Notizen: A low cost powdered infant formula made from soybeans, oats and sucrose, fortified with methionine, vitamins and minerals, has been developed. A dry process for making the formula, which employs a low-cost Brady extruder, has also been developed. The resulting product is of good microbiological quality and disperses readily in water to give a milk-like suspension of satisfactory stability to settling. The essential amino acid pattern of the formula, its PER and NPU values, nitrogen balance data with infants, proximal chemical analysis, calorie distribution and low level of trypsin inhibitor indicate it to be adequate for feeding infants 3 months or older. Clinical trials with infants also showed that the formula was well accepted and tolerated, and produced adequate weight gain. Retail prices of the product, packed in cans and polyethylene bags, are appreciably lower than those of comparable cow's milk and other soya-based infant formulas available in the market. The product is currently being manufactured and sold in several parts of Mexico.

Notizen: A nitrogen balance study, comparing a low-cost soy-oats infant formula with and without supplemental methionine, was carried out with infants of ages 3 - 26 months at a single equivalent level of nitrogen intake (approximately 850 mg/kg/day). Nitrogen retention was found to be unaffected by methionine supplementation. Com- parison of data obtained in this study with previous research re- vealed that a critical value for intake of total L-sulfur amino acids apparently exists in infants 3 - 26 months of age, below which methionine supplementation will increase nitrogen retention, but above which it will not. This value appears to coincide with that of 49 mg/kg/day recommended for infants by the FNB.

Notizen: Mesquite beans (Prosopis juliflora) consisted of pericarp, hulls and kernels. With the exception of kernels, composition of beans and their fractions were protein, 10–15%; fat, 2–3%; crude fiber, 20–30%; sucrose, 21%; reducing sugars, 2–6%. Kernels contained 38% protein, 3% fat and 9% crude fiber. Bean trypsin inhibitor content was 1.4 TIU/mg. Bean protein isoelectric point was pH 5. Mesquite protein concentrate and isolate were prepared, however, by increasing solubility at the extraction pH (pH 10) with NaCl and decreasing it at the isoelectric point with (NH4)2SO4. First and second limiting amino acids for bean protein were threonine and isoleucine, respectively; the protein was high in total sulfur amino acids and tryptophan. Corrected PER was 1.4.

Notizen: Abstract In Schizosaccharomyces pombe, wee1 encodes a tyrosine kinase that inhibits entry into mitosis by phophorylating Cdc2, the universal cyclin-dependent kinase (Cdk) that regulates the G2/M transition in all eukaryotic cells. A search for suppressors of the G2 arrest caused by overexpression of wee1 led to the isolation of a new allele of swo1 (named swo1-w1), the gene coding for chaperone Hsp90, which is required to stabilise Wee1. The swo1-w1 allele carries a glycine to aspartic acid substitution at amino acid 155 that results in a partial loss of Hsp90 function. Cells bearing the swo1-w1 mutation in combination with the point mutation cdc2-33 or cdc2-M26 showed severe mitotic defects. Genetic interactions were not observed in combination with point mutations in other cdc genes, suggesting that Cdc2 specifically interacts with Hsp90. This synthetic lethal swo1-w1 cdc2-33 (or cdc2-M26) strain had normal levels of Cdc2 protein and histone H1 phosphorylation activity, indicating that Hsp90 is required to enable Cdc2 to interact with its mitotic substrates or regulators, rather than for its proper folding or stabilisation. In a wild-type background, swo1-w1 mutant cells were sensitive to temperature as well as to other stress agents, such as KCl, ethanol and formamide. Under these stressful growth conditions, the swo1-w1 cells displayed anaphase B arrest and aberrant septation patterns, indicating that a subset of proteins involved in mitosis and cytokinesis is highly dependent on chaperone Hsp90 for function.

Notizen: Abstract Thecdc2 + gene product (p34cdc2) is a protein kinase that regulates entry into mitosis in all eukaryotic cells. The role that p34cdc2 plays in the cell cycle has been extensively investigated in a number of organisms, including the fission yeastSchizosaccharomyces pombe. To study the degree of functional conservation among evolutionarily distant p34cdc2 proteins, we have constructed aS. pombe strain in which the yeastcdc2 + gene has been replaced by itsDrosophila homologue CDC2Dm (theCDC2Dm strain). ThisCDC2Dm S. pombe strain is viable, capable of mating and producing four viable meiotic products, indicating that the fly p34CDC2Dm recognizes all the essentialS. pombe cdc2 + substrates, and that it is recognized by cyclin partners and other elements required for its activity. The p34CDC2Dm protein yields a lethal phenotype in combination with the mutant B-type cyclin p56cdc13-117, suggesting that thisS. pombe cyclin might interact less efficiently with theDrosophila protein than with its native p34cdc2 counterpart. ThisCDC2Dm strain also responds to nutritional starvation and to incomplete DNA synthesis, indicating that proteins involved in these signal transduction pathways, interact properly with p34CDC2Dm (and/or that p34cdc2-independent pathways are used). TheCDC2Dm gene produces a ‘wee’ phenotype, and it is largely insensitive to the action of theS. pombe weel + mitotic inhibitor, suggesting thatDrosophila weel + homologue might not be functionally conserved. ThisCDC2Dm strain is hypersensitive to UV irradiation, to the same degree asweel-deficient mutants. A strain which co-expresses theDrosophila and yeastcdc2+ genes shows a dominantwee phenotype, but displays a wild-type sensitivity to UV irradiation, suggesting that p34cdc2 triggers mitosis and influences the UV sensitivity by independent mechanisms.

