    Description: Closed-loop neurophysiology has been accelerated by recent software and hardware developments and by the emergence of novel tools to control neuronal activity with spatial and temporal precision, in which stimuli are delivered in real time based on recordings or behavior. Real-time stimulation feedback enables a wide range of innovative studies of information processing and plasticity in neuronal networks. This Research Topic e-Book comprises 16 Original Research Articles, seven Methods Articles, and seven Reviews, Mini- Reviews, and Perspectives, all peer-reviewed and published in Frontiers in Neural Circuits. The contributions deal with closed loop neurophysiology experiments at a variety of levels of neural circuit complexity. Some include modeling and theoretical analyses. New enabling technologies and techniques are described. Novel work is presented from experiments in vitro, in vivo, and in humans, along with their clinical and technological implications for improving the human condition.

    Description: Gas-puff Z-pinch experiments were conducted on the 1 MA, 200 ns pulse duration Cornell Beam Research Accelerator (COBRA) pulsed power generator in order to achieve an understanding of the dynamics and instability development in the imploding and stagnating plasma. The triple-nozzle gas-puff valve, pre-ionizer, and load hardware are described. Specific diagnostics for the gas-puff experiments, including a Planar Laser Induced Fluorescence system for measuring the radial neutral density profiles along with a Laser Shearing Interferometer and Laser Wavefront Analyzer for electron density measurements, are also described. The results of a series of experiments using two annular argon (Ar) and/or neon (Ne) gas shells (puff-on-puff) with or without an on- (or near-) axis wire are presented. For all of these experiments, plenum pressures were adjusted to hold the radial mass density profile as similar as possible. Initial implosion stability studies were performed using various combinations of the heavier (Ar) and lighter (Ne) gasses. Implosions with Ne in the outer shell and Ar in the inner were more stable than the opposite arrangement. Current waveforms can be adjusted on COBRA and it was found that the particular shape of the 200 ns current pulse affected on the duration and diameter of the stagnated pinched column and the x-ray yield.

    Description: ABSTRACT Inhibiting the NF-κB signaling pathway provides morphological and functional benefits for the mdx mouse, a model for Duchenne muscular dystrophy characterized by chronic elevations in the nuclear expression of p65, the transactivating component of the NF-κB complex. The purpose of this study was to examine p65 expression in nondystrophic and mdx myotubes using confocal immunofluorescence, and determine whether inhibitors of the NF-κB pathway alter myotube development. Primary cultures of nondystrophic and mdx myotubes had identical levels of nuclear and cytosolic p65 expression and exhibited equivalent responses to TNF-α, thus excluding the hypothesis that the lack of dystrophin is sufficient to induce increases in NF-κB signaling. The NF-κB inhibitors pyrrolidine dithiocarbamate (PDTC) and sulfasalazine decreased spontaneous contractile activity and reduced myotube viability in a dose- and time-dependent manner. Similarly, a vivo-morpholino designed to block translation of murine p65 (m-p65tb-vivomorph1) rapidly abolished spontaneous contractile activity, reduced p65 expression measured by confocal immunofluorescence, and induced cell death in primary cultures of nondystrophic and mdx myotubes. Similar effects on p65 immunofluorescence and cell viability were observed following m-p65tb-vivomorph1 exposure to spontaneously inactive C2C12 myotubes, while exposure to a control scrambled vivo morpholino had no effect. These results indicate a direct role of the NF-κB pathway in myotube development and identify a potential therapeutic limitation to the use of NF-κB inhibitors in treating Duchenne and related muscular dystrophies.     Description: Increased phosphorus (P) fertilizer use and livestock production has fundamentally altered the global P cycle. We calculated spatially explicit P balances for cropland soils at 0.5° resolution based on the principal agronomic P inputs and outputs associated with production of 123 crops globally for the year 2000. Although agronomic inputs of P fertilizer (14.2 Tg of P·y−1) and manure (9.6 Tg of P·y−1) collectively exceeded P removal by harvested crops (12.3 Tg of P·y−1) at the global scale, P deficits covered almost 30% of the global cropland area. There was massive variation in the magnitudes of these P imbalances across most regions, particularly Europe and South America. High P fertilizer application relative to crop P use resulted in a greater proportion of the intense P surpluses (〉13 kg of P·ha−1·y−1) globally than manure P application. High P fertilizer application was also typically associated with areas of relatively low P-use efficiency. Although manure was an important driver of P surpluses in some locations with high livestock densities, P deficits were common in areas producing forage crops used as livestock feed. Resolving agronomic P imbalances may be possible with more efficient use of P fertilizers and more effective recycling of manure P. Such reforms are needed to increase global agricultural productivity while maintaining or improving freshwater quality.

    Description: Nitrous oxide (N2O) is a potent greenhouse gas that contributes to climate change and stratospheric ozone destruction. Anthropogenic nitrogen (N) loading to river networks is a potentially important source of N2O via microbial denitrification that converts N to N2O and dinitrogen (N2). The fraction of denitrified N that escapes as N2O rather than N2 (i.e., the N2O yield) is an important determinant of how much N2O is produced by river networks, but little is known about the N2O yield in flowing waters. Here, we present the results of whole-stream 15N-tracer additions conducted in 72 headwater streams draining multiple land-use types across the United States.     Description: Despite conservation efforts, most forest ecosystems worldwide are affected by biotic invasions; however, the specific impacts vary across different geographic regions and forest types. The relative contributions of the main drivers such as propagule pressure (e.g., due to human population, travel, and trade), climate, land use, and habitat invasibility remain uncertain. The special issue “Impacts of Nonnative Species on the Health of Natural and Planted Forests” was organized to facilitate timely communications among scientists and managers in different regions and to assist in attempts to improve forest health and maintain long-term sustainability. The special issue addresses broad issues related to forest invasions, including the impacts of nonnative species in various forest ecosystems (e.g., natural vs. urban) and the contributions of land use (e.g., fragmentation), human activity, and climate change to invasion. The new findings include identifying hotspots of potential invasion impacts and their causes, which can help inform policy makers as they develop effective strategies for prevention, early detection or eradication, and forest management.

    Description: Diseases and insects, particularly those that are non-native and invasive, arguably pose the most destructive threat to North American forests. Currently, both exotic and native insects and diseases are producing extensive ecological damage and economic impacts. As part of an effort to identify United States tree species and forests most vulnerable to these epidemics, we compiled a list of the most serious insect and disease threats for 419 native tree species and assigned a severity rating for each of the 1378 combinations between mature tree hosts and 339 distinct insect and disease agents. We then joined this list with data from a spatially unbiased and nationally consistent forest inventory to assess the potential ecological impacts of insect and disease infestations. Specifically, potential host species mortality for each host/agent combination was used to weight species importance values on approximately 132,000 Forest Inventory and Analysis (FIA) plots across the conterminous 48 United States. When summed on each plot, these weighted importance values represent an estimate of the proportion of the plot’s existing importance value at risk of being lost. These plot estimates were then used to identify statistically significant geographic hotspots and coldspots and of potential forest impacts associated with insects and diseases in total, and for different agent types. In general, the potential impacts of insects and diseases were greater in the West, where there are both fewer agents and less diverse forests. The impact of non-native invasive agents, however, was potentially greater in the East. Indeed, the impacts of current exotic pests could be greatly magnified across much of the Eastern United States if these agents are able to reach the entirety of their hosts’ ranges. Both the list of agent/host severities and the spatially explicit results can inform species-level vulnerability assessments and broad-scale forest sustainability reporting efforts, and should provide valuable information for decision-makers who need to determine which tree species and locations to target for monitoring efforts and pro-active management activities.
