    Description: 〈p〉Despite the importance of glucose and amino acids for energy metabolism, interactions between the two nutrients are not well understood. We provide evidence for a role of leucyl-tRNA synthetase 1 (LARS1) in glucose-dependent control of leucine usage. Upon glucose starvation, LARS1 was phosphorylated by Unc-51 like autophagy activating kinase 1 (ULK1) at the residues crucial for leucine-binding. The phosphorylated LARS1 showed decreased leucine-binding, which may inhibit protein synthesis and help save energy. Leucine, not used to anabolic process, may be available to catabolic pathway for energy generation. The LARS1-mediated changes in leucine utilization might help support cell survival deprived of glucose. Thus, dependent on the availability of glucose, LARS1 may help regulate whether leucine is used for protein synthesis or energy production.〈/p〉

    Notes: Abstract  Disruptive selection for responsiveness to queen mandibular gland pheromone (QMP) in the retinue bioassay resulted in the production of high and low QMP responding strains of honeybees (Apis mellifera L.). Strains differed significantly in their retinue response to QMP after one generation of selection. By the third generation the high strain was on average at least nine times more responsive than the low strain. The strains showed seasonal phenotypic plasticity such that both strains were more responsive to the pheromone in the spring than in the fall. Directional selection for low seasonal variation indicated that phenotypic plasticity was an additional genetic component to retinue response to QMP. Selection for high and low retinue responsiveness to QMP was not an artifact of the synthetic blend because both strains were equally responsive or non-responsive to whole mandibular gland extracts compared with QMP. The use of these strains clearly pointed to an extra-mandibular source of retinue pheromones (Pankiw et al. 1995; Slessor et al. 1998; Keeling et al. 1999).

    Notes: Abstract Dexamethasone-β-D-glucoside is a potential prodrug for colonic delivery of the antiinflammatory agent, dexamethasone. The ability of this prodrug to deliver dexamethasone selectively to the colon depends not only on its being slowly absorbed from the alimentary canal, but also on its having chemical and enzymatic stability in the stomach and small intestine. Once reaching the large bowel, it should be quantitatively hydrolyzed to release the active agent. The potential of dexamethasone-β-D-glucoside for colon-specific delivery of dexamethasone is assessed by determining the rates of its hydrolysis down the alimentary canal of the guinea pig, an animal in which an inflammatory bowel disease model has been developed. The hydrolytic activity is examined in tissues and luminal contents of the stomach, proximal and distal segments of the small intestine, cecum, and colon. For the tissues, the greatest hydrolytic activity is in the proximal small intestine, while the stomach, cecum, and colon have only moderate activity. In contrast, the contents of the cecum and colon show greater activity than the contents of the small intestine and stomach. The luminal contents retained β-glucosidase activity even after repeated centrifugation and resuspension in a buffer. The activity was unaffected by homogenization. These observations suggest that hydrolytic activity is associated with enzymes located on the surface of luminal cells. The movement and hydrolysis of dexamethasone-β-D-glucoside down the gastrointestinal tract of the guinea pig are also examined. About 20 to 30% of an oral dose appears to reach the cecum. Here the prodrug is rapidly hydrolyzed to the active drug. From intravenous administration of the prodrug and drug, it is apparent that dexamethasone-β-D-glucoside is poorly absorbed in the gastrointestinal tract (bioavailability, 〈1%). There is a ninefold selective advantage for delivery of dexamethasone in cecal tissues in the guinea pig under the conditions of this experiment. Thus, there is a potential for a decrease in the usual dose and a concomitant reduction in the systemic exposure to dexamethasone. Because humans have much less glucosidase activity in the small intestine, even greater site-selective delivery to the cecum and colon is expected.

    Notes: Abstract There have been numerous reports of genetic influences on division of labor in honey bee colonies, but the effects of worker genotypic diversity on colony behavior are unclear. We analyzed the effects of worker genotypic diversity on the phenotypes of honey bee colonies during a critical phase of colony development, the “nest initiation” phase. Five groups of colonies were studied (n = 5–11 per group); four groups had relatively low genotypic diversity compared to the fifth group. Colonies were derived from queens that were instrumentally inseminated with the semen of four different drones according to one of the following mating schemes: group A, 4 A-source drones; group B, 4 B-source drones; group C, 4 C-source drones; group D, 4 D-source drones; and group E, 1 drone of each of the A-D drone sources. There were significant differences between colonies in groups A-D for 8 out of 19 colony traits. Because the queens in all of these colonies were super sisters, the observed differences between groups were primarily a consequence of differences in worker genotypes. There were very few differences (2 out of 19 traits) between colonies with high worker genotypic diversity (group E) and those with low diversity (groups A-D combined). This is because colonies with greater diversity tended to have phenotypes that were average relative to colonies with low genotypic diversity. We hypothesize that the averaging effect of genotypic variability on colony phenotypes may have selective advantages, making colonies less likely to “fail” because of inappropriate colony responses to changing environmental conditions.

    Notes: Summary Two experiments were performed to determine whether worker reproduction in queenless honey bee colonies is influenced by colony genetic structure. In Experiment 1, allozyme analyses of workers and worker-derived drone larvae revealed that in half the colonies, there were genotypic differences in worker egg-laying behavior (presumed to involve actual oviposition), but biases in drone production were not always consistent with biases in egg-laying behavior. In Experiment 2, allozyme analyses again revealed intracolonial differences in egg-laying behavior and in behavior patterns thought to involve oophagy and larval care. Data support the hypothesis of a genetic influence on this intracolonial behavioral variation. Differences in the genotypic distributions of worker-derived drones relative to workers engaged in oviposition behavior in queenless colonies may be a consequence of genetic variability for egg production or for treatment of eggs and larvae (possibly coupled with kin recognition), or both.

    Notes: Abstract Pollen storage in a colony of Apis mellifera is actively regulated by increasing and decreasing pollen foraging according to the “colony's needs.” It has been shown that nectar foragers indirectly gather information about the nectar supply of the colony from nestmates without estimating the amount of honey actually stored in the combs. Very little is known about how the actual colony need is perceived with respect to pollen foraging. Two factors influence the need for pollen: the quantity of pollen stored in cells and the amount of brood. To elucidate the mechanisms of perception, we changed the environment within normal-sized colonies by adding pollen or young brood and measured the pollen-foraging activity, while foragers had either direct access to them or not. Our results show that the amount of stored pollen, young brood, and empty space directly provide important stimuli that affect foraging behavior. Different mechanisms for forager perception of the change in the environment are discussed.
