    Description: The Hegemony of Heritage makes an original and significant contribution to our understanding of how the relationship of architectural objects and societies to the built environment changes over time. Studying two surviving medieval monuments in southern Rajasthan—the Ambikā Temple in Jagat and the Śri Ékliṅgjī Temple Complex in Kailāshpurī—the author looks beyond their divergent sectarian affiliations and patronage structures to underscore many aspects of common practice. This book offers new and extremely valuable insights into these important monuments, illuminating the entangled politics of antiquity and revealing whether a monument’s ritual record is affirmed as continuous and hence hoary or dismissed as discontinuous or reinvented through various strategies. The Hegemony of Heritage enriches theoretical constructs with ethnographic description and asks us to reexamine notions such as archive and text through the filter of sculpture and mantra.

    Description: Traditional medicinal literature and previous studies have reported the possible role of Cissus quadrangularis (CQ) as an anti-osteoporotic agent. This study examines the effectiveness of CQ in promoting osteoblast differentiation of the murine pre-osteoblast cell line, MC3T3-E1. Ethanolic extract of CQ (CQ-E) was found to affect growth kinetics of MC3T3-E1 cells in a dosage dependent manner. High concentrations of CQ-E (more than 10 µg/ml) have particularly adverse effects, while lower concentrations of 0.1 and 1 µg/ml were non-toxic and did not affect cell viability. Notably, cell proliferation was significantly increased at the lower concentrations of CQ-E. CQ-E treatment also augmented osteoblast differentiation, as reflected by a substantial increase in expression of the early osteoblast marker ALP activity, and at later stage, by mineralization of extracellular matrix compared to the control group. These findings suggest dose-dependent effect of CQ-E with lower concentrations exhibiting anabolic and osteogenic properties. This article is protected by copyright. All rights reserved

    Description: The cell cycle in pluripotent human embryonic stem cells is governed by unique mechanisms that support unrestricted proliferation and competency for endodermal, mesodermal and ectodermal differentiation. The abbreviated G1 period with retention of uncompromised fidelity for genetic and epigenetic mechanisms operative in control of proliferation support competency for expansion of the pluripotent cell population that is fundamental for initial stages of development. Regulatory events during the G1 period of the pluripotent cell cycle are decisive for the transition from pluripotency to lineage commitment. Recent findings indicate that a G2 cell cycle pause is present in both endodermal and mesodermal lineage cells, and is obligatory for differentiation to endoderm.     Description: Three-dimensional organization of the chromatin has important roles in transcription, replication, DNA repair, and pathologic events such as translocations. There are two fundamental ways to study higher-order chromatin organization: microscopic and molecular approaches. In this review, we briefly introduce the molecular approaches, focusing on chromosome conformation capture or“3C” technology and its derivatives, which can be used to probe chromatin folding at resolutions beyond that provided by microscopy techniques.     Description: Regional climate models (RCMs) are a standard tool for downscaling climate forecasts to finer spatial scales. The evaluation of RCMs against observational data is an important step in building confidence in the use of RCMs for future prediction. In addition to model performance in climatological means and marginal distributions, a model's ability to capture spatio-temporal relationships is important. This study develops two approaches: (1) spatial correlation/variogram for a range of spatial lags, with total monthly precipitation and non-seasonal precipitation components used to assess the spatial variations of precipitation; and (2) spatio-temporal correlation for a wide range of distances, directions, and time lags, with daily precipitation occurrence used to detect the dynamic features of precipitation. These measures of spatial and spatio-temporal dependence are applied to a high-resolution RCM run and to the National Center for Environmental Prediction (NCEP)-U.S. Department of Energy (DOE) AMIP II reanalysis data (NCEP-R2), which provides initial and lateral boundary conditions for the RCM. The RCM performs significantly better than NCEP-R2 in capturing both the spatial variations of total and non-seasonal precipitation components and the spatio-temporal correlations of daily precipitation occurrences, which are related to dynamic behaviors of precipitating systems. The improvements are apparent not just at resolutions finer than that of NCEP-R2, but also when the RCM and observational data are aggregated to the resolution of NCEP-R2.

    Description: Mammalian fertilization is accompanied by oscillations in egg cytoplasmic calcium (Ca2+) concentrations that are critical for completion of egg activation. These oscillations are initiated by Ca2+ release from inositol 1,4,5-trisphosphate (IP3)-sensitive intracellular stores. We tested the hypothesis that Ca2+ influx across the plasma membrane was a requisite component of egg activation signaling, and not simply a Ca2+ source for store repletion. Using intracytoplasmic sperm injection (ICSI) and standard in vitro fertilization (IVF), we found that Ca2+ influx was not required to initiate resumption of meiosis II. However, even if multiple oscillations in intracellular Ca2+ occurred, in the absence of Ca2+ influx, the fertilized eggs failed to emit the second polar body, resulting in formation of three pronuclei. Additional experiments using the Ca2+ chelator, BAPTA/AM, demonstrated that Ca2+ influx is sufficient to support polar body emission and pronucleus formation after only a single sperm-induced Ca2+ transient, whereas BAPTA/AM-treated ICSI or fertilized eggs cultured in Ca2+-free medium remained arrested in metaphase II. Inhibition of store-operated Ca2+ entry had no effect on ICSI-induced egg activation, so Ca2+ influx through alternative channels must participate in egg activation signaling. Ca2+ influx appears to be upstream of CaMKIIγ activity because eggs can be parthenogenetically activated with a constitutively active form of CaMKIIγ in the absence of extracellular Ca2+. These results suggest that Ca2+ influx at fertilization not only maintains Ca2+ oscillations by replenishing Ca2+ stores, but also activates critical signaling pathways upstream of CaMKIIγ that are required for second polar body emission.
