    Notes: Summary Three mutations in the cystic fibrosis transmembrane conductance regulator (CFTR) gene were discovered in a pancreas-insufficient patient with cystic fibrosis (CF) who displayed an uncommon combination of almost normal chloride concentration in sweat tests and typical symptoms of gastrointestinal and pulmonary disease. The R553Q mutation was found on the maternal ΔF508-CFTR gene. Codon 553 is located within a consensus motif of the ATP-binding cassette transport proteins at a less conserved position. Other members of this protein superfamily contain a glutamine instead of arginine at the homologous position, suggesting a modulating rather than disease-causing role of the R553Q mutation in CFTR. The amplification refractory mutation system did not detect the R553Q mutation in a further 65 normal, 113 ΔF508, and 91 non-ΔF508 CF chromosomes. The index case carried the R553X nonsense mutation on the paternal chromosome. The R553X mutation was present on a further 9 out of 86 German nonΔF508 CF chromosomes linked with the XV2c-KM19Mp6d9-J44-GATT haplotypes 2-2-2-1-1 and 1-1-2-1-2. The location of R553X on separate haplotypes including both alleles of the intragenic GATT repeat suggests an ancient and/or multiple origins of the R553X mutations. The association of the genotype of the CFTR mutation and the clinical phenotype was assessed for the patients carrying the related genotypes ΔF508/ΔF508 (n = 80), ΔF508/R553X (n = 9) and ΔF508-R553Q/R553X (n = 1). In compound heterozygotes, the median chloride concentration in pilocarpine iontophoresis sweat tests was significantly lower than in the ΔF508 homozygotes (P 〈 0.01). The patient groups were significantly different with respect to the distributions of the centiles for height (P 〈 0.001) and weight (P 〈 0.01) as the most sensitive predictors of the course and prognosis in CF. Growth retardation was more pronounced in the compound heterozygotes.

    Notes: Abstract The dispersion of two immiscible fluids in a static micromixer comprising interdigital channels with corrugated walls was investigated using silicon oil dispersed in dyed as well as pure water as test systems. Silicon oil and water flow rates between 20 mL/h to 500 mL/h and 150 mL/h to 700 mL/h were used, respectively. The experiments revealed the dependence of the average droplet size and size distribution on geometrical parameters of the micromixer and operating conditions. Dispersions with average droplet sizes as small as 5.6 μm and monomodal size distributions having small standard deviations of the droplet size down to 3.6 μm could be generated using the micromixer. The droplet size decreased with increasing total flow and ratio of the flow rates of the two liquids. In addition, a decrease of the droplet size was found when decreasing the channel width of the mixing device. Generally, the silicon oil – dyed water dispersion showed smaller average droplet sizes and were more stable compared to the dispersions based on silicon oil and pure water.

    Notes: Abstract For a comprehensive study of phototropism in sporangiophores of the fungus Phycomyces, quantitative treatment of spatial aspects is necessary. The first step in quantifying spatial factors of phototropic signal processing is the elucidation of the non-uniform light profile, predominantly caused by a lens effect in the cylindrical body of the sporangiophore. Herein we compare recently presented theoretical and experimental studies of light profiles. Errors and ambiguities arising from instrumental limitations and arbitrary assumptions are revealed. On the other hand, by combining theoretical and experimental results we have been able to select out the reliable information, which can now be applied in phototropic studies.

    Notes: Abstract Many data sets used by EPA's Environmental Monitoring and Assessment Program (EMAP) will be collected and managed by groups other than EPA as the Committee on Environment and Natural Resources develops the inter-agency National Environmental Monitoring Initiative. Managing these data requires a change from a database managed solely by EPA to a model where there is, in addition to distributed databases, truly distributed ownership and responsibility. Common standards, data directories, and data descriptions allow data of interest to be located, understood, and downloaded. The level of EMAP data management practices applied to a data set is based on the degree of EMAP responsibility for the data. The EMAP Data Directory is an Oracle database that tracks data sets of interest and contains sufficient information about a data set for a user to determine if the data are of interest. Some of the data sets listed in the Directory are in the possession of EMAP and are accessible on the EMAP WWW site (http://www.epa.gov/emap). Other data sets in the Directory are managed, documented, and made available by other organizations. The Data Catalog contains metadata about data sets in the possession of EMAP and provides the user with information about methods, assumptions, and data quality.
