    Description: We report here the identification of the key sites for the beta structure motifs of the islet amyloid polypeptide (IAPP) analogs by using scanning tunneling microscopy (STM). Duplex folding structures in human IAPP8–37 (hIAPP8–37) assembly were observed featuring a hairpin structure. The multiplicity in rIAPP assembly structures indicates the polydispersity of the rat IAPP8–37 (rIAPP8–37) beta-like motifs. The bimodal length distribution of beta structure motifs for rIAPP8–37 R18H indicates the multiple beta segments linked by turns. The IAPP8–37 analogs share common structure motifs of IAPP8–17 and IAPP26–37 with the most probable key sites at positions around Ser19/Ser20 and Gly24. These observations reveal the similar amyloid formation tendency in the C and N terminus segments because of the sequence similarity, while the differences in specific amino acids at each key site manifest the effect of sequence variations. The results could be beneficial for studying structural polymorphism of amyloidal peptides with multiple beta structure motifs.

    Description: The results of application of the quantum-mechanical adiabatic theory to vibrational predissociation (VPD) of water dimers, (H 2 O) 2 and (D 2 O) 2 , are presented. We consider the VPD processes including the totally symmetric OH mode of the dimer and the bending mode of the fragment. The VPD in the adiabatic representation is induced by breakdown of the vibrational adiabatic approximation, and two types of nonadiabatic coupling matrix elements are involved: one provides the VPD induced by the low-frequency dissociation mode and the other provides the VPD through channel interactions induced by the low-frequency modes. The VPD rate constants were calculated using the Fermi golden rule expression. A closed form for the nonadiabatic transition matrix element between the discrete and continuum states was derived in the Morse potential model. All of the parameters used were obtained from the potential surfaces of the water dimers, which were calculated by the density functional theory procedures. The VPD rate constants for the two processes were calculated in the non-Condon scheme beyond the so-called Condon approximation. The channel interactions in and between the initial and final states were taken into account, and those are found to increase the VPD rates by 3(1) orders of magnitude for the VPD processes in (H 2 O) 2 ((D 2 O) 2 ). The fraction of the bending-excited donor fragments is larger than that of the bending-excited acceptor fragments. The results obtained by quantum-mechanical approach are compared with both experimental and quasi-classical trajectory calculation results.

    Description: Environment reconstruction is to rebuild the physical environment in the cyberspace using the sensory data collected by sensor networks, which is a fundamental method for human to understand the physical world in depth. A lot of basic scientific work such as nature discovery and organic evolution heavily relies on the environment reconstruction. However, gathering large amount of environmental data costs huge energy and storage space. The shortage of energy and storage resources has become a major problem in sensor networks for environment reconstruction applications. Motivated by exploiting the inherent feature of environmental data, in this paper, we design a novel data gathering protocol based on compressive sensing theory and time series analysis to further improve the resource efficiency. This protocol adapts the duty cycle and sensing probability of every sensor node according to the dynamic environment, which cannot only guarantee the reconstruction accuracy, but also save energy and storage resources. We implement the proposed protocol on a 51-node testbed and conduct the simulations based on three real datasets from Intel Indoor, GreenOrbs and Ocean Sense projects. Both the experiment and simulation performances demonstrate that our method significantly outperforms the conventional methods in terms of resource efficiency and reconstruction accuracy.

    Description: DNA Topoisomerases are essential to resolve topological problems during DNA metabolism in all species. However, the prevalence and function of RNA topoisomerases remain uncertain. Here, we show that RNA topoisomerase activity is prevalent in Type IA topoisomerases from bacteria, archaea, and eukarya. Moreover, this activity always requires the conserved Type IA core domains and the same catalytic residue used in DNA topoisomerase reaction; however, it does not absolutely require the non-conserved carboxyl-terminal domain (CTD), which is necessary for relaxation reactions of supercoiled DNA. The RNA topoisomerase activity of human Top3β differs from that of Escherichia coli topoisomerase I in that the former but not the latter requires the CTD, indicating that topoisomerases have developed distinct mechanisms during evolution to catalyze RNA topoisomerase reactions. Notably, Top3β proteins from several animals associate with polyribosomes, which are units of mRNA translation, whereas the Top3 homologs from E. coli and yeast lack the association. The Top3β-polyribosome association requires TDRD3, which directly interacts with Top3β and is present in animals but not bacteria or yeast. We propose that RNA topoisomerases arose in the early RNA world, and that they are retained through all domains of DNA-based life, where they mediate mRNA translation as part of polyribosomes in animals.

