    Notes: Summary Polyclonal antibodies against a part of pea (Pisum sativum L.) LOXG protein have been raised to study the pattern of distribution of related lipoxygenases in pea carpels. The antiserum recognized three lipoxygenase polypeptides in carpels. One of them became undetectable 24 hours after fruit development induction, suggesting that it may correspond to the protein derived from loxg cDNA. Immunolocalization experiments showed that lipoxygenase protein was present only in pod tissues: it was abundant in the mesocarp and, from the day of anthesis, in the endocarp layers. Lipoxygenase distribution is regulated throughout development. The association of lipoxygenase with cells in which processes of expansion and growth will potentially take place support a role in pod growth and development.

    Notes: Abstract. The enzyme 3-hydroxy-3-methylglutaryl coenzyme A reductase (HMGR, EC 1.1.1.34) is encoded by a small multigene family in tomato (Lycopersiconesculentum Mill.) and catalyzes the synthesis of mevalonic acid (MVA), a committed step in the biosynthesis of sterols and isoprenoids. A chimeric HMG1::GUS reporter gene fusion was used to analyze the regulation of HMG1 gene expression in detail. HMG1 promoter 5′ deletion mutants established the boundary of a fully inducible promoter. In HMG1::GUS transgenic tomato plants, histochemical staining with 5-bromo-3-indolyl-glucuronide demonstrated that HMG1 was primarily expressed in shoot and root meristems, and in young tomato fruit. This result was confirmed by both HMG1 in-situ hybridization and RNA gel blot analysis. Tomato suspension cell experiments showed that steady-state HMG1 mRNA accumulated during lag and exponential growth phases, but not during the stationary phase. Transient expression of the HMG1::GUS in tissue culture cells treated with mevinolin indicated that HMG1 expression was subject to feedback regulation by a biosynthetic product derived from MVA. These results suggest that a primary, although not exclusive, role of HMG1 is to supply the MVA demand associated with cell division and growth.

    Notes: Abstract A cDNA clone (loxg) corresponding to a gene repressed during carpel development has been isolated from a cDNA library of unpollinated carpels induced to grow by treatment with gibberellic acid (GA3). The sequences of loxg cDNA and the deduced polypeptide have a high similarity with legume type 2 lipoxygenases, especially with Phaseolus lox1 (78.5% similarity at the protein level) and pea and soybean lox3 (83.6% and 85.4%, respectively). loxg expression is constant in unstimulated carpels but it decreases in carpels induced to keep growing by fertilization or hormone treatment. A similar pattern of repression was observed in lipoxygenase activity of pea and tomato carpels. In situ hybridization studies showed that loxg mRNAs are present in the endocarp and the mesocarp of pea pods; no loxg expression was detectable either in the pod exocarp or in the ovules. Loxg is also expressed in other young growing tissues, especially in flower organs. Nevertheless, the natural pattern of flower and fruit development is associated with loxg repression.

    Notes: A bubble column absorber was used to investigate kinetics of the reaction between carbon dioxide and aqueous solutions of diisopropanolamine (DIPA), by means of gas absorption experiments. These were conducted in the temperature range of 20 to 40°C, with DIPA concentrations from 5 to 500 mol/m3, and CO2 partial pressures between 5 and 101 kPa. A model based on the Danckwerts' surface reneval theory was used to analyze the experimental results and to determine the rate constant.     Notes: -Reaction of 3 with (cyanomethylene)triphenylphosphorane in refluxing dichloromethane or methanol gave mixtures of 4a and 4b in ratios of about 3:1 and 1:3, respectively. The same reaction performed with the furanoid isomer 5 afforded 6a and 6b in (E)/(Z) ratios of approximately 9:1 and 1:2, respectively. Catalytic hydrogenation of the α,β-unsaturated nitriles 4 and 6 with either 10% Pd-C or Raney nickel afforded the saturated nitriles 7 and 9, or the 1-amino-1,2,3-trideoxy-4-octulose derivatives 8 and 10, respectively. In an attempt to transform 6a into the appropriate polyhydroxylated branched-chain pyrrolidine 15, (5R,8S,9R,10S)-8,9,10-trihydroxy-1-aza-6-oxaspiro[4.5]decane, which was identified as its peracylated derivative 16, was isolated. Following an alternative synthetic strategy, partial hydrolysis of compound 9 afforded 17 which was regioselectively transformed into the corresponding derivative 22 via its 8-O-p-toluenesulfonyl derivative 18. Removal of the 4,5-O-isopropylidene group in 22 with aqueous trifluroacetic acid gave the free octulose 23, which was hydrogenated in the presence of 10% Pd-C, to afford the expected (6S,7R,8R,8aR)-6,7,8-trihydroxyindolizidine (1-deoxycasta-nospermine, 11).
