Notes: Abstract Purpose. To determine the human jejunal permeabilities of compounds utilizing different transport mechanisms using a regional perfusion approach and to establish a standard procedure for determining drug permeability class to be used for the establishment of drug product bioequivalence standards. Methods. Six healthy male volunteers participated in this study. A multi-lumen perfusion tube was inserted orally and positioned in the proximal region of the jejunum. A solution containing piroxicam, phenylalanine, propranolol, PEG 400 and PEG 4000 was perfused through the intestinal segment at a rate of 3.0 ml/min. Perfusate samples were quantitatively collected every 10 minutes for two 100 minute periods with an intermediate wash out period to determine intra and intersubject variation. Results. The mean Peff (±SD) of piroxicam, phenylalanine, propranolol, and PEG 400 were 10.40 ± 5.93, 6.67 ± 3.42, 3.59 ± 1.60, 0.80 ± 0.46 × 10−4 cm/sec, respectively. The coefficient of variation for the intersubject variability, first and second perfusion periods were: piroxicam, 60.5% and 57.1%; phenylalanine, 52.8% and 57.8%: propranolol, 62.1 % and 44.6%; and PEG 400, 81.7% and 42.3%, indicating a slightly lower CV for the second perfusion period in the same subject. The intrasubject CV's between the two perfusion periods were: 19.4%, 21.3%, 23.6% and 41.0% respectively, indicating a smaller intraindividual variation for all compounds studied. Conclusions. Piroxicam, a nonpolar drug exhibited the highest permeability of the compounds studied. The intrasubject CV was lower than the intersubject CV, indicating consistent permeability estimation within subjects. The methodology is useful for permeability estimation regardless of absorption mechanism and can be used to establish a consistent data base of human permeabilities for estimation of human drug absorption and for establishing the biopharmaceutic permeability class of drugs.

    Topics: Medicine , Technology
Notes: Abstract Tunable diode laser spectrometry was used to measure breath carbon monoxide (CO) concentrations in nonsmoking subjects. Corresponding blood carboxyhemoglobin (COHb) saturations between physiological levels and levels after 1 and 8 hours exposure to 9 ppm CO, the maximum permissible concentration in the air under federal regulations, were also determined. A good linear correlation between breath CO and blood COHb saturation for low levels representing ambient conditions was established, with a correlation coefficient better than 0.999 for the means and 0.945 for the individual data points. Similar studies on the other compounds of concern are also possible using tunable diode laser spectrometry, requiring only a breath sample and with no sample preparation. This technique will facilitate fast, noninvasive environmental health effect and biomedical studies.

Notes: Abstract HealthyVinca rosea plants, connected by dodder strands with spike-diseased sandal trees developed witches' broom symptoms. Electron microscopy of petioles of the infectedVinca plants revealed the presence of numerous mycoplasma-like bodies. Fresh dodder established on theVinca plants with witches' broom symptoms and trained onto healthy sandal trees, transmitted the spike disease back to the latter.

Notes: A coupled cellular automaton-finite difference (CA-FD) model is used to simulate thedetailed dendritic structure evolution of the columnar-to-equiaxed transition (CET) for Al-Cu alloysduring solidification. The effects of material properties (nucleation undercooling, density of nuclei inbulk liquid and alloy solidification range) on the CET are investigated. Simulated results reveal that:(1) equiaxed grains form at an earlier stage with a smaller critical nucleation undercooling; (2) CET ispromoted if the density of nuclei in bulk liquid is increased; (3) extending the alloy solidificationrange promotes the CET. Finally, CET maps corresponding to different alloy concentrations areconstructed, illustrating the relationship between processing conditions and the resulting grainstructures for alloys with different solidification ranges

Notes: Models describing the critical steps in the manufacturing process of aluminum alloywheels were integrated with a model of in-service loading to predict component fatigueperformance. A multiscale solidification model was coupled with models of the subsequent heattreatment and machining to predict the residual stress distribution in a finished wheel. A secondmultiscale model was incorporated into a service model to relate the pore size and local stress stateto fatigue performance. The predictions resulting from this through process modeling approachhave been validated by comparisons with a series of measurements: 1) thermocouple measurementsduring casting; 2) characterization of pore size and distribution using X-ray microtomography andoptical metallography; 3) residual strain measurements on finished wheels; 4) in-service strainmeasurements during a rotating bend test; and 5) in-service fatigue performance during rotatingbend tests

Notes: Abstract Threads can easily be implemented using first-class continuations, but the straightforward approaches for doing so lead to space leaks, especially in a language with exceptions like Standard ML. We show how these space leaks arise and give a new implementation for threads that is safe for space.

Notes: Abstract This paper examines the transformation of call-by-need λ terms into continuation-passing style (CPS). It begins by presenting a simple transformation of call-by-need λ terms into program graphs and a reducer for such graphs. From this, an informal derivation is carried out, resulting in a translation from λ terms into self-reducing program graphs, where the graphs are represented as CPS terms involving storage operations. Though informal, the derivation proceeds in simple steps, and the resulting translation is taken to be our canonical CPS transformation for call-by-need λ terms. In order to define the CPS transformation more formally, two alternative presentations are given. The first takes the form of a continuation semantics for the call-by-need language. The second presentation follows Danvy and Hatcliff's two-stage decomposition of the call-by-name CPS transformation, resulting in a similar two-stage CPS transformation for call-by-need. Finally, a number of practical matters are considered, including an improvement to eliminate the so-called administrative redexes, as well as to avoid unnecessary memoization and take advantage of strictness information. These improvements make it feasible to consider potential applications in compilers for call-by-need programming languages.
