    Description: Imaging spectroscopy, also known as hyperspectral remote sensing, is based on the characterization of Earth surface materials and processes through spectrally-resolved measurements of the light interacting with matter. The potential of imaging spectroscopy for Earth remote sensing has been demonstrated since the 1980s. However, most of the developments and applications in imaging spectroscopy have largely relied on airborne spectrometers, as the amount and quality of space-based imaging spectroscopy data remain relatively low to date. The upcoming Environmental Mapping and Analysis Program (EnMAP) German imaging spectroscopy mission is intended to fill this gap. An overview of the main characteristics and current status of the mission is provided in this contribution. The core payload of EnMAP consists of a dual-spectrometer instrument measuring in the optical spectral range between 420 and 2450 nm with a spectral sampling distance varying between 5 and 12 nm and a reference signal-to-noise ratio of 400:1 in the visible and near-infrared and 180:1 in the shortwave-infrared parts of the spectrum. EnMAP images will cover a 30 km-wide area in the across-track direction with a ground sampling distance of 30 m. An across-track tilted observation capability will enable a target revisit time of up to four days at the Equator and better at high latitudes. EnMAP will contribute to the development and exploitation of spaceborne imaging spectroscopy applications by making high-quality data freely available to scientific users worldwide.

    Notes: Abstract  Opaque-2 (O2) encodes a transcriptional activator of the basic domain-leucine zipper (bZIP) class, which controls the expression level in maize endosperm of the 22 kD α-zeins and a number of non-storage proteins. The interaction of the O2 protein at three clustered binding sites on an isolated 22 kD zein gene promoter has been investigated. O2 is shown to transactivate transcription from these sites in tobacco mesophyll protoplasts as well as in maize endosperm cells transformed by particle bombardment. The binding sites have been mutated by base exchanges, singly or in different combinations, to determine their contribution to transactivation in vivo in both the leaf protoplast and the maize endosperm system. The effect of these mutations on binding of O2 in vitro was determined by electrophoretic mobility shift assays (EMSA), using O2 protein expressed in E. coli. Two of the sites seemed to be equally effective in responding to Opaque-2 in vivo in both cell types, although one of them does not contain an ACGT core sequence, and has a lower affinity for O2 in vitro than the ACGT-containing binding site. A third site, which has the lowest affinity of all three, confers no detectable O2-dependent promoter activation alone, but significantly increases activation in combination with either one of the other sites. Hence, weaker O2 binding sites can still mediate major O2-dependent effects when present in target promoters in vivo.

    Notes: X-ray scattering techniques have been a very useful tool for the non-destructive analysis of the wood structure. X-rays are sensitive to structural parameters such as the composite structure of wood cell walls, the crystal structure of cellulose microfibrils and their helical arrangement in the cell wall, which is usually described by the microfibril angle (MFA). With the availability of synchrotron radiation sources novel experiments on wood have become possible. The increased flux of X-rays makes the in situ and time-resolved investigation of structural changes upon mechanical stress possible. The low-divergence synchrotron radiation X-rays can be focused down to sub-micrometer size, enabling scanning studies of the wood nanostructure with (sub-)microscopic position resolution. This chapter highlights very recent advances in the understanding of wood micro- and nanostructure, which were only possible using synchrotron radiation. Examples include the MFA determination in the individual layers of the secondary cell wall, the imaging of the helical structure of the cellulose microfibrils in the cell wall, lattice strain as induced by applied mechanical stress and the structural changes of different wood types under external tensile stress

    Notes: Abstract A 1420 bp genomic fragment (λ-horl-17) encompassing a Hor-1 gene encoding a C-hordein polypeptide is presented. The deduced amino acid sequence is 261 residues long. It comprises a 20 amino acid signal peptide, unique NH2- and COOH-terminal regions and a coding region comprised of pentapeptide (PQQPY) and octapeptide (PQQPFPQQ) repeat motifs. The 431 bp of 5′ non-coding region contains a ‘TATA box’ at −105, a ‘CACA box’ (−181 to −201) and a −300 prolamin element. In the 3′ noncoding region there are two putative polyadenylation signals located 88 and 142 bp downstream of the stop codon. The structure of λ-hor1-17 is compared with that of another gene (λ-hor1-14) encoding a C-hordein polypeptide, which contains an amber codon interrupting the ORF. A functional assay in which the 5′ non-coding regions of the two genes were fused to the β-glucuronidase (GUS) gene demonstrated that both genes were transcriptionally active and that circa 430 bp of the C-hordein promoters were sufficient to drive the expression of the GUS gene in developing barley endosperms. It also demonstrated that both promoters had transcriptional efficiencies comparable with that of the 35S CaMV promoter. The in vitro translation of the coding region of λ-hor1-14 in the wheat germ system showed that the premature stop codon could be partially suppressed. The suppression was also demonstrated in a transient expression assay in vivo using isolated barley endosperms.

    Notes: Abstract Mitochondrial inclusion bodies are often described in skeletal muscle of patients suffering diseases termed mitochondrial myopathies. A major component of these structures was discovered as being creatine kinase. Similar creatine kinase enriched inclusion bodies in the mitochondria of creatine depleted adult rat cardiomyocytes have been demonstrated. Structurally similar inclusion bodies are observed in mitochondria of ischemic and creatine depleted rat skeletal muscle. This paper describes the various methods for inducing mitochondrial inclusion bodies in rodent skeletal muscle, and compares their effects on muscle metabolism to the metabolic defects of mitochondrial myopathy muscle. We fed rats with a creatine analogue guanidino propionic acid and checked their soled for mitochondrial inclusion bodies, with the electron microscope. The activity of creatine kinase was analysed by measuring creatine stimulated oxidative phosphorylation in soleus skinned fibres using an oxygen electrode . The guanidino propionic acid-rat soleus mitochondria displayed no creatine stimulation, whereas control soleus did, even though the GPA soled had a five fold increase in creatine kinase protein per mitochondrial protein. The significance of these results in light of their relevance to human mitochondrial myopathies and the importance of altered muscle metabolism in the formation of these crystalline structures are discussed.     Notes: Abstract Feature trees are the formal basis for algorithms manipulating record like structures in constraint programming, computational linguistics and in concrete applications like software configuration management. Feature trees model records, and constraints over feature trees yield extensible and modular record descriptions. We introduce the constraint system FT≤ of ordering constraints interpreted over feature trees. Under the view that feature trees represent symbolic information, the relation ≤ corresponds to the information ordering (“carries less information than”). We present two algorithms in cubic time, one for the satisfiability problem and one for the entailment problem of FT≤. We show that FT≤ has the independence property. We are thus able to handle negative conjuncts via entailment and obtain a cubic algorithm that decides the satisfiability of conjunctions of positive and negated ordering constraints over feature trees. Furthermore, we reduce the satisfiability problem of Dörre's weak subsumption constraints to the satisfiability problem of FT≤ and improve the complexity bound for solving weak subsumption constraints from O(n5) to O(n3) .

    Notes: Abstract Sourdough is the foremost cereal fermentation performed in a variety of technologies with almost any cereal. The lactobacilli studied most intensely include Lactobacillus sanfranciscensis, L. reuteri and L. pontis isolated from traditional and modern rye and wheat fermentations. Molecular biology tools are available for their rapid identification and monitoring throughout a process. The currently available insight on their metabolism can be used to explain their prevalence in this environment and their interactions. Key genes of the sugar degradation pathway were cloned and characterised from L. sanfranciscensis. In addition some strains were found to have special properties including the production of antagonistic compounds or the adhesion to human intestinal cells.
