    Description: In plants, plant fructokinases (FRKs) are considered to be the main gateway of fructose metabolism as they can phosphorylate fructose to fructose-6-phosphate. Chinese white pears ( Pyrus bretschneideri ) are one of the popular fruits in the world market; sugar content is an important factor affecting the quality of the fruit. We identified 49 FRKs from four Rosaceae species; 20 of these sequences were from Chinese white pear. Subsequently, phylogenic relationship, gene structure and micro-collinearity were analysed. Phylogenetic and exon–intron analysis classified these FRK s into 10 subfamilies, and it was aimed to further reveal the variation of the gene structure and the evolutionary relationship of this gene family. Remarkably, gene expression patterns in different tissues or different development stages of the pear fruit suggested functional redundancy for PbFRKs derived from segmental duplication or genome-wide duplication and sub-functionalization for some of them. Additionally, PbFRK11 , PbFRK13 and PbFRK16 were found to play important roles in regulating the sugar content in the fruit. Overall, this study provided important insights into the evolution of the FRK gene family in four Rosaceae species, and highlighted its roles in both pear tissue and fruits. Results presented here provide the appropriate candidate of PbFRK s that might contribute to fructose efflux in the pear fruit.

    Description: Members of the myocyte enhancer factor-2 (MEF2) family of MADS (MCM1, agamous, deficiens, serum response factor)-box transcription factors bind an A-T-rich DNA sequence associated with muscle-specific genes. The murine MEF2C gene is expressed in heart precursor cells before formation of the linear heart tube. In mice homozygous for a null mutation of MEF2C, the heart tube did not undergo looping morphogenesis, the future right ventricle did not form, and a subset of cardiac muscle genes was not expressed. The absence of the right ventricular region of the mutant heart correlated with down-regulation of the dHAND gene, which encodes a basic helix-loop-helix transcription factor required for cardiac morphogenesis.     Description: Glial cell line-derived neurotrophic factor (GDNF) supports growth and survival of dopaminergic (DA) neurons. A replication-defective adenoviral (Ad) vector encoding human GDNF injected near the rat substantia nigra was found to protect DA neurons from the progressive degeneration induced by the neurotoxin 6-hydroxydopamine (6-OHDA) injected into the striatum. Ad GDNF gene therapy reduced loss of DA neurons approximately threefold 6 weeks after 6-OHDA lesion, as compared with no treatment or injection of Ad lacZ or Ad mGDNF (encoding a biologically inactive deletion mutant GDNF).     Description: Type VII collagen defects cause recessive dystrophic epidermolysis bullosa (RDEB), a blistering skin disorder often accompanied by epidermal cancers. To study the role of collagen VII in these cancers, we examined Ras-driven tumorigenesis in RDEB keratinocytes. Cells devoid of collagen VII did not form tumors in mice, whereas those retaining a specific collagen VII fragment (the amino-terminal noncollagenous domain NC1) were tumorigenic. Forced NC1 expression restored tumorigenicity to collagen VII-null epidermis in a non-cell-autonomous fashion. Fibronectin-like sequences within NC1 (FNC1) promoted tumor cell invasion in a laminin 5-dependent manner and were required for tumorigenesis.     Description: The wide application of anaerobic digestion (AD) for waste sludge results in a huge amount of digested sludge, while the appropriate reuse of digested sludge depends on effective solid-liquid separation. Thus, chemical (acid/alkali) pretreatment effects on dewaterability of digested paper mill sludge (DPMS) for better downstream reuse based on enhanced solid- liquid separation were investigated in this research. The dewatering properties of paper mill sludge (PMS) were also investigated to elucidate the impact of AD on sludge dewaterability. The results indicated that a higher DPMS dewaterability was noted with acid pretreatment (pH5). A 41.37% moisture content and 74.41% dewatering efficiency were determined for DPMS after acid (pH5) pretreatment within 25 min. In addition, a 7.13 mg•g-1 VSS of extracellular polymeric substances (EPS) and 101.50 μm of average particle size were observed.     Description: Cytosolic RNA sensing is a prerequisite for initiation of innate immune response against RNA viral pathogens. Signaling through RIG-I (retinoic acid–inducible gene I)–like receptors (RLRs) to TBK1 (Tank-binding kinase 1)/IKK (IB kinase ) kinases is transduced by mitochondria-associated MAVS (mitochondrial antiviral signaling protein). However, the precise mechanism of how MAVS-mediated TBK1/IKK activation is strictly controlled still remains obscure. We reported that protein phosphatase magnesium-dependent 1A (PPM1A; also known as PP2Cα), depending on its catalytic ability, dampened the RLR-IRF3 (interferon regulatory factor 3) axis to silence cytosolic RNA sensing signaling. We demonstrated that PPM1A was an inherent partner of the TBK1/IKK complex, targeted both MAVS and TBK1/IKK for dephosphorylation, and thus disrupted MAVS-driven formation of signaling complex. Conversely, a high level of MAVS can dissociate the TBK1/PPM1A complex to override PPM1A-mediated inhibition. Loss of PPM1A through gene ablation in human embryonic kidney 293 cells and mouse primary macrophages enabled robustly enhanced antiviral responses. Consequently, Ppm1a –/– mice resisted to RNA virus attack, and transgenic zebrafish expressing PPM1A displayed profoundly increased RNA virus vulnerability. These findings identify PPM1A as the first known phosphatase of MAVS and elucidate the physiological function of PPM1A in antiviral immunity on whole animals.

    Description: 〈p〉Dielectric capacitors with ultrahigh power densities are fundamental energy storage components in electrical and electronic systems. However, a long-standing challenge is improving their energy densities. We report dielectrics with ultrahigh energy densities designed with polymorphic nanodomains. Guided by phase-field simulations, we conceived and synthesized lead-free BiFeO〈sub〉3〈/sub〉-BaTiO〈sub〉3〈/sub〉-SrTiO〈sub〉3〈/sub〉 solid-solution films to realize the coexistence of rhombohedral and tetragonal nanodomains embedded in a cubic matrix. We obtained minimized hysteresis while maintaining high polarization and achieved a high energy density of 112 joules per cubic centimeter with a high energy efficiency of ~80%. This approach should be generalizable for designing high-performance dielectrics and other functional materials that benefit from nanoscale domain structure manipulation.〈/p〉

    Description: [1]  We employ a global magnetohydrodynamic model to study the effects of the interplanetary magnetic field (IMF) strength and direction upon the cross-section of the magnetotail at lunar distances. The anisotropic pressure of draped magnetosheath magnetic field lines and the inclusion of a reconnection-generated standing slow mode wave fan bounded by a rotational discontinuity within the definition of the magnetotail result in cross-sections elongated in the direction parallel to the component of the IMF in the plane perpendicular to the Sun-Earth line. Tilted cross-tail plasma sheets separate the northern and southern lobes within these cross-sections. Greater fast mode speeds perpendicular than parallel to the draped magnetosheath magnetic field lines result in greater distances to the bow shock in the direction perpendicular than parallel to the component of the IMF in the plane transverse to the Sun-Earth line. The magnetotail cross-section responds rapidly to variations in the IMF orientation. The rotational discontinuity associated with newly reconnected magnetic field lines requires no more than the magnetosheath convection time to appear at any distance downstream, and further adjustments of the cross-section in response to the anisotropic pressures of the draped magnetic field lines require no more than 10-20 minutes. Consequently for typical ecliptic IMF orientations and strengths, the magnetotail cross-section is oblate while the bow shock is prolate.
