    Description: We invite you to contribute to a Special Issue of Nutrients, entitled “Dietary Supplements”. The purpose of this Special Issue is to advance dietary supplement science by presenting commissioned overviews on four nutrients of particular current interest and controversy: Vitamin D, iodine, iron, and omega-3 fatty acids. To supplement these contributions, we welcome the submission of manuscripts describing original research or providing systematic reviews related to various issues in dietary supplement science. Manuscripts across a broad range of topics will be considered, but we are particularly interested in manuscripts that address the following areas: • Health effects, both positive and negative, of single nutrients, such as vitamin D, iodine, iron, and omega-3 fatty acids, with an emphasis on human studies. • Human clinical trials of dietary supplement use • Motivations for and prevalence of the use of dietary supplements • Mechanisms of action of nutrients and other bioactive components of dietary supplements. • Biomarkers of nutritional status, especially those measured in human samples. • Development and application of analytical tools for the measurement of bioactive components of dietary supplements. • Databases of dietary supplements' composition for use in improving the assessment of nutrient intake and of the exposure to ingredients present in these products in human populations.

    Description: Some 20% of the population is required to work outside the regular 9:00 a.m.–5:00 p.m. working day, and this number is likely to increase as economic demands push work hours into the night for many companies. These irregular schedules mean workers often have to sleep during the day and be awake at night. This causes a misalignment between normal day-light entrained internal physiological processes, such as metabolism and digestion, and the external environment. As a direct consequence, night workers have poorer health than day workers, even after controlling for lifestyle and socioeconomic status. The purpose of this Special Issue is to highlight the interrelationships between timing of food intake and diet quality with sleep and work patterns in humans with an emphasis on randomized controlled trials or meta-analyses of data from published studies.

    Description: Almost nothing is known about the mechanisms of dissimilatory metal reduction by Gram-positive bacteria, although they may be the dominant species in some environments. Thermincola potens strain JR was isolated from the anode of a microbial fuel cell inoculated with anaerobic digester sludge and operated at 55 °C. Preliminary characterization revealed that T. potens coupled acetate oxidation to the reduction of hydrous ferric oxides (HFO) or anthraquinone-2,6-disulfonate (AQDS), an analog of the redox active components of humic substances. The genome of T. potens was recently sequenced, and the abundance of multiheme c-type cytochromes (MHCs) is unusual for a Gram-positive bacterium. We present evidence from trypsin-shaving LC-MS/MS experiments and surface-enhanced Raman spectroscopy (SERS) that indicates the expression of a number of MHCs during T. potens growth on either HFO or AQDS, and that several MHCs are localized to the cell wall or cell surface. Furthermore, one of the MHCs can be extracted from cells with low pH or denaturants, suggesting a loose association with the cell wall or cell surface. Electron microscopy does not reveal an S-layer, and the precipitation of silver metal on the cell surface is inhibited by cyanide, supporting the involvement of surface-localized redox-active heme proteins in dissimilatory metal reduction. These results provide unique direct evidence for cell wall-associated cytochromes and support MHC involvement in conducting electrons across the cell envelope of a Gram-positive bacterium.

    Description: Many oomycete and fungal plant pathogens are obligate biotrophs, which extract nutrients only from living plant tissue and cannot grow apart from their hosts. Although these pathogens cause substantial crop losses, little is known about the molecular basis or evolution of obligate biotrophy. Here, we report the genome sequence of the oomycete Hyaloperonospora arabidopsidis (Hpa), an obligate biotroph and natural pathogen of Arabidopsis thaliana. In comparison with genomes of related, hemibiotrophic Phytophthora species, the Hpa genome exhibits dramatic reductions in genes encoding (i) RXLR effectors and other secreted pathogenicity proteins, (ii) enzymes for assimilation of inorganic nitrogen and sulfur, and (iii) proteins associated with zoospore formation and motility.     Description: With more than 55,000 members identified so far in all forms of life, the family of terpene or terpenoid natural products represents the epitome of molecular biodiversity. A well-known and important member of this family is the polycyclic diterpenoid Taxol (paclitaxel), which promotes tubulin polymerization and shows remarkable efficacy in cancer chemotherapy. The first committed step of Taxol biosynthesis in the Pacific yew (Taxus brevifolia) is the cyclization of the linear isoprenoid substrate geranylgeranyl diphosphate (GGPP) to form taxa-4(5),11(12)diene, which is catalysed by taxadiene synthase. The full-length form of this diterpene cyclase contains 862 residues, but a roughly 80-residue amino-terminal transit sequence is cleaved on maturation in plastids. We now report the X-ray crystal structure of a truncation variant lacking the transit sequence and an additional 27 residues at the N terminus, hereafter designated TXS. Specifically, we have determined structures of TXS complexed with 13-aza-13,14-dihydrocopalyl diphosphate (1.82 A resolution) and 2-fluorogeranylgeranyl diphosphate (2.25 A resolution). The TXS structure reveals a modular assembly of three alpha-helical domains. The carboxy-terminal catalytic domain is a class I terpenoid cyclase, which binds and activates substrate GGPP with a three-metal ion cluster. The N-terminal domain and a third 'insertion' domain together adopt the fold of a vestigial class II terpenoid cyclase.
