    Notes: Summary Tissue composition, membrane potentials and cellular activity of potassium, sodium and chloride have been measured in innervated and denervated rat skeletal muscles incubatedin vitro. After denervation for 3 days, tissue water, sodium and chloride were increased but cellular potassium content and measured activity were little affected, despite a decrease of 16 mV in resting membrane potential which would have necessitated a decrease in cellular potassium activity of almost 50% were potassium distributed at electrochemical equilibrium. These findings, therefore, preclude a decreased electrochemical potential gradient for potassium as the cause of the membrane depolarization characteristic of denervated muscle fibers. Analysis of the data excludes an important contribution of rheogenic sodium transport to the resting potential of innervated muscles. These results strongly support the hypothesis that the decreased membrane potential in denervated fibers reflects a relative increase in the membrane permeability to sodium.

    Notes: Summary Measurement of intracellular chloride activity in theextensor digitorum longus (EDL) muscle of the rat using liquid ion-exchanger microelectrodes gave an apparent resting value of 10 to 11 mmol liter−1. If chloride ions were distributed passively across the muscle fiber membrane the predicted value would be 4 mmol liter−1. In experiments in which the bathing fluid was changed by (a) reduction of external chloride, or incubation at low external chloride followed by a return to normal concentrations, (b) an increase or reduction in external potassium, (c) alteration in potassium and chloride in the bathing medium so as to maintain a constant [K]×[Cl] product, or in other experiments in which the membrane potential was caused to change by anoxia, or by addition of ouabain to the medium, changes in intracellular chloride activity were invariably consistent with the hypothesis that this ion is passively distributed. Measurements of intracellular chloride activity with recessed-tip solid-state Ag/AgCl electrodes gave a value of 4.6 mmol liter−1. Since the liquid ion-exchanger is known to be poorly selective for chloride, it is concluded that the chloride ion is passively distributed in rat EDL muscle.

    Notes: Abstract Marine ammonia oxidising bacteria were enriched by incubation of sea water, amended with ammonium sulphate, and subsequent subculture in liquid inorganic medium. PCR primers were designed to be specific for rDNA sequences from ammonia oxidisers belonging to the β-rsub-group of the proteobacteria. These primers were then used to amplify rRNA genes from ammonia oxidiser enrichment cultures containing heterotrophs. PCR products were recovered from all cultures in which complete ammonia oxidation occurred. Subsequent rDNA sequence analysis indicated the presence of three new lineages within the clade defined by sequences of cultured β-sub-group ammonia oxidisers. Two of the new lineages showed moderate similarity to sequences from pure cultures of ammonia oxidisers previously isolated from marine and brackish environments. The third lineage (AEM-3) was deep branching and occupied an intermediate position between clades defined by Nitrosomonas or Nitrosospira, which were isolated from soil or sewage. The phylogenetic analysis suggests that, in enrichment cultures, the primers are specific for members of the target group, the β-proteobacteria ammonia oxidisers. The results also indicate the presence of previously unknown ammonia oxidisers in marine samples. The approach enabled analysis of ammonia oxidiser enrichments at an early stage and without the requirement for isolation of pure cultures, significantly reducing the time required and facilitating quantitative assessment of relatedness of strains.

    Notes: A mathematical model was developed to solve a steady free surface flow problem and a rapid drawdown problem in a two-dimensional porous medium. The same problem was also solved by an analogue device and excellent agreement was found to exist between the two solutions. This paper contains the formulation of the numerical problem from first principles and a discussion of measures that had to be taken in order to assure numerical stability and proper convergence of the solution.Although the scope of this study was limited to a two-dimensional flow case, the elements of simulation discussed are general in nature and applicable to three-dimensional problems. It was demonstrated that numerical solution can be obtained for the position of the free surface at given time intervals, for the piezometric head distribution within the flow field and for flow quantities across given boundaries. In addition, the mathematical model will permit consideration of nonhomogeneous or anisotropic characteristics of the porous medium, without difficulty.It is concluded that mathematical models, incorporating some or all of the techniques discussed in this paper, in conjunction with some analogue control device, can be very efficient and reliable tools for solving complex porous flow problems, including those which, so far, have eluded comprehensive analysis, due to physical and/or cost limitation.
