    Notes: Abstract An understanding of spawning and larval development can be fundamental to interpreting the abundance, distribution, and population structure of marine invertebrate taxa. Tegula funebralis (A. Adams, 1855), the black turban snail, has been the focus of numerous ecological studies on the Pacific coast of North America. To date, there are only conflicting and anecdotal reports of spawning, and there is no information on larval or juvenile development for this conspicuous and abundant species. On 19 September 1995, two individuals of T. funebralis were observed free-spawning gametes into seawater in tanks at the Oregon Institute of Marine Biology. Embryos and larvae were subsequently reared to metamorphosis and beyond. Development was pelagic and similar to development described for other trochids, and larvae were observed not to feed at any stage. Larvae began to metamorphose at 5.7 to 6.7 d and settled at 260 μm shell length. Juveniles grew ≃ 10 μm in shell length per day and appeared to feed on detritus. Juveniles lacked some adult diagnostic shell characters, including two columellar nodes and a closed umbilicus. In the field, small (〈3 mm) juveniles occurred in the adult habitat on all sampling dates between October and March. Small juveniles were found only under rocks and were most abundant under rocks partially buried in coarse sand, suggesting that juveniles may utilize a specific microhabitat within the adult T. funebralis habitat.

    Notes: Abstract A nontoxic method of marking juvenile animals is a prerequisite for many field studies investigating growth and survivorship in marine invertebrates. This study investigates the effectiveness of low concentrations of calcein in marking hatchling snails (Nucella ostrina), the durability of the calcein mark, and the effects of marking on survivorship and growth. I also describe an inexpensive means of visualizing the calcein mark under a dissecting microscope. Results demonstrate that calcein provides a long-lasting, readily detected fluorescent shell mark that can be used to measure shell growth accurately. In addition, marking with calcein did not affect survivorship or growth, and had no size-dependent effects on growth or survivorship.

    Notes: Summary The stoichiometric properties of the renal outer cortical brush-border membraned-glucose transporter are studied. Experiments which establish the glucose/sodium, glucose/phlorizin and phlorizin/sodium stoichiometries are reported. Three independent methods of determining the substrate/activator (glucose/sodium) stoichiometry for coupled transport systems are presented and discussed. One of these, the “Static Head Method,” is introduced here for the first time. This type of experiment appears to be more generally applicable than the usual procedure of directly measuring the coupled fluxes of substrate and activator to determine stoichiometric coupling ratios. The results presented in this paper demonstrate that the glucose/sodium/phlorizin stoichiometry of the renal outer cortical brush-border membraned-glucose transport system is 1∶1∶1.

    Notes: Summary We have shown previously that the concentration of glucose in the growth medium regulates sodium-coupled hexose transport in epithelia formed by the porcine renal cell line LLC-PK1. Assayed in physiological salt solution, the ratio of the concentration of α-methyl glucoside (AMG) accumulated inside the cell at steady state to its concentration outside, and the number of glucose transporters, as measured by phlorizin binding, was inversely related to the glucose concentration in the growth medium. In this study, using a cloned line of LLC-PK1 cells, we provide evidence that the difference in AMG concentrating capacity is the result of a regulatory signal and not simply due to a selection process where the growth of cells with enhanced glucose transport is favored by low glucose medium or vice-versa. By adding glucose to conditioned medium (collected after 48 hr incubation with cells and therefore containing less than 0.1mm glucose), we demonstrate that the signal in the growth medium is indeed the concentration of glucose rather than another factor secreted into or depleted from the medium. Fructose and mannose, two sugars not transported by the sodium-dependent glucose transporter, can substitute for glucose as a carbohydrate source in the growth medium and have a modest glucose-like effect on the transporter. Growth in medium containing AMG does not affect the transporter, indicating that the regulatory signal is not a direct effect of the hexose on its carrier but involves hexose metabolism.

    Notes: A commercially available enzyme immunoassay (ELISA) in which a myeloma protein (MOPC 467) is used for detection of salmonellae was compared with two conventional cultural methods for detection of salmonellae in naturally contaminated meat and poultry products. Products tested included mechanically deboned poultry, chitterlings, poultry carcass rinsings, chicken necks, luncheon loaf emulsion, pork sausage and basturma. There was 100% agreement between ELISA and cultural methods. The ELISA technique is specific and rapid. Identification of Salmonella-contaminated meat and poultry products was accomplished in 2-3 days compared to the 4-6 days required by conventional cultural methods.
