    Description: A member of the four-and-a-half-LIM (FHL) domain protein family, FHL1, is highly expressed in human adult skeletal and cardiac muscle. Mutations in FHL1 have been associated with diverse X-linked muscle diseases: scapuloperoneal (SP) myopathy, reducing body myopathy, X-linked myopathy with postural muscle atrophy, rigid spine syndrome (RSS) and Emery-Dreifuss muscular dystrophy. In 2008, we identified a missense mutation in the second LIM domain of FHL1 (c.365 G〉C, p.W122S) in a family with SP myopathy. We generated a knock-in mouse model harboring the c.365 G〉C Fhl1 mutation and investigated the effects of this mutation at three time points (3–5 months, 7–10 months and 18–20 months) in hemizygous male and heterozygous female mice. Survival was comparable in mutant and wild-type animals. We observed decreased forelimb strength and exercise capacity in adult hemizygous male mice starting from 7 to 10 months of age. Western blot analysis showed absence of Fhl1 in muscle at later stages. Thus, adult hemizygous male, but not heterozygous female, mice showed a slowly progressive phenotype similar to human patients with late-onset muscle weakness. In contrast to SP myopathy patients with the FHL1 W122S mutation, mutant mice did not manifest cytoplasmic inclusions (reducing bodies) in muscle. Because muscle weakness was evident prior to loss of Fhl1 protein and without reducing bodies, our findings indicate that loss of function is responsible for the myopathy in the Fhl1 W122S knock-in mice.

    Description: Whole-genome sequencing of the protozoan pathogen Trypanosoma cruzi revealed that the diploid genome contains a predicted 22,570 proteins encoded by genes, of which 12,570 represent allelic pairs. Over 50% of the genome consists of repeated sequences, such as retrotransposons and genes for large families of surface molecules, which include trans-sialidases, mucins, gp63s, and a large novel family (〉1300 copies) of mucin-associated surface protein (MASP) genes. Analyses of the T. cruzi, T. brucei, and Leishmania major (Tritryp) genomes imply differences from other eukaryotes in DNA repair and initiation of replication and reflect their unusual mitochondrial DNA.     Description: Mus81-Eme1 endonuclease has been implicated in the rescue of stalled replication forks and the resolution of meiotic recombination intermediates in yeast. We used gene targeting to study the physiological requirements of Mus81 in mammals. Mus81-/- mice are viable and fertile, which indicates that mammalian Mus81 is not essential for recombination processes associated with meiosis. Mus81-deficient mice and cells were hypersensitive to the DNA cross-linking agent mitomycin C but not to gamma-irradiation. Remarkably, both homozygous Mus81-/- and heterozygous Mus81+/- mice exhibited a similar susceptibility to spontaneous chromosomal damage and a profound and equivalent predisposition to lymphomas and other cancers.     Description: New seismic profiles, bathymetric data and sediment-rock sampling document for the first time the discovery of hydrothermal vent complexes and volcanic cones at 4800-5200 m depth related to recent volcanic and intrusive activity in an unexplored area of the Canary Basin (Eastern Atlantic Ocean, 500 km west of the Canary Islands). A complex of sill intrusions is imaged on seismic profiles showing saucer-shaped, parallel or inclined geometries. Three main types of structures are related to these intrusions. Type I consists of cone-shaped depressions developed above inclined sills interpreted as hydrothermal vents. Type II is the most abundant and is represented by isolated or clustered hydrothermal domes bounded by faults rooted at the tips of saucer-shaped sills. Domes are interpreted as seabed expressions of reservoirs of CH 4 - and CO 2 -rich fluids formed by degassing and contact metamorphism of organic-rich sediments around sill intrusions. Type III are hydrothermal-volcanic complexes originated above stratified or branched inclined sills connected by a chimney to the seabed volcanic edifice. Parallel sills sourced from the magmatic chimney formed also domes surrounding the volcanic cones. Core and dredges revealed that these volcanoes, which must be among the deepest in the world, are constituted by OIB-type, basanites with an outer ring of blue-green hydrothermal Al-rich smectite muds. Magmatic activity is dated, based on lava samples, at 0.78±0.05 and 1.61±0.09 Ma (K/Ar methods) and on tephra layers within cores at 25-237 ky. The Subvent hydrothermal-volcanic complex constitutes the first modern system reported in deep-water oceanic basins related to intraplate hotspot activity.

    Description: A novel x-ray imager, which takes time-resolved gated images along a single line-of-sight, has been successfully implemented at the National Ignition Facility (NIF). This Gated Laser Entrance Hole diagnostic, G-LEH, incorporates a high-speed multi-frame CMOS x-ray imager developed by Sandia National Laboratories to upgrade the existing Static X-ray Imager diagnostic at NIF. The new diagnostic is capable of capturing two laser-entrance-hole images per shot on its 1024 × 448 pixels photo-detector array, with integration times as short as 1.6 ns per frame. Since its implementation on NIF, the G-LEH diagnostic has successfully acquired images from various experimental campaigns, providing critical new information for understanding the hohlraum performance in inertial confinement fusion (ICF) experiments, such as the size of the laser entrance hole vs. time, the growth of the laser-heated gold plasma bubble, the change in brightness of inner beam spots due to time-varying cross beam energy transfer, and plasma instability growth near the hohlraum wall.

    Description: We report an early science discovery of the 12 CO(1–0) emission line in the collisional ring galaxy VII Zw466, using the Redshift Search Receiver instrument on the Large Millimeter Telescope Alfonso Serrano. The apparent molecular-to-atomic gas ratio either places the interstellar medium (ISM) of VII Zw466 in the H i -dominated regime or implies a large quantity of CO-dark molecular gas, given its high star formation rate. The molecular gas densities and star formation rate densities of VII Zw466 are consistent with the standard Kennicutt–Schmidt star formation law even though we find this galaxy to be H 2 -deficient. The choice of CO-to- H 2 conversion factors cannot explain the apparent H 2 deficiency in its entirety. Hence, we find that the collisional ring galaxy, VII Zw466, is either largely deficient in both H 2 and H i or contains a large mass of CO-dark gas. A low molecular gas fraction could be due to the enhancement of feedback processes from previous episodes of star formation as a result of the star-forming ISM being confined to the ring. We conclude that collisional ring galaxy formation is an extreme form of galaxy interaction that triggers a strong galactic-wide burst of star formation that may provide immediate negative feedback towards subsequent episodes of star formation – resulting in a short-lived star formation history or, at least, the appearance of a molecular gas deficit.