    Description: Aims Studying the ecological significance of highly specialized morphological traits evolved by alpine plants could help us to understand the adaptation and evolution of these plants under severe alpine environment. We explored the adaptive significance of woolly and overlapping leaves for reproduction in Eriophyton wallichii , a perennial herb native to the subnival belt of the Himalaya-Hengduan Mountains. Methods We examined whether the trichomes could influence the leaf wettability, temperature and leaf reflectance spectra in the lab. And we investigated the thermal benefits of the woolly and overlapping leaves for flowers and fruits in the field. Pollen viability and seed germination were also examined in the lab to assess whether these leaves enhance reproductive fitness. Important Findings Our results showed that dense trichomes impart good water repellency, absorption of solar radiation and accumulation of leaf heat. The woolly and overlapping leaves increased the interior temperature of flowers and fruits to an optimal level on sunny hours, but prevented them from overheating when transient intense solar radiation occurs. This kept optimal temperatures in plants’ reproductive organs, thus promoting the development of pollen and seed in alpine environment.

    Description: In geochemical elemental logging, the elemental sensitivity factor is a key parameter for converting relative yields of elements obtained from spectral analysis to actual concentrations of the elements in the formation. Here we perform a quantitative analysis of the Monte Carlo N -particle (MCNP) simulated spectra of elements in a model formation and compare the obtained relative yield of each element with its actual dry weight in a model formation to study their relationship. For a series of model formations with simple and complex lithologies, the mixed formation spectrum was obtained through MCNP simulation, and elemental sensitivity factors in these formations were calculated according to elemental sensitivity factor theory and applied in calculating dry weights of actual formation elements. It is thus inferred that the elemental sensitivity factor is a constant quantity and does not vary with the nature of the formation. The elemental sensitivity factors calculated for the simulated formations are close to actually measured sensitivity factors and can be used to analyse actual formation elements.

    Description: The transfer of alien genes to crop plants using chromosome engineering has been attempted infrequently in tetraploid durum wheat ( Triticum turgidum L. subsp. durum ). Here, we report a highly efficient approach for the transfer of two genes conferring resistance to stem rust race Pgt -TTKSK (Ug99) from goatgrass ( Aegilops speltoides ) to tetraploid wheat. The durum line DAS15, carrying the stem rust resistance gene Sr47 derived from Ae. speltoides , was crossed, and backcrossed, to durum 5D(5B) aneuploids to induce homeologous pairing. After a final cross to ‘Rusty’ durum, allosyndetic recombinants were recovered. The Ae. speltoides chromosomal segment carrying Sr47 was found to have two stem rust resistance genes. One gene conditioning an infection type (IT) 2 was located in the same chromosomal region of 2BS as Sr39 and was assigned the temporary gene symbol SrAes7t . Based on ITs observed on a diverse set of rust races, SrAes7t may be the same as Sr39 . The second gene conditioned an IT 0; and was located on chromosome arm 2BL. This gene retained the symbol Sr47 because it had a different IT and map location from other stem rust resistance genes derived from Ae. speltoides . Allosyndetic recombinant lines carrying each gene on minimal alien chromosomal segments were identified as were molecular markers distinguishing each alien segment. This study demonstrated that chromosome engineering of Ae. speltoides segments is feasible in tetraploid wheat. The Sr47 gene confers high-level and broad spectrum resistance to stem rust and should be very useful in efforts to control TTKSK.

    Description: The hypothesis that the joint effects of predation and cyanobacteria favor the performance of small-bodied cladocerans relative to large-bodied species was tested using three competition experiments in a two-factor full-factorial design. The large-bodied Daphnia pulex outcompeted the medium-bodied Simocephalus vetulus when fed a green alga Chlorella pyrenoidosa . However, the presence of the toxic cyanobacterium Microcystis aeruginosa and the coelenterate predator Hydra oligactis reversed the competitive outcome. Their joint effect was significant and contributed to the strongest dominance of S. vetulus . Similar dominance shifts were also observed in the competition between D. pulex and the small-bodied Bosmina longirostris , and in the competition between S. vetulus and B. longirostris . Life table and predation experiments showed that the order of sensitivity of three cladoceran species to Microcystis and to Hydra was the same as the order of competitive superiority in the absence of cyanobacteria. Therefore, cyanobacteria, predation and their joint effects are important drivers for competitive outcomes among cladocerans, potentially creating new opportunities for small-bodied species to dominate in a community. The present study underscores the importance of considering the combined effects of dietary palatability and predation on competitive dominance among herbivores, when predicting zooplankton responses to frequent cyanobacterial blooms.
