Zusammenfassung 14 männliche kastrierte Ratten wurden zu unterschiedlichen Zeiten nach einer einmaligen Gabe von 2, 5 μg β-Östradiol/100 g Körpergewicht getötet. 60 min vor dem Tod erhielten alle Tiere zusätzlich 200 μC 3H-Cytidin/100 g Körpergewicht ip. An PAS-gefärbten Paraffinschnitten der Niere wurde die Kornzahl über den Zellkernen der gewundenen und gestreckten Hauptstücke, bei letzteren unter besonderer Berücksichtigung des Nukleolus, autoradiographisch bestimmt. Die Ergebnisse wurden varianzanalytisch ausgewertet. Unter der Annahme, daß die Kornzahl die unterschiedliche Einbaugröße von 3H-Cytidin in der Ribonukleinsäure wiedergibt, kommen wir zu folgenden Resultaten: 1. Spätestens 3 Std nach Östradiol ist die Einbaurate in den gestreckten Hauptstücken im Vergleich zu den gewundenen Abschnitten erhöht. Die Einbaurate ist auch noch 12 Std nach Östradiol nachweisbar gesteigert, kehrt danach aber zur Norm zurück. 2. Östradiol fördert den 3H-Cytideneinbau in den Nukleolus in genau der gleichen Weise wie in das umgebende Karyoplasma. 3. Actinomycin D (70 (μg/100 g Körpergewicht) wirkt entgegengesetzt wie Östradiol. Es hemmt jedoch den 3H-Cytidineinbau in den Nukleolus ungleich stärker als in das umgebende Karyoplasma. 4. Es wird ein Zusammenhang zwischen dem 3H-Cytidineinbau und der östradiolabhängigen Steigerung der alkalischen Phosphatase der gestreckten Hauptstücke vermutet.

Summary 14 male castrated rats were sacrified at various times after a single dose of 2.5 (μg β-estradiol/100 g b. w. sc. 60 min before death all animals additionally received 200 μC 3H-Cytidine/100 g b. w. ip. The number of silver grains located above the nuclei of the convoluted and straight portion of proximal tubules of the kidney was measured autoradiographically on PAS-stained paraffin sections. Special reference was given to the relationship between the number of grains located above the nucleoli and the surrounding parts of the nucleus. If one assumption is made that the number of grains reflects the different rate of incorporation of 3H-Cytidine the following conclusions can be drawn: 1. 3 hours after estradiol administration at the latest the rate of incorporation in the straight proximal tubules is raised in comparison to the convoluted tubules. Even 12 hours after estradiol administration the rate of incorporation is significantly increased and then turns back to the normal level. 2. There is an equivalent enhancement by estradiol of incorporation of 3H-Cytidine in the nucleoli as well as in the surrounding parts of the nuclei. 3. Actinomycin D counteracts the effect of estradiol. The incorporation is far more inhibited by Actinomycin D in the nucleoli compared with the surrounding parts of the nuclei. 4. An interrelationship between the incorporation of 3H-Cytidine and the estradiol dependent increase of alkaline phosphatase in the straight proximal tubules is suggested.
