    Notes: A copolymer of vinylidene fluoride and tetrafluoroethylene is solution cast on a p-type silicon wafer. A strong inversion is produced at the silicon surface when the copolymer film is corona charged or when a subsequently evaporated aluminum gate is biased. Although the dipole alignment in the copolymer is stable, the inversion decays. The decay of the inversion layer is monitored by measuring the decay of the surface channel currents between a source and a drain. For the case of corona charging, an initial rapid decay not observed in the gated device is explained by the motion of excess positive charges (dielectric relaxation). The slow decay subsequently observed is explained in terms of the thermionic injection of inversion electrons across an interface barrier and also using a space-charge-limited injection model. Both models fit the results and their validity is discussed.

    Notes: Abstract: In Mollusca, the mantle produces an organic matrix that mineralizes in time to make shell. Primary mantle cell cultures from the nacreous gastropod Haliotis tuberculata have been established as useful experimental model to investigate in vitro synthesis of both proteoglycans/glycosaminoglycans (PGs/GAGs) and collagen. First, we tested different enzymatic digestion procedures to find the method that gives the highest percentage of viable and adherent cultured cells. Enzymatic digestion with 0.1% pronase plus 0.1% collagenase was routinely used. Six days after the initiation of culture, about 80% of cells were viable, among which 20% were adherent as quantified by the MTT reduction assay. In addition, the protein synthesis estimated by [3H]leucine incorporation remained constant during this period. For the first time, we demonstrated a de novo synthesis of PGs/GAGs and collagen in primary cultures of mantle cells. After 48 hours of labeling, among the [3H]-d-glucosamine macromolecules synthesized, [3H]PGs/GAGs represented 43%, divided into 45% heparan sulfate, 37% chondroitin/dermatan sulfate, and 6% hyaluronic acid. Early elution on anion-exchange chromatography of these PGs/GAGs indicated that most of them appeared as undersulfated GAG molecules. De novo synthesis of collagen represents 4.52% ± 0.84% (SD) with respect to the total protein synthesis. Such a model will facilitate studies on the synthesis of PGs/GAGs and collagen as components of the extracellular matrix and its regulation in Mollusca. Both PGs/GAGs and collagen participate in molecular events that regulate cell adhesion, migration, and proliferation. Further studies with this type of in vitro model should provide knowledge about novel aspects of molluscan cell signaling, in relation to extracellular matrix components.

    Notes: Abstract The incorporation of mercury into the food chain and its assimilation by humans is a universally recognized potential health hazard. Studies carried out in the Amazon Basin have shown that mercury (Hg) is present in fish and in humans, however, the relation between fish diet and human exposure has received limited attention in this region. The present study focused on a small village, Brasília Legal (3°59′00″S, 55°30′00″W), situated on the banks of the Rio Tapajós. A total of 181 fish (40 species) were captured in March, 1995 and analysed for Hg concentration. Of these, 132 fish were among species consumed by the population during the rainy season (mid-November to mid-May) and the dry season (mid-May to mid-November). Wide intra- and inter-species variations in Hg concentrations were observed. Thirty four fish (25.8% of the consumed species) had levels above 0.5 μg/g Hg fresh weight; all were among the piscivorous and omnivorous species. Hair Hg concentrations (HHg), showed that villagers with a high fish diet (n=31; median HHg=16.1 μg/g) and mixed fish diet (n=36; median HHg=14.8 μg/g) had significantly higher HHg concentrations compared to the low fish diet group (n=29; mean HHg=7.8 μg/g). Time series function of HHg measurements, made for 26 persons with over 24 cm of hair, revealed sinusoidal variations, with peaks during the rainy season and troughs during the low water period, paralleling the seasonal shift in dietary habits. Piscivorous and omnivorous fish species, with higher mercury levels, are the main component of the fish diet during the rainy season, while herbivorous fish species predominate during the dry season. Preventive actions should take into account the risk to human health, particularly for fetal and neonatal development, the importance of fish in the riparian diet, the wide intra- and inter-species variations in mercury content and seasonal fluctuations in diet.

    Notes: Abstract In relation with the digestive cycle, the digestive gland cells of bivalve molluscs undergo a sequence of cytological changes which is controlled by external and internal effectors such as putative gastrointestinal hormones and growth differentiation factors. A tissue dissociation method was developed to investigate the in vitro effect of the vertebrate growth and differentiation factors: insulin, insulin growth factor I (IGF-I), basic fibroblast growth factor (bFGF) and epidermal growth factor (EGF) on the digestive gland cells of the scallop Pecten maximus. All these vertebrate peptides induced a dose-dependent increased incorporation of 3H-leucine and 14C-uridine in whole digestive gland cell suspensions. However, after Percoll density gradient purification of the digestive cells, only stem and undifferentiated enriched cell fractions were responsive to the different peptides. In addition, insulin and IGF-I, but not EGF and bFGF, stimulated 3H-leucine incorporation in control dispersed mantle edge cells. These results suggest that insulin-related peptides could work as general growth promoting factors in molluscs. On the other hand, EGF and bFGF, or at least their molluscan counterparts, may be efficient growth differentiation factors in the regenerative processes occurring in the digestive gland of molluscs.

    Description: In a world of increasing exposure of populations to natural hazards, the mapping of extreme rainfall remains a key subject of study. Such maps are required for both flood risk management and civil engineering structure design, the challenge being to take into account the local information provided by point rainfall series as well as the necessity of some regional coherency. Two approaches based on the extreme value theory are compared here, with an application to extreme rainfall mapping in West Africa. The first approach is a local fit and interpolation (LFI) consisting of a spatial interpolation of the generalized extreme value (GEV) distribution parameters estimated independently at each station. The second approach is a spatial maximum likelihood estimation (SMLE); it directly estimates the GEV distribution over the entire region by a single maximum likelihood fit using jointly all measurements combined with spatial covariates. Five LFI and three SMLE methods are considered, using the information provided by 126 daily rainfall series covering the period 1950–1990. The methods are first evaluated in calibration. Then the predictive skills and the robustness are assessed through a cross validation and an independent network validation process. The SMLE approach, especially when using the mean annual rainfall as covariate, appears to perform better for most of the scores computed. Using the Niamey 104 year time series, it is also shown that the SMLE approach has the capacity to deal more efficiently with the effect of local outliers by using the spatial information provided by nearby stations.
