    Notes: An analysis of Satellite Laser Ranging (SLR) data to the LAGEOS satellite has yielded improved estimates of the horizontal motion for a subset of 34 tracking sites within the global tracking network. The analysis, called SL8.3, utilized data acquired between 1980 January and 1993 June by the global network composed of 71 sites. The solution design provides for the simultaneous estimation of site positions and their velocities within a pre-defined kinematic frame. The solution is statistically rigorous and retains the full correlation information content. Least-squares estimates of relative poles of rotation, which are used to model the motion of one plate relative to another, were made based on the SLR estimated velocities for sites known to be well away from deformation zones. The resulting SLR-based relative rotation poles differ slightly from those of NUVEL-1, but in general, indicate that the magnitude of the SLR implied velocities is slower than those implied by NUVEL-1, consistent with the 4–5 per cent slowing in relative spherical rates noted in earlier comparisons. Spherical rates between sites in western North America support models of extension in the Basin and Range Province and the rotation of the Sierra Nevada microplate. An analysis of the spherical rates crossing the North Atlantic shows that SL8.3 estimated extension between North America-Eurasia sites is generally smaller than those implied by NUVEL-1: meanwhile SL8.3 rates between North America-Africa sites are in better agreement with NUVEL-1, although they are not so well determined. The maintenance and ongoing monitoring of global SLR site kinematics provides a well-defined global reference which will aid in combination global kinematic solutions where information from other technologies are merged (e.g. Very Long Baseline Interferometry and Global Positioning System) and in providing the context for densification studies of regional kinematics derived from terrestrial and Global Positioning System observations.

    Notes: The permanent GPS tracking site at Annapolis, MD shows a 7-mm seasonal signal primarily in its horizontal position. It is suggested that thermal expansion of the pier on which the antenna rests is the source of this motion. A simple numerical model of the pier reproduces the observed motion of the GPS antenna, lending credence to this hypothesis. Although adding an additional level of complexity, this motion is predictable and the site retains it s value for high precision monitoring. Although the arrangement of this GPS site it somewhat uncommon, these results emphasize the importance of the underlying antenna monumentation when measuring crustal motions. © 1999 John Wiley & Sons, Inc.

    Notes: Abstract Turbulence during pulsatile flow has been suggested as a possible mechanism to enchance the transport of gases during high-frequency ventilation. Experimental studies on oscillatory flow in straight, circular tubes have identified three types of flow: (a) laminar; (b) conditionally turbulent, in which high-frequency disturbances occur during the decelerating phase of the flow cycle but relaminarize by the beginning of the subsequent accelerating phase; and (c) fully turbulent flow, in which disturbances occur throughout the flow cycle. Fully turbulent flow has been observed only when a mean flow is present, and only laminar or conditionally turbulent flow has been observed for purely oscillatory flow. A critical Reynolds number based on the Stokes layer can be defined, and transition Reynolds numbers between 400 and 550 have been experimentally determined for purely oscillatory flow in a circular tube, although lower values are expected for physiological flows. There are some indications that the structure of oscillating turbulent flow is similar to steady turbulent flow, and preliminary work in our laboratory shows that the spectral content of flows during high-frequency ventilation is similar to that in steady turbulent flow.

    Notes: Abstract Flow and the associated shear stress have been shown to play an active role in the regulation of the structure and function of endothelial cells (EC) in vitro. Although cultured EC subjected to flow exhibit an elongated morphology and a decreased cell growth rate rather like those observed in vivo, there are differences in morphology and growth rate, as well as other characteristics, between in vitro and in vivo EC. This suggests that flow is only one of the many factors affecting EC differentiation in vivo. In this study, a co-culture model system was designed, which includes smooth muscle cells (SMC), a matrix of collagen type I, and a confluent monolayer of EC, and this simplified model of the arterial wall was subjected to a steady, laminar shear stress of 10 and 30 dyn/cm2. Under non-flow conditions, EC exhibited an elongated shape, but with a random orientation. In response to flow, there was an alignment with the direction of flow. This alignment occurred more rapidly at 30 dyn/cm2 than at 10 dyn/cm2. The collagen matrix was found to be primordial in the maintenance of a quiescent endothelium, even in the absence of SMC and flow, suggesting the importance of an organized extracellular matrix (ECM) in the differentiation of cells in vivo.

    Notes: Abstract Tissue engineering is the application of the principles and methods of engineering and the life sciences towards the development of biological substitutes to restore, maintain or improve functions. It is an area which is emerging in importance worldwide. In the USA it has been actively fostered by the National Science Foundation, both through research grants and the sponsorship of a series of workshops starting in 1988. This brief review of activities in the USA focuses on cell culture technology as a foundation for tissue engineering and then discusses examples of applications. These include artificial skin and the use of encapsulated cells in the development of bioartificial organs. Also discussed is the reconstitution of a blood vessel in culture, both for use in basic research and for implantation as an artificial blood vessel in bypass surgery. In conclusion, other potential applications are mentioned as well as generic areas of technology for future development.

    Notes: The fluid dynamic design and characterization of a low mechanical stress agitator and bioreactor vessel for large-scale bioprocessing using anchorage dependent mammalian cells is considered. The complex and fragile nature of mammalian cells puts stringent constraints on the design of agitators for stirred tank bioreactors. Traditional agitators have difficulty meeting the competing requirements of fluidization and mixing while maintaining low enough mechanical stress levels to avoid cell damage. A rotating disc agitator design is proposed. Flow visualization and laser Doppler velocimeter measurements reveal fluidization of microcarriers with a gentle (low turbulence level), highly three-dimensional flow characterized by good mixing at low hydrodynamic shear stress levels.
