    Description: Whereas many genes associated with intellectual disability (ID) encode synaptic proteins, transcriptional defects leading to ID are less well understood. We studied a large, consanguineous pedigree of Arab origin with seven members affected with ID and mild dysmorphic features. Homozygosity mapping and linkage analysis identified a candidate region on chromosome 17 with a maximum multipoint logarithm of odds score of 6.01. Targeted high-throughput sequencing of the exons in the candidate region identified a homozygous 4-bp deletion (c.169_172delCACT) in the METTL23 (methyltransferase like 23) gene, which is predicted to result in a frameshift and premature truncation (p.His57Valfs*11). Overexpressed METTL23 protein localized to both nucleus and cytoplasm, and physically interacted with GABPA (GA-binding protein transcription factor, alpha subunit). GABP, of which GABPA is a component, is known to regulate the expression of genes such as THPO (thrombopoietin) and ATP5B (ATP synthase, H+ transporting, mitochondrial F1 complex, beta polypeptide) and is implicated in a wide variety of important cellular functions. Overexpression of METTL23 resulted in increased transcriptional activity at the THPO promoter, whereas knockdown of METTL23 with siRNA resulted in decreased expression of ATP5B , thus revealing the importance of METTL23 as a regulator of GABPA function. The METTL23 mutation highlights a new transcriptional pathway underlying human intellectual function.

    Description: The Bakken Formation of the Williston Basin is a prime example of an unconventionally produced petroleum system, with a low-permeability reservoir that requires application of advanced technologies for commercial production. A three-dimensional model of the Williston Basin was constructed to integrate and assess the parameters that influence the generation and migration of hydrocarbons in the Bakken Formation. This study tests the applicability of available basin and petroleum system modeling technology on an unusually low-permeability petroleum system. The model is based on nine surfaces constructed from log tops of thousands of wells in the study area and additional depth and isopach maps of the Bakken Formation members. These were integrated with the established basin evolution in line with published research. Temperature and thermal maturity were calibrated during model construction from well temperature and geochemistry data. The resulting heat-flow map supports the existence of a heat-flow anomaly along longitude 103°W, discussed controversially in the literature. Furthermore, the results indicate that the invasion-percolation migration approach best describes the distribution of petroleum accumulation and the saturated areas in the Bakken members. The volume of generated hydrocarbons was calculated, and the extent of the highly saturated accumulation beyond the area of the high-mature source was mapped. Furthermore, it was demonstrated that petroleum accumulations beyond the high-saturation zones have to be related to stratigraphic pinch-outs, lateral variability in permeability of the Bakken members, or smaller structural influences that were lost because of the resolution applied in the model.

    Description: Olfaction and taste are of critical importance to insects and other animals, since vital behaviours, including mate, food and host seeking, as well as predator and toxin avoidance, are guided by chemosensory cues. Mate and habitat choice are to a large extent determined by chemical signals, and chemoreceptors contribute accordingly to pre-mating isolation barriers and speciation. In addition to fundamental physiological, ecological and evolutionary consideration, the knowledge of insect taste and especially olfaction is also of great importance to human economies, since it facilitates a more informed approach to the management of insect pests of agricultural crops and forests, and insect vectors of disease. Chemoreceptors, which bind to external chemical signals and then transform and send the sensory information to the brain, are at the core of the peripheral olfactory and gustatory system and have thus been the focus of recent research in chemical ecology. Specifically, emphasis has been placed on functional characterization of olfactory receptor genes, which are derived from three large gene families, namely the odorant receptors, gustatory receptors and ionotropic receptors. Spatial expression patterns of olfactory receptors in diverse chemosensory tissues provide information on divergent functions, with regards to ecologically relevant behaviours. On the other hand, characterization of olfactory receptor activation profiles, or “deorphanization”, provides complimentary data on the molecular range of receptivity to the fundamental unit of the olfactory sense. The aim of this Research Topic is to give an update on the breadth and depth of research currently in progress related to understanding the molecular mechanisms of insect chemoreception, with specific emphasis on the olfactory receptors.

    Description: Abstract Submarine turbidity currents are one of the most important processes for moving sediment across our planet; they are hazardous to offshore infrastructure, deposit petroleum reservoirs worldwide, and may record tsunamigenic landslides. However, there are few studies that have monitored these submarine flows in action, and even fewer studies that have combined direct monitoring with longer‐term records from core and seismic data of deposits. This article provides one of the most complete studies yet of a turbidity current system. The aim here is to understand what controls changes in flow frequency and character along the turbidite system. The study area is a 12 km long delta‐fed fjord (Howe Sound) in British Columbia, Canada. Over 100 often powerful (up to 2 to 3 m sec−1) events occur each year in the highly‐active proximal channels, which extend for 1 to 2 km from the delta lip. About half of these events reach the lobes at the channel mouths. However, flow frequency decreases rapidly once these initially sand‐rich flows become unconfined, and only one to five flows run out across the mid‐slope each year. Many of these sand‐rich, channelized, delta‐sourced flows therefore dissipated over a few hundred metres, once unconfined, rather than eroding and igniting. Upflow migrating bedforms indicate that supercritical flow dominated in the proximal channels and lobes, and also across the unconfined mid‐slope. These supercritical flows deposited thick sand beds in proximal channels and lobes, but thinner and finer beds on the unconfined mid‐slope. The distal flat basin records far larger volume and more hazardous events that have a recurrence interval of ca 100 years. This study shows how sand‐rich delta‐fed flows dissipate rapidly once they become unconfined, that supercritical flows dominate in both confined and unconfined settings, and how a second type of more hazardous, and much less frequent event is linked to a different scale of margin failure.

    Description: Gas separation membranes are one of the lowest energy technologies available for the separation of carbon dioxide from flue gas. Key to handling the immense scale of this separation is maximised membrane permeability at sufficient selectivity for CO2 over N2. For the first time it is revealed that metals can be post-synthetically exchanged in MOFs to drastically enhance gas transport performance in membranes. Ti-exchanged UiO-66 MOFs have been found to triple the gas permeability without a loss in selectivity due to several effects that include increased affinity for CO2 and stronger interactions between the polymer matrix and the Ti-MOFs. As a result, it is also shown that MOFs optimized in previous works for batch-wise adsorption applications can be applied to membranes, which have lower demands on material quantities. These membranes exhibit exceptional CO2 permeability enhancement of as much as 153% when compared to the non-exchanged UiO-66 mixed-matrix controls, which places them well above the Robeson upper bound at just a 5 wt.% loading. The fact that maximum permeability enhancement occurs at such low loadings, significantly less than the optimum for other MMMs, is a major advantage in large-scale application due to the more attainable quantities of MOF needed.
