    Description: Non-long terminal repeat (non-LTR) retrotransposons are highly abundant elements that are present in chromosomes throughout the eukaryotic domain of life. The long interspersed nuclear element (LINE-1) (L1) clade of non-LTR retrotransposons has been particularly successful in mammals, accounting for 30–40% of human genome sequence. The current model of LINE retrotransposition, target-primed reverse transcription, culminates in a chromosomally integrated end product. Using a budding yeast model of non-LTR retrotransposition, we show that in addition to producing these ‘classical’, chromosomally integrated products, a fungal L1 clade member (Zorro3) can generate abundant, RNA-derived episomal products. Genetic evidence suggests that these products are likely to be formed via a variation of target-primed reverse transcription. These episomal products are a previously unseen alternative fate of LINE retrotransposition, and may represent an unexpected source for de novo retrotransposition.

    Description: The E3 ubiquitin ligase HUWE1/Mule/ARF-BP1 plays an important role in integrating/coordinating diverse cellular processes such as DNA damage repair and apoptosis. A previous study has shown that HUWE1 is required for the early step of DNA damage-induced apoptosis, by targeting MCL-1 for proteasomal degradation. However, HUWE1 is subsequently inactivated, promoting cell survival and the subsequent DNA damage repair process. The mechanism underlying its regulation during this process remains largely undefined. Here, we show that the Cullin4B-RING E3 ligase (CRL4B) is required for proteasomal degradation of HUWE1 in response to DNA damage. CUL4B is activated in a NEDD8-dependent manner, and ubiquitinates HUWE1 in vitro and in vivo . The depletion of CUL4B stabilizes HUWE1, which in turn accelerates the degradation of MCL-1, leading to increased induction of apoptosis. Accordingly, cells deficient in CUL4B showed increased sensitivity to DNA damage reagents. More importantly, upon CUL4B depletion, these phenotypes can be rescued through simultaneous depletion of HUWE1 , consistent with the role of CUL4B in regulating HUWE1. Collectively, these results identify CRL4B as an essential E3 ligase in targeting the proteasomal degradation of HUWE1 in response to DNA damage, and provide a potential strategy for cancer therapy by targeting HUWE1 and the CUL4B E3 ligase.

    Description: Being used in the fixed-target mode, the multi-TeV LHC proton and lead beams allow for studies of heavy-flavour hadroproduction with unprecedented precision at backward rapidities, far negative Feynman-, using conventional detection techniques. At the nominal LHC energies, quarkonia can be studied in detail in , , and collisions at  GeV and in Pb + and Pb + collisions at  GeV with luminosities roughly equivalent to that of the collider mode that is up to 20 fb−1 yr−1 in and collisions, up to 0.6 fb−1 yr−1 in collisions, and up to 10 nb−1 yr−1 in Pb + collisions. In this paper, we assess the feasibility of such studies by performing fast simulations using the performance of a LHCb-like detector.

    Description: 〈span〉〈div〉ABSTRACT〈/div〉The detailed depositional systems and basin evolution of Lower Cretaceous coal-bearing strata in the Erlian Basin of northeastern China were analyzed based on extensive borehole and outcrop data. A total of 7 facies associations are interpreted and consist of 14 distinct lithofacies, with lithologies including conglomerates, sandstones, siltstones, mudstones, shales, and coals. Five third-order sequences were recognized, and their internal lowstand, transgressive, and highstand systems tracts were defined based on six key sequence stratigraphic boundaries. These boundaries were represented by regional unconformities, basal erosional surfaces of incised valley fills, interfluvial paleosols, and abrupt depositional facies-reversal surfaces. Sequences I–V correspond to the rift-initiation stage, the early-rift climax stage, the late-rift climax stage, the immediate postrift stage, and the late postrift stage of the basin, respectively. The preferred sites for coal accumulation were braided fluvial delta plain, meandering fluvial delta plain, and littoral–shallow lake environments. The major coal seams formed during the early and late transgressive systems tract of sequences III, IV, and V, which were well developed in the eastern, northeastern, and northeastern parts of the Erlian Basin, respectively. Three coal depositional models were summarized in the sequence stratigraphic framework, including types 1, 2, and 3, corresponding to the Newark type, Newark–Richmond type, and Richmond type, respectively. These coal depositional models were closely related to the basin evolution. These results could provide preferred depositional environments and favorable areas of coal and coalbed methane (CBM) for the exploration and development of coal and CBM in the Erlian Basin, with the Jiergalangtu, Huolinhe, Baiyinhua, and A’nan sags recommended as the key sags.〈/span〉

    Description: TDP-43 aggregation in the cytoplasm or nucleus is a key feature of the pathology of amyotrophic lateral sclerosis and frontotemporal dementia and is observed in numerous other neurodegenerative diseases, including Alzheimer's disease. Despite this fact, the inciting events leading to TDP-43 aggregation remain unclear. We observed that endogenous TDP-43 undergoes reversible aggregation in the nucleus after the heat shock and that this behavior is mediated by the C-terminal prion domain. Substitution of the prion domain from TIA-1 or an authentic yeast prion domain from RNQ1 into TDP-43 can completely recapitulate heat shock-induced aggregation. TDP-43 is constitutively bound to members of the Hsp40/Hsp70 family, and we found that heat shock-induced TDP-43 aggregation is mediated by the availability of these chaperones interacting with the inherently disordered C-terminal prion domain. Finally, we observed that the aggregation of TDP-43 during heat shock led to decreased binding to hnRNPA1, and a change in TDP-43 RNA-binding partners suggesting that TDP-43 aggregation alters its function in response to misfolded protein stress. These findings indicate that TDP-43 shares properties with physiologic prions from yeast, in that self-aggregation is mediated by a Q/N-rich disordered domain, is modulated by chaperone proteins and leads to altered function of the protein. Furthermore, they indicate that TDP-43 aggregation is regulated by chaperone availability, explaining the recurrent observation of TDP-43 aggregates in degenerative diseases of both the brain and muscle where protein homeostasis is disrupted.

    Description: Vinyl carbocations have been the subject of extensive experimental and theoretical studies over the past five decades. Despite this long history in chemistry, the utility of vinyl cations in chemical synthesis has been limited, with most reactivity studies focusing on solvolysis reactions or intramolecular processes. Here we report synthetic and mechanistic studies of vinyl cations generated through silylium–weakly coordinating anion catalysis. We find that these reactive intermediates undergo mild intermolecular carbon-carbon bond–forming reactions, including carbon-hydrogen (C–H) insertion into unactivated sp 3 C–H bonds and reductive Friedel-Crafts reactions with arenes. Moreover, we conducted computational studies of these alkane C–H functionalization reactions and discovered that they proceed through nonclassical, ambimodal transition structures. This reaction manifold provides a framework for the catalytic functionalization of hydrocarbons using simple ketone derivatives.