Beschreibung: During the ADRIMED (Aerosol Direct Radiative Impact on the regional climate in the Mediterranean region) special observation period (SOP-1a), conducted in June 2013 in the framework of the ChArMEx (Chemistry-Aerosol Mediterranean Experiment) project, a moderate Saharan dust event swept the Western and Central Mediterranean Basin (WCMB) from west to east during a 9-day period between 16 and 24 June. This event was monitored from the ground by six EARLINET/ACTRIS (European Aerosol Research Lidar Network/Aerosols, Clouds, and Trace gases Research Infrastructure Network) lidar stations (Granada, Barcelona, Naples, Potenza, Lecce and Serra la Nave) and twoADRIMED/ChArMEx lidar stations specially deployed for the field campaign in Cap d’en Font and Ersa, in Minorca and Corsica Islands, respectively. The first part of the study shows the spatio-temporal monitoring of the dust event during its transport over the WCMB with ground-based lidar and co-located AERONET (Aerosol Robotic Network) Sun-photometer measurements. Dust layer optical depths, Ångström exponents, coarse mode fractions, linear particle depolarization ratios (LPDRs), dust layer heights and the dust radiative forcing estimated in the shortwave (SW) and longwave (LW) spectral ranges at the bottom of the atmosphere (BOA) and at the top of the atmosphere (TOA) with the Global Atmospheric Model (GAME), have been used to characterize the dust event. Peak values of the AERONETaerosol optical depth (AOD) at 440 nm ranged between 0.16 in Potenza and 0.37 in Cap d’en Font. The associated Ångström exponent and coarse mode fraction mean values ranged from 0.43 to 1.26 and from 0.25 to 0.51, respectively. The mineral dust produced a negative SWdirect radiative forcing at the BOA ranging from −56.9 to −3.5Wm−2. TheLW radiative forcing at the BOAwas positive, ranging between +0.3 and +17.7 W m-2. The BOA radiative forcing estimates agree with the ones reported in the literature. At the TOA, the SW forcing varied between −34.5 and +7.5 W m−2. In seven cases, the forcing at the TOA resulted positive because of the aerosol strong absorbing properties (0.83 〈 single-scattering albedo (SSA) 〈 0.96). The multi-intrusion aspect of the event is examined by means of air- and space-borne lidar measurements, satellite images and back trajectories. The analysis reported in this paper underline the arrival of a second different intrusion of mineral dust observed over southern Italy at the end of the considered period which probably results in the observed heterogeneity in the dust properties.

Beschreibung: In the framework of the Chemistry-Aerosol Mediterranean Experiment (ChArMEx; http://charmex.lsce.ipsl.fr/) initiative, a field campaign took place in the western Mediterranean Basin between 10 June and 5 July 2013 within the ADRIMED (Aerosol Direct Radiative Impact on the regional climate in the MEDiterranean region) project. The scientific objectives of ADRIMED are the characterization of the most common ‘Mediterranean aerosols’ and their direct radiative forcing (column closure and regional scale). During 15–24 June a multiintrusion dust event took place over the western and central Mediterranean Basin. Extra measurements were carried out by some EARLINET/ACTRIS (European Aerosol Research Lidar Network /Aerosols, Clouds, and Trace gases Research InfraStructure Network, http://www.actris.net/) lidar stations in Spain and Italy, in particular on 22 June in support to the flight over southern Italy of the Falcon 20 aircraft involved in the campaign. This article describes the physical and optical properties of dust observed at the different lidar stations in terms ofdust plume centre of mass, optical depth, lidar ratio, and particle depolarization ratio. To link the differences found in the origin of dust plumes, the results are discussed on the basis of back-trajectories and air- and space-borne lidars. This work puts forward the collaboration between a European research infrastructure (ACTRIS) and an international project (ChArMEx) on topics of interest for both parties, and more generally for the atmospheric community.
