende der Präpatenz ab 56 Tage p.i. Im weiteren Infektionsverlauf war ab ca. 100 Tage p.i. trotz steigender Mikrofilarienzahl im peripheren Blut nur noch ein minimaler Globulinanstieg zu verzeichnen. Die relativen Globulinanteile im Serum stiegen von 30–40% zu Beginn der Untersuchung bis zum Ende des Untersuchungszeitraums auf 75–80% an. Eine Erhöhung des Gesamtproteingehalts im Serum konnte nicht festgestellt werden. Die Veränderungen im Serumeiweißbild in der 4. Woche p.i. werden mit der Häutung der Parasiten von der Larve III zur Larve IV und diejenigen am Ende der Präpatenz mit der beginnenden Fertilität der Filarien in Zusammenhang gebracht. Im weiteren Verlauf der L. carinii-Infektion bis ca. 240 Tage p.i. stellte sich ein Gleichgewichtszustand ein. Korrelationen zwischen den Veränderungen im Albumin-Globulin-Verhältnis und der Zahl adulter L. carinii konnten statistisch nicht gesichert werden. Hochsignifikant korreliert war dagegen die prozentuale Reduktion der Albumine am 218. und 240. Tag p.i. mit der Höhe der Mikrofilariämie zu diesen Zeitpunkten. In den früheren Phasen der Infektion ergaben sich hierbei keine gesicherten Korrelationen. Weiterhin konnte die Beobachtung gesichert werden, daß das Mikrofilarienmaximum um so später auftritt, je höher die Befallsrate mit weiblichen L. carinii ist.
    Notes: Summary The albumine/globuline ratio in the serum of Mastomys natalensis has been studied with cellulose-acetate-folia-electrophoresis over a period of 240 days after single experimental infection with Litomosoides carinii. During this period a sudden decrease of albumine combined with an increase in globuline percentage could be observed twice during the course of experimentation. It was noticed especially (1) between 21st–28th day p.i. and (2) at the end of the prepatent period i.e. 56 days p.i. In spite of the increase in the number of microfilariae in the peripheral blood, there was a minimal increase of globuline throughout the patent period (recorded from 100 days p.i.). The relative increase in serum globulines could be observed from 30–40% at the beginning to 75–80% at the end of experiment. However, no increase in the total protein contents of serum could be observed. The changes in the albumine/globuline ratio during the 4th week after infection is presumed to be related to the moulting of 3rd stage to 4th stage larvae, and that at the end of the prepatent period may be correlated with the sexual maturity of the worms. In the latter infection period up to 240 days p.i. a stage of equilibrium could be noticed with minor changes in the albumine/globuline ratio. The correlation between the changes in the albumine-globuline-relation and the number of adult Litomosoides carinii were not significant. On the other hand high significant correlation was found between the percentage reduction of albumine and the level of microfilariae in blood on 218th and 240th day of infection. In the early phase of infection, however, significant correlations could not be noticed. When more female Litomosoides carinii were present in the host, it was statistically proved that the maximum level of microfilariae was reached after prolonged period of infection.
